The time dependence of atropisomer interconversion has limited the pursuit of single atropisomer drug candidates, even in circumstances where one atropisomer presents favorable biological activity over another. Moderate interconversion energy barriers risk compromising drug stability. As a result, examples of atropisomerically pure drugs in current clinical use are rare. However, in recent years, there has been a shift towards the development of single, stable atropisomer drug candidates with enhanced activity. Consequently, development of methods which effectively restrict rotation in a configuration which favors activity is highly beneficial. The picket fence porphyrin α4 atropisomer configuration has been previously demonstrated to improve the cell internalization of the pre-clinical drug, redaporfin, applied in photodynamic therapy. In this work, the α4 configuration was modelled with novel porphyrin photosensitizers through strapped moieties which effectively fixed the atropisomeric configuration. The stable cis-αα configuration demonstrated enhanced cell membrane permeation, effectively predicting the behavior of the α4 configuration and indicates that strapped porphyrins can serve as stable model systems for the investigation of photoactive drugs.  相似文献

The steady forced convection flow of a power-law fluid over a horizontal plate embedded in a saturated Darcy-Brinkman porous medium is considered. The flow is driven by a constant pressure gradient. In addition to the convective inertia, also the “porous Forchheimer inertia” effects are taken into account. The pertinent boundary value problem is investigated analytically, as well as numerically by a finite difference method. It is found that far away from the leading edge, the velocity boundary layer always approaches an asymptotic state with identically vanishing transverse component. This holds for pseudoplastic (0 < n < 1), Newtonian (n = 1), and dilatant (n > 1) fluids as well. The asymptotic solution is given for several particular values of the power-law index n in an exact analytical form. The main flow characteristics of physical and engineering interest are discussed in the paper in some detail.  相似文献

A theoretical study of the effect of variable fluid properties on the classical Blasius and Sakiadis flow is presented in this paper. The investigation concerns engine oil, water and air taking into account the variation of their physical properties with temperature. The results are obtained with the numerical simulation of the governing equations and cover large temperature differences. Velocity and temperature profiles are presented, as well as values of wall shear stress and wall heat transfer, for a variety of temperatures between the plate and the ambient fluid. It is found that the variation of fluid properties and especially viscosity have a strong influence on the results. The results of oil and water are, in general, similar and are generalized to liquids whereas air results are different and are generalized to gases. Except of the new results found in the present work some inaccurate results existing in the literature have been identified.  相似文献

The Blasius and Sakiadis flows with uniform suction or with zero transverse velocity, at the asymptotic state, in a Darcy–Brinkman porous medium are investigated in this note. Exact analytical solutions are derived for velocity as well as for the integral quantities. It is found that both the dimensional and non-dimensional displacement thickness, momentum thickness, energy thickness and the absolute wall shear stress are identical in both Blasius and Sakiadis flows at the asymptotic state.  相似文献

 In most studies concerning laminar natural convection along a vertical isothermal cylinder a linear relationship between fluid density and temperature has been used and kinematic viscosity and thermal diffusivity have been considered constant calculated at ambient temperature. However, it is known that the density–temperature relationship for water is non-linear at low temperatures and kinematic viscosity and thermal diffusivity are functions of temperature. In this study the problem of laminar natural convection of pure and saline water along a vertical isothermal cylinder has been investigated in the temperature range between 20 and 0 C taking into account the temperature dependence of ν, α and ρ. The results are obtained with the numerical solution of the boundary layer equations. The variation of ν, α and ρ with temperature has a strong influence on free convection characteristics.
