Chlorogenic acid and caffeine are among the important components in coffee beans, determining the taste and aroma. In addition, phenols and antioxidants content possess vital health values. The main aim of this study is to determine the levels of caffeine and chlorogenic acid in several coffee samples of different origins and degrees of roasting. The coffee samples were extracted using hot water. The levels of caffeine and chlorogenic acid were quantified using high-performance liquid chromatography (HPLC) equipped with a diode array detector, a reverse phase system, and an ODS column (C18). Total phenol and antioxidant contents were previously determined for the same samples. The results showed that the highest content of caffeine was found in the medium roasted coffee (203.63 mg/L), and the highest content of chlorogenic acid content was found in the green coffee (543.23 mg/L). The results demonstrated a negative correlation between the chlorogenic acid levels with the degree of roasting, while it showed a positive correlation between the caffeine levels with the degree of roasting till a certain point where the levels dropped in the dark roasted coffee. The origin of coffee samples did not show any effect on any of the measured variables. Antioxidant effects of coffee samples were largely determined by chlorogenic acid content.  相似文献

Phenolic and antioxidant compounds have received considerable attention due to their beneficial effects on human health. The aim of this study is to determine the content of total phenols and antioxidants in fifty-two coffee samples of different origins, purchased from the Jordanian local market, and investigate the effect of the degree of roasting on the levels of these compounds. The coffee samples were extracted using the hot water extraction method, while Folin–Ciocalteu (FC) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay methods were used to analyze these compounds. The results showed that the highest content of total phenol (16.55 mg/g equivalent to GAE) was found in the medium roasted coffee, and the highest content of antioxidants (1.07 mg/g equivalent to TEAC) content was found in the green coffee. Only light and medium roasted coffee showed a significant correlation (p < 0.05, R2 > 0.95) between the average of total phenolic and antioxidant content. A negative correlation between the antioxidant content and the degree of roasting (p < 0.05, R2 > 0.95) were shown, while it did not correlate with phenolic contents. Previously, a positive correlation between antioxidant and chlorogenic acids content was observed, with no correlation between the origin of coffee samples nor heavy metal content, which was previously determined for the same coffee samples. These findings suggest that the antioxidant content for coffee extracts is largely determined by its chlorogenic acid content, rather than the coffee origin or total phenolic and heavy metals content.  相似文献

The search for new plasmonic materials that are low-cost, chemically and thermally stable, and exhibit low optical losses has garnered significant attention among researchers. Recently, metal nitrides have emerged as promising alternatives to conventional, noble-metal-based plasmonic materials, such as silver and gold. Many of the initial studies on metal nitrides have focused on computational prediction of the plasmonic properties of these materials. In recent years, several synthetic methods have been developed to enable empirical analysis. This review highlights synthetic techniques for the preparation of plasmonic metal nitride nanoparticles, which are predominantly free-standing, by using solid-state and solid–gas phase reactions, nonthermal and arc plasma methods, and laser ablation. The physical properties of the nanoparticles, such as shape, size, crystallinity, and optical response, obtained with such synthetic methods are also summarized.  相似文献

