A possible DNA damage after interaction of kainic acid (KA) with calf thymus double stranded DNA and genomic DNA was herein determined in in vitro and in vivo conditions using; electrochemical assay and agarose gel electrophoresis. The changes in guanine signal were detected as an indicator of DNA damage in genomic DNA samples isolated from 1 or 10 mg/kg KA‐treated animals. The decreased levels of guanine signal were found as 29% and 33% by 1 and 10 mg/kg KA treatment when compared to controls, respectively. The results of gel electrophoresis confirmed DNA damage obtained in identical samples by electrochemical method.  相似文献

In this study, the first micro-total analysis system (μ-TAS) for catecholamines (dopamine, epinephrine, and norepinephrine) analysis in which preconcentration, separation, and determination steps were integrated on a microchip was developed. Electrophoresis microchips in a variety of channel lengths and designs were produced in borofloat glass for the μ-TAS studies. Chambers for the preparation of monolithic disks were formed in the microchips at the intersection of the injection and separation channels. Vinyl phenylboronic acid–ethylene glycol dimethacrylate polymers were prepared as monolithic disks in these chambers with a depth of 0.05 mm and a diameter of 2.1 mm. The microchips could be used more than 50 times if mechanical problems such as plugging or fracturing did not occur. Adsorption and elution of catecholamines were realized electrokinetically, with catecholamines determined via laser-induced native fluorescence detection following elution and electrophoretic separation. The most promising results were obtained with 100 mM phosphate buffer (pH 2) for elution with 25% propanol added to the separation buffer (100 mM phosphate, pH 3).  相似文献

The development of amino-terminated G4 PAMAM dendrimer (PDR) modified disposable electrodes were developed as the first time in our study by using the dendrimer modified disposable graphite (PDR-PGE) and multiwalled carbon nanotube based screen-printed graphite (PDR-MWCNT-SPE) electrodes. Firstly, the microscopic characterization of bare PGEs and PDR modified PGEs was performed. These sensors were then applied for electrochemical monitoring of an anticancer drug, Daunorubicin (DNR). The enhanced oxidation signal of DNR was measured at +0.50 V by using differential pulse voltammetry (DPV) in combination with the PDR-PGEs. The detection limit, estimated from S/N = 3, corresponds accordingly to 317 nM and 128 nM for DNR respectively at the PGE and PDR-PGE. The voltammetric results were consistent with electrochemical impedance spectroscopy (EIS) that was used to characterize the successful modification of PDR onto the surface of PGE and MWCNT-SPE.  相似文献

A novel solid phase extraction microchip with a boronic acid functional affinity monolithic disc was developed in this work. Vinyl phenylboronic acid–ethylene glycol dimethacrylate co-polymer monoliths, which have pore sizes up to 20 μm, were investigated for extraction of catecholamines using adsorption and desorption studies in a batch system. Desorption yields of greater than 90% were achieved for catecholamines at pH 3 and below. Monolithic discs were then formed in chambers in borofloat glass microfluidic chips using in situ UV polymerization. Adsorption on the monolithic discs was performed via electrokinetic flow, with catecholamines determined via laser-induced native fluorescence (LINF) detection following electrokinetic elution. Microchips containing the boronic acid functional polymer discs worked well for extraction of catecholamines, providing greater than 100 fold concentration enrichment. This study demonstrated that a solid phase extraction microchip, containing an easily prepared monolith disc, will be useful for boronate affinity extraction of cis-diol containing compounds.  相似文献

In this study, convective heat transfer and pressure drop in a cross-flow heat exchanger with hexagonal, square and circular (HSC) pin–fin arrays were studied experimentally. The pin–fins were arranged in an in-line manner. For the applied conditions, the optimal spacing of the pin–fin in the span-wise and stream-wise directions has been determined. The variable parameters are the relative longitudinal pitch (S L /D = 2, 2.8, 3.5), and the relative transverse pitch was kept constant at S T /D = 2. The performances of all pin–fins were compared with each other. The experimental results showed that the use of hexagonal pin–fins, compared to the square and circular pin–fins, can lead to an advantage in terms of heat transfer enhancement. The optimal inter-fin pitches are provided based on the largest Nusselt number under the same pumping power, while the optimal inter-fin pitches of hexagonal pin–fins are S T /D = 2 and S L /D = 2.8. Empirical equations are derived to correlate the mean Nusselt number and friction coefficient as a function of the Reynolds number, pin–fin frontal surface area, total surface area, and total number. Consequently, the general empirical formula is given in the present form.

The effect of the SnO(2) nanoparticles (SNPs) on the behaviour of voltammetric carbon paste electrodes were studied for possible use of this material in biosensor development. The electrochemical behaviour of SNP modified carbon paste electrodes (CPE) was first investigated by using cyclic voltammetry (CV), differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS) techniques. The performance of the SNP modified electrodes were compared to those of unmodified ones and the parameters affecting the response of the modified electrode were optimized. The SNP modified electrodes were then tested for the electrochemical sensing of DNA purine base adenine to explore their further development in biosensor applications.  相似文献

Spin polarized Density Functional Theory combined with Non-Equilibrium Green?s Function Formalism is applied to investigate the spin dependent transport in carbon based monatomic systems. Both one-dimensional linear and ring structures sandwiched between spin polarized nickel electrodes are examined. Incorporating of nickel electrodes and rings leads to interesting spin dependent properties. The influence of electrode structure is also addressed, using the Ni(100) pyramidal and plane electrodes. It is revealed that spin dependent behavior is largely determined by the atomic arrangement of the monatomic system, and that both the transport and magnetic properties can be tuned by odd–even disparity and/or appropriate ring(s). The mechanisms governing the spin dependent properties in these structures are discussed.  相似文献

This paper presents the results of the electron paramagnetic resonance (EPR) study of the anion radical formed from 3-nitroacetophenone (C8H7NO3) (3NAP) single crystal, by gamma irradiation. The EPR spectra of gamma-irradiated single crystals of 3NAP have been recorded at 10-degree intervals for different orientations of crystals in a magnetic field, at room temperature. The EPR analysis of gamma-irradiated crystals of 3NAP has shown that the radiation damage center produced by gamma irradiation is the carbon-centered 3NAP anion radical. One-electron reduction of 3NAP results in general bond loosening. The single crystals have been investigated between 120 and 450?K. The spectra have been found to be temperature-dependent. The EPR parameters of the 3NAP anion radical have been evaluated.  相似文献
