We adapted the genetic algorithm to minimize the AMBER potential energy function. We describe specific recombination and mutation operators for this task. Next we use our algorithm to locate low energy conformation of three polypeptides (AGAGAGAGA, A9, and [Met]-enkephalin) which are probably the global minimum conformations. Our potential energy minima are –94.71, –98.50, and –48.94 kcal/mol respectively. Next, we applied our algorithm to the 46 amino acid protein crambin and located a non-native conformation which had an AMBER potential energy 150 kcal/mol lower than the native conformation. This is not necessarily the global minimum conformation, but it does illustrate problems with the AMBER potential energy function. Although frit-fast atom bombardment (frit-FAB) and continuous-flow FAB mass spectrometry have become standard methods for the analysis of peptides and peptide mixtures, these techniques have not been applied previously to the analysis of oligonucleotides. Mobilephase composition, flow rate, and sample size were optimized for the analysis of oligonucleotides by negative ion frit-FAB mass spectrometry (a type of continuous-flow FAB mass spectrometry). With a mobile phase consisting of methanol/water/triethanolamine (80:20:0.5, v/v/w), flow injection frit-FAB analysis of oligonucleotides showed lower limits of detection compared to standard probe FAB mass spectrometry. For example, in order to obtain a signal-to-noise ratio of 3:1, 38 prnol of d(GTIAAC) were required for frit-FAB mass spectrometry and 62 pmol were required for standard probe FAB mass spectrometry. The largest difference between frit-FAB and standard probe FAB was observed for d(pC)5, for which the limit of detection by frit-FAB was approximately 11-fold lower than by standard FAB mass spectrometry. Adjustment of the mobile phase to pH 7 with trifluoroacetic acid increased the limit of detection (reduced sensitivity) a minimum of sixfold. Equimolar mixtures of two or three oligonucleotides produced deprotonated molecules in identical relative abundances whether analyzed by frit-FAB or standard probe FAB mass spectrometry. Finally, frit-FAB liquid chromatography mass spectrometry was demonstrated by separating mixtures of oligonucleotides on a β -cyclodextrin high-performance liquid chromatography column with a mobile phase containing methanol, water, and triethanolamine. 相似文献

A new method was developed for the determination of 2-furfural (2-F) and 5-methylfurfural (5-MF), two products of Maillard reaction in vinegar, with head-space solid-phase microextraction (HS-SPME) coupled to gas chromatography-mass spectrometry (GC-MS). A divinylbenzene (DVB)/carboxen (CAR)/polydimethylsiloxane (PDMS) fibre was used and SPME conditions were optimised, studying ionic strength effect, temperature effect and adsorption time. Both analytes were determined by calibration established on 2-furfural-d4 (2-F-d4). The method showed good linearity in the range studied (from 16 to 0.12 mg/l), with a regression coefficient r2 of 0.9999. Inter-batch precision and accuracy were found between 14.9 and 6.0% and between -11.7 and 0.2%, respectively. Detection limit was 15 microg/l. The method is simple and accurate and it has been applied to a series of balsamic and non-balsamic vinegars. 相似文献

13C NMR measurements were performed on [Re3(μ-H)3(CO)10]2? at various temperatures and field strengths. Selective decoupling allowed assignments of the carbonyl resonances. Spin-lattice relaxation time measurements indicated that two mechanisms, scalar coupling and chemical shielding anisotropy, contribute to the relaxation of carbon-13. The composition of the essential oil of Bifora radians, an aldehyde-producing weed, has been investigated by capillary gas chromatography, coupled gas chromatography – mass spectrometry, on-line catalytic hydrogenation and coupled gas chromatography – infrared spectrometry. The nineteen compounds identified included eighteen aldehydes: seven alkanals (C6, C9, C10, C11, C12, C13, and C14), ten alkenals, including five (E)-2-alkenals (C12, C13, C14, C15, and C16), and one (E,E)-2,4-alkadienal (C13). The gas-phase acidity of ionized benzyl alcohol and of some of its derivatives with selected reference bases has been studied by Fourier Transform Ion Cyclotron Resonance (FT-ICR) mass spectrometry. The aim was to relate the gas-phase reactivity to the behavior in aqueous solution of the radical cations of benzyl alcohols bearing methoxy substituent(s) on the phenyl ring which are known to undergo deprotonation at both the CH2 and OH groups. The dual reactivity behavior is confirmed in the gas phase, in which the prototypical ion, C6H5CH2OH*+, is deprotonated at both the CH2 and OH groups, whereas the ring hydrogens are not involved. An increasing extent of O-deprotonation is shown as the strength of the base increases. Appropriate methyl substitution, as in the radical cations of C6H5C(Me)2OH and C6H5CH2OMe, allows only O- or C-acidity. The two processes are characterized by comparable thermodynamic features with a Gas-phase Basicity (GB) value of 852 kJ mol(-1) for the cumyloxyl radical and 850 kJ mol(-1) for the alpha-methoxybenzyl radical. The possible origin of the observed mechanistic dichotomy is discussed. 相似文献

In previous work (Le Bizec et al., Rapid Commun. Mass Spectrom. 2000; 14: 1058), it was demonstrated that a boar meal intake could lead to possible false accusations of abuse of 17beta-nortestosterone in antidoping control. The aim of the present study was to identify and quantify endogenous 19-norsteroids in boar edible tissue at concentrations that can alter the steroid urinary profile in humans, and lead to excretion of 19-norandrosterone (19-NA) and 19-noretiocholanolone (19-NE). The samples were analysed in two laboratories. The methodologies used for extraction and detection (GC/MS(EI) and LC/MS/MS(APCI+)) are compared and discussed. 19-Norandrostenedione (NAED), 17beta- and 17alpha-nortestosterone (bNT, aNT), and 17beta- and 17alpha-testosterone (bT, aT) were quantified. The largest concentrations of NAED and bNT were observed in testicles (83 and 172 microg/kg), liver (17 and 63 microg/kg) and kidney (45 and 38 microg/kg). A correlation between the bNT and NAED content of a typical meal prepared with boar parts and the excreted concentrations of 19-NA and 19-NE in human urine was demonstrated. 相似文献

The split-root technique was used to study the effect of varying the growth media on the elemental content of nutrient elements in the roots of grape vines. The varieties 2-1 (R99 x Jacquez) and Sauvignon blanc (Vitis vinifera) were grown in Hoagland water culture with and without added aluminium. The elemental concentrations of Mg, Al, Si, P. S, Cl, K and Ca in the dried roots were determined by PIXE. Roots grown in Al-rich media were deficient in Mg and Ca, but enriched in Al. There was a correlation between Al and Si but the uptake differed in the two varieties. 相似文献