The first examples of α‐amino boronate complexes stabilized by amino cyanoborane complexation were tested as trypsin and chymotrypsin inhibitors, and they showed moderate inhibition. The structure of compound 1 that contains two different boron atoms reveals that the geometry around the boron atom in the cyano group is tetrahedral, whereas a trigonal planar geometry exists around the boron atom attached to two oxygen atoms and a carbon atom. An organometallic NO‐bidentate Schiff base, (2‐(1‐((1‐carboxyethyl)imino)ethyl) cyclopenta‐2,4‐dien‐1‐yl)(cyclopenta‐2,4‐dien‐1‐yl) iron (HL) was synthesized by condensation of 2‐acetylferrocene with amino acid alanine. Then its octahedral Cr (III), Mn (II), Fe (III), Co (II), Ni (II), Cu (II), Zn (II) and Cd (II) complexes were synthesized. All compounds were characterized on the basis of elemental analysis (C, H, N and M), molar conductivity, FT‐IR, UV–Vis, 1H‐NMR, SEM, mass analysis and thermal studies. Furthermore, computational studies of HL ligand have been carried out by DFT/B3LYP method. HOMO and LUMO energy values, chemical hardness‐softness, electronegativity, electrophilic index and other parameters were calculated. SEM micrographs of HL ligand and its [Cd (HL)(H2O)2Cl2].2H2O complex, showed that they were prepared in nano‐structure forms with particle size 54 and 41 nm, respectively. Antifungal and antibacterial activities of HL ligand and its metal complexes have been screened in vitro against different species such as Aspergillus fumigatus, Candida albicans, Bacillus subtilis, Staphylococcus aureus, Escherichia coli and Salmonella typhimurium. The synthesized compounds were evaluated for their anticancer activities against breast cancer cell line (MCF‐7) and normal melanocytes cell line (HFB‐4). It was found that [Co (HL)(H2O)2Cl2].3H2O complex had the lowest IC50 value (10.9 μg/ml) and hence was the most active one. Finally, the optimized structures of the Schiff base and its Co (II) complex have been used to accomplish molecular docking studies with receptors of 3HB5, 3MIW, 5IBV and 4WM8 to determine the most preferred mode of interaction.  相似文献

Human SELENOF is an endoplasmic reticulum (ER) selenoprotein that contains the redox active motif CXU (C is cysteine and U is selenocysteine), resembling the redox motif of thiol-disulfide oxidoreductases (CXXC). Like other selenoproteins, the challenge in accessing SELENOF has somewhat limited its full biological characterization thus far. Here we present the one-pot chemical synthesis of the thioredoxin-like domain of SELENOF, highlighted by the use of Fmoc-protected selenazolidine, native chemical ligations and deselenization reactions. The redox potential of the CXU motif, together with insulin turbidimetric assay suggested that SELENOF may catalyze the reduction of disulfides in misfolded proteins. Furthermore, we demonstrate that SELENOF is not a protein disulfide isomerase (PDI)-like enzyme, as it did not enhance the folding of the two protein models; bovine pancreatic trypsin inhibitor and hirudin. These studies suggest that SELENOF may be responsible for reducing the non-native disulfide bonds of misfolded glycoproteins as part of the quality control system in the ER.  相似文献

We developed and validated an HPLC method with intramolecular excimer-forming fluorescence derivatization to determine methylmalonic acid, a unique biochemical marker for methylmalonic aciduria. Methylmalonic acid in urine and an internal standard were derivatized with pyrenebutyric hydrazide and separated on a C8 column. The derivatives were detected by monitoring the fluorescence at 475 nm (excitation wavelength 345 nm). At a signal-to-noise ratio of 3, the detection limit was 0.33 pmol on the column and the calibration curve was linear up to 1 mmol[sol ]L in urine. In a retrospective study on a relatively large number of known methylmalonic aciduria cases (n = 48), the method enabled us to differentiate methylmalonic aciduria cases from healthy controls (n = 52), regardless of age of patients at sampling or years of specimen storage. No interference was observed from isomeric or other dicarboxylic acids, or other urine constituents. As described, the method can be used retrospectively or prospectively for the diagnosis of methylmalonic aciduria and can be easily adopted by laboratories with no access to gas chromatography-mass spectrometry.  相似文献

The proton transfer reactions between chromotropic acid (CTA) and some amines including benzylamine (BA), triethylamine (TEA), pyrrolidine (PY) and 1,8-bis(dimethylamino) naphthalene (DMAN) have been investigated spectrophotometrically in methanol. A long wavelength band at 365 nm has been recorded due to the proton transfer (PT) complex formation. The proton transfer equilibrium constants KPT were estimated utilizing the minimum–maximum absorbances method. It has been found that KPT were not depend on the amine pKa values, but strongly depend on the formed structures of the PT complexes. Jobs method of continuous variations and photometric titrations were applied to identify the compositions of the formed PT complexes where 1:1 complexes (proton donor: proton acceptor) were produced. Due to the rapidity and simplicity of the proton transfer reactions and the stability of the formed complexes, a rapid and accurate spectrophotometric method for the determination of CTA was proposed for the first time.  相似文献
