Nanoparticle assemblies interconnected with DNA triple helixes can be used to colorimetrically screen for triplex DNA binding molecules and simultaneously determine their relative binding affinities based on melting temperatures. Nanoparticles assemble only when DNA triple helixes form between DNA from two different particles and a third strand of free DNA. In addition, the triple helix structure is unstable at room temperature and only forms in the presence of triplex DNA binding molecules which stabilize the triple helix. The resulting melting transition of the nanoparticle assembly is much sharper and at a significantly higher Tm than the analogous triplex structure without nanoparticles. Upon nanoparticle assembly, a concomitant red-to-blue color change occurs. The assembly process and color change do not occur in the presence of duplex DNA binders and therefore provide a significantly better screening process for triplex DNA binding molecules compared to standard methods.  相似文献

In this work two monoiodothyronines, 3-T1 and 3'-T1, have been analyzed using electrospray ionization tandem mass spectrometry (ESI-MS/MS). Fragmentation patterns were proposed based on our data obtained by ESI-MS/MS. MS2 spectra in either negative or positive ion mode can be used to differentiate 3-T1 and 3'-T1. Based on the relative abundance of fragment ions in MS2 spectra in the negative ion mode, quantification of the 3-T1 and 3'-T1 isomers in mixtures is achieved without prior separation. Solid-phase extraction in combination with ESI-MS/MS provides a practicable procedure that can be used to determine the molar ratio of 3-T1 and 3'-T1 in human serum with an error less than 3%. The detection limits for 3-T1 and 3'-T1 were 0.5 and 0.7 pg/microL, respectively.  相似文献

Diiodothyronines 3,5-diiodothyronine (3,5-T2), 3',5'-diiodothyronine (3',5'-T2), and 3,3'-diiodothyronine (3,3'-T2) are important metabolites of 3,5,3'-triiodothyronine (T3) and 3,3',5'-triiodothyronine (rT3; reverse T3). In this paper, a novel and rapid method for identifying and quantifying 3,5-T2, 3',5'-T2 and 3,3'-T2 has been introduced using electrospray ionization tandem mass spectrometry (ESI-MS/MS). Fragmentation patterns were proposed on the basis of our data obtained by ESI-MS/MS. MS2 spectra in either negative ionization mode or positive ionization mode can be used to differentiate 3,5-T2, 3',5'-T2 and 3,3'-T2. Langmuir monolayers constitute a powerful platform for self‐assembly and organization of amhiphilic molecules. Controlling the structural features of condensed domains formed within Langmuir monolayers, however, is a challenging task. The formation of remarkably diverse condensed microstructures is demonstrated in binary monolayers comprising of a surfactant (octadecylmelamine) and a diacetylene monomer. The mole ratio between the two constituents and composition of the aqueous subphases (specifically pH and which dissolved metal ions are present) dramatically modulated the shapes and dimensions of microstructures formed at the air–water interface. The self‐assembled microstructures could be transferred from the water surface onto solid substrates, and subsequently further served as templates for gold coating, yielding electrically conductive microwires.  相似文献

Bottom‐up synthesis offers novel routes to obtain nanostructures for nanotechnology applications. Most self‐assembly processes are carried out in three dimensions (i.e. solutions); however, the large majority of nanostructure‐based devices function in two dimensions (i.e. on surfaces). Accordingly, an essential and often cumbersome step in bottom‐up applications involves harvesting and transferring the synthesized nanostructures from the solution onto target surfaces. We demonstrate a simple strategy for the synthesis and chemical transformation of tellurium nanorods, which is carried out directly at the solid–solution interface. The technique involves binding the nanorod precursors onto amine‐functionalized surfaces, followed by in situ crystallization/oxidation. We show that the surface‐anchored tellurium nanorods can be further transformed in situ into Ag2Te, Cu2Te, and SERS‐active Au–Te nanorods. This new approach offers a way to construct functional nanostructures directly on surfaces.  相似文献

采用溶剂蒸发自组装法调控载体形貌及孔道结构,成功制备了有序介孔氧化铝载体。以铬氧物种为活性组分,碱金属钾为助剂,采用浸渍法制备负载型催化剂,用于异丁烷催化脱氢反应,研究了反应温度、原料流速、催化剂粒径等因素对催化性能的影响。采用X射线粉末衍射、透射电子显微镜、N2物理吸附、氢气程序升温还原及热重等表征方法探讨了载体形貌、孔道结构与催化性能的构效关系,结果表明,低温下有利于控制异丙醇铝的水解和缩合及介孔γ-Al2O3的研制。与常规的γ-Al2O3相比,所制备的介孔γ-Al2O3的有更大的比表面积和良好的有序性,在600℃、101.325kPa、GHSV=1 000 h-1的条件下,10%(w/w)Cr2O3/γ-Al2O3催化剂性能最佳,异丁烷的转化率达63.1%,异丁烯的选择性达到85.5%。与传统的催化剂相比,介孔Cr2O3/γ-Al2O3催化剂具有大的比表面积,高度分散的活性组分,优良的催化性能和良好的抗积碳能力。  相似文献

Artifacts are the nemesis of trace element analysis in electron-excited energy dispersive X-ray spectrometry. Peaks that result from nonideal behavior in the detector or sample can fool even an experienced microanalyst into believing that they have trace amounts of an element that is not present. Many artifacts, such as the Si escape peak, absorption edges, and coincidence peaks, can be traced to the detector. Others, such as secondary fluorescence peaks and scatter peaks, can be traced to the sample. We have identified a new sample-dependent artifact that we attribute to Compton scattering of energetic X-rays generated in a small feature and subsequently scattered from a low atomic number matrix. It seems likely that this artifact has not previously been reported because it only occurs under specific conditions and represents a relatively small signal. However, with the advent of silicon drift detectors and their utility for trace element analysis, we anticipate that more people will observe it and possibly misidentify it. Though small, the artifact is not inconsequential. Under some conditions, it is possible to mistakenly identify the Compton scatter artifact as approximately 1% of an element that is not present.  相似文献

We report excellent reactivity and enantioselectivity of a C(2)-symmetric copper-bound N-heterocyclic carbene (NHC) in the hydrosilylation of a variety of structurally diverse ketones. This catalyst exhibits extraordinary enantioselctivity in the reduction of such challenging substrates as 2-butanone and 3-hexanone. Even at low catalyst loading (2.0 mol %), the reactions occur in under an hour at room temperature and often do not require purification beyond catalyst and solvent removal. The scope of this transformation was investigated in the reduction of 10 aryl-alkyl and alkyl-alkyl ketones.  相似文献
