The time-dependent Gross-Pitaevskii equation describes the dynamics of initially trapped Bose-Einstein condensates. We present a rigorous proof of this fact starting from a many-body bosonic Schr?dinger equation with a short-scale repulsive interaction in the dilute limit. Nanoparticles’ uptake by cancer cells upon reaching the tumor microenvironment is often the rate-limiting step in cancer nanomedicine. Herein, we report that the inclusion of aminopolycarboxylic acid conjugated lipids, such as EDTA- or DTPA-hexadecylamide lipids in liposome-like porphyrin nanoparticles (PS) enhanced their intracellular uptake by 25-fold, which was attributed to these lipids’ ability to fluidize the cell membrane in a detergent-like manner rather than by metal chelation of EDTA or DTPA. EDTA-lipid-incorporated-PS (ePS) take advantage of its unique active uptake mechanism to achieve >95 % photodynamic therapy (PDT) cell killing compared to <5 % cell killing by PS. In multiple tumor models, ePS demonstrated fast fluorescence-enabled tumor delineation within minutes post-injection and increased PDT potency (100 % survival rate) compared to PS (60 %). This study offers a new nanoparticle cellular uptake strategy to overcome challenges associated with conventional drug delivery. 相似文献

A fluorescent polypyridyl ruthenium complex was successfully prepared using an amide bond linkage to link two rhodamine moieties through bipyridine groups. Although photo‐induced electron transfer (PET) quenched the fluorescent intensity, the quantum yield of the rhodamine‐modified Ru(II) complex was 0.17 in water, sufficient for observing the fluorophore behaviour in biological systems. The rhodaminemodified Ru(II) complex was found to inhibit the bacterial growth of E. coli. In vitro fluorescence images of human hepatoma cells (SK‐Hep1) showed that a fluorescent polypyridyl ruthenium complex not only supported the above observation but also preferably accumulated in the cytoplasmic region inside the cell. These observations suggest that in addition to strong Ru–DNA interactions, Ru‐protein interactions in the cytoplasmic regions are strong and are therefore important to the development of metallopharmaceuticals. 相似文献

The roots of Fissistigma oldhamii (FO) are widely used as medicine with the effect of dispelling wind and dampness, promoting blood circulation and relieving pains, and its fruits are considered delicious. However, Hakka people always utilize its above-ground parts as a famous folk medicine, Xiangteng, with significant differences from literatures. Studies of chemical composition showed there were multiple aristolactams that possessed high nephrotoxicity, pending evaluation research about their distribution in FO. In this study, a sensitive, selective, rapid and reliable method was established to comparatively perform qualitative and semi-quantitative analysis of the constituents in roots, stems, leaves, fruits and insect galls, using an Ultra-High-Performance Liquid Chromatography coupled with Hybrid Quadrupole Orbitrap Mass Spectrometry (UPLC-Q-Exactive Orbitrap MS, or Q-Exactive for short). To make more accurate identification and comparison of FO chemicals, all MS data were aligned and screened by XCMS, then their structures were elucidated according to MSn ion fragments between the detected and standards, published ones or these generated by MS fragmenter. A total of 79 compounds were identified, including 33 alkaloids, 29 flavonoids, 11 phenylpropanoids, etc. There were 54 common components in all five parts, while another 25 components were just detected in some parts. Six toxic aristolactams were detected in this experiment, including aristolactam AII, AIIIa, BII, BIII, FI and FII, of which the relative contents in above-ground stems were much higher than roots. Meanwhile, multivariate statistical analysis was performed and showed significant differences both in type and content of the ingredients within all FO parts. The results implied that above-ground FO parts should be carefully valued for oral administration and eating fruits. This study demonstrated that the high-resolution mass spectrometry coupled with multivariate statistical methods was a powerful tool in compound analysis of complicated herbal extracts, and the results provide the basis for its further application, scientific development of quality standard and utilization. 相似文献

Detection of α‐Methylacyl‐CoA Racemase in Serum and Urine Using a Highly Sensitive Electrochemical Immunodetector 下载免费PDF全文
The detection of α‐methylacyl‐CoA racemase (AMACR), a novel biomarker for prostate cancer, is demonstrated in serum and urine using a novel immuno‐detection method. The detection system consists of a three‐electrode conventional electrochemical cell modified with a gating electrode for applying a gating voltage VG to the immune complex immobilized on the working electrode to provide signal amplification. The detection system is realized by integrating gating electrodes with screen‐printed electrodes. This detection method does not require involved sample preparation procedures. The detection was demonstrated in serum and urine samples on the nanogram/mL level with VG equal to 0.6 V. Detection in serum was also performed on the picogram/mL level with a limit of 100 picogram/mL with VG=0.6 V being a necessary condition. 相似文献

Three new donor–π–donor (D‐π‐D) tetrathienoacene (thieno[2′,3′:4,5]thieno[3,2‐b]thieno[2,3‐d]thiophene (TTA))‐cored chromophores, end‐functionalized with electron‐donating triphenylamine (TPA) groups, were developed and characterized for their two‐photon‐related properties by using both nano‐ and femtosecond laser pulses as the probing tools. TTA‐based chromophores exhibit stronger and more widely dispersed two‐photon absorption (2PA) than those of dithienothiophene (DTT)‐based congeners. As a consequence, the bithiophene‐conjugated TTA chromophore exhibits the highest maximum 2PA cross‐section value (up to 2500 GM) with good thermal stability, and thus, it is the best performing two‐photon chromophore among the studied model compounds. The bithiophene‐conjugated DTT analogue exhibits the second highest maximum two‐photon absorptivity of 1950 GM, which is nearly 7 times larger than that of previously reported DTT‐based chromophores. 相似文献
