We previously studied the response of mammalian cultured cells to weak, 60 Hz-electromagnetic (EM) fields. Two time constants, similar to those observed in chemotaxis, were found to govern the cellular response to the field. We concluded that a system of temporal sensing, similar to that employed in chemotaxis by motile bacteria, was operative. We termed the shorter time (approximately 0.1 s) the "sensing" time, and the longer time (approximately 10 s) the "memory" time. To investigate the possibility that temporal sensing was a general property of EM field-cell interaction, the temporal properties of another EM field-induced effect was studied. The EM field-induced protection against the effects of extreme hypoxia was examined in chick embryos. Embryos were exposed to 60 Hz-magnetic fields, the amplitudes of which were regularly altered throughout the 20-min exposure. Alteration was accomplished either by turning the field off and on at regular intervals (1-50 s), or by introducing brief (10 or 100 ms), zero amplitude gaps, once each second, throughout exposure. When the field was turned on and off at 0.1 s intervals, the protective effect conferred by a constant field was lost. At progressively longer on/off intervals, protection was progressively restored, maximizing at intervals of 10-30 s. Gapping the magnetic field for 10 ms, each second of exposure conferred the same protection as that observed for an uninterrupted field, but gapping the field at 100 ms each second produced a significant reduction in protection. These data exhibit remarkable consistency with those obtained in similar temporal studies of the magnetic field-induced enhancement of ornithine decarboxylase activity in L929 fibroblasts. A method is described for the identification and relative quantification of proteomes using accurate mass tags (AMT) generated by nLC-dual ESI-FT-ICR-MS on a 7T instrument in conjunction with stable isotope labeling using 16O/18O ratios. AMTs were used for putative peptide identification, followed by confirmation of peptide identity by tandem mass spectrometry. For a combined set of 58 tryptic peptides from bovine serum albumin (BSA) and human transferrin, a mean mass measurement accuracy of 1.9 ppm +/-0.94 ppm (CIM99%) was obtained. This subset of tryptic peptides was used to measure 16O/18O ratios of 0.36 +/- 0.09 (CIM99%) for BSA (micro = 0.33) and 1.48 +/- 0.47 (CIM99%) for transferrin (micro = 1.0) using a method for calculating 16O/18O ratios from overlapping isotopic multiplets arising from mixtures of 16O, 18O1, and 18O2 labeled C-termini. The model amino acid averagine was used to calculate a representative molecular formula for estimating and subtracting the contributions of naturally occurring isotopes solely as a function of peptide molecular weight. The method was tested against simulated composite 16O/18O spectra where peptide molecular weight, 16O/18O ratio, 18O1/18O2 ratios, and number of sulfur atoms were varied. Relative errors of 20% or less were incurred when the 16O/18O ratios were less than three, even for peptides where the number of sulfur atoms was over- or under-estimated. These data demonstrate that for biomarker discovery, it is advantageous to label the proteome representing the disease state with 18O; and the method is not sensitive to variations in 18O1/18O2 ratio. This approach allows a comprehensive differentiation of expression levels and tentative identification via AMTs, followed by targeted analysis of over- and under-expressed peptides using tandem mass spectrometry, for applications such as the discovery of disease biomarkers. 相似文献

A miniaturised technique to analyse and detect heterocyclic aromatic amines (HAs) using micro solid-phase extraction (SPE) coupled on-line (in-capillary) to capillary electrophoresis (CE) separation with nanospray (nESI) mass spectrometry (MS) detection has been developed. HAs are mutagenic and carcinogenic compounds formed at low levels in protein-rich food during cooking. Due to the low concentrations of HAs and the high complexity of the matrix in which they exist, sensitive and selective analytical methods are required for quantification. SPE was performed on a packed bed of C18 particles inside the CE capillary, which minimised the dead volume. The on-line coupling of SPE, CE and nESI-MS reduced the time for extraction and identification to less than half an hour, which will allow for screening of several samples per day. The new technique provides short analysis time, low sample and solvent consumption, and HAs in standard solutions were easily detected at 12–17 fmol injections, and in spiked urine samples at 750–810 fmol injections. 相似文献

Near-UV irradiation of structurally characterized [Re(I)(CO)3(1,10-phenanthroline)(Q107H)](W48F/Y72F/H83Q/Y108W)AzM(II) [Az = Pseudomonas aeruginosa azurin, M = Cu, Zn]/[Co(NH3)5Cl]Cl2 produces a tryptophan radical (W108*) with unprecedented kinetic stability. After rapid formation (k = 2.8 x 106 s-1), the radical persists for more than 5 h at room temperature in the folded ReAzM(II) structure. The absorption spectrum of ReAz(W108*)M(II) exhibits maxima at 512 and 536 nm. The structure and chemical composition are determined by X-ray diffraction analysis and element analysis. The four S2 sites of the big cyclic ligand [As4W40O140]28- are occupied by two Na+ and two Ni2+ respectively, and each site supplies four Od coordinating to metal ion. The coordination number of Ni2+ is six, and that of two Na+ is five and six respectively. Observation that rates of dehydrobromination of trans-β-bromostyrene (1) and the Hofmann degradation of tetrabutyl ammonium cation depend on strength of base in different ways and that treatment of 1 with base results in fast abstraction of the β-proton imply the possibility that the dehydrobromination of 1 could proceed via α-elimination and Ph migration. In order to clarify this question, β-13C-labeled 1 was obtained and subjected to PTC dehydrobromination which proceeds without migration of Ph. Novel rhenium(I) tricarbonyl complexes have been prepared by reactions of (Et4N)2[Re(CO)3Br3] with acetylpyridine benzoylhydrazone, Hapbhyd, di(2‐pyridyl)ketone benzoylhydrazone, Hpy2bhyd, bis(2‐pyridine)ketone, py2CO, and pyridinealdehyde terephtalaldehydebishydrazone, pytehyd. The ligands remain protonated when no supporting base is added and the following complexes have been isolated: [Re(CO)3Br(Hapbhyd)], [Re(CO)3Br(Hpy2bhyd‐py, hyd)], [Re(CO)3Br(Hpy2bhyd‐py1, py2)], [Re(CO)3Br(py2CO‐N, N)] and [Re(CO)3Br(pytehyd)]. Addition of triethyl amine results in deprotonation of Hapbhyd and the formation of [Re(CO)3(OH2)(apbhyd)], whereas Hpy2bhyd is hydrolysed and a rhenium complex with the monoanionic bis(2‐pyridyl)hydroxymethanolato ligand, {py2C(OH)O}‐, is formed. The same compound, [Re(CO)3{py2C(OH)O}], is obtained when triethyl amine and water are added to a mixture of (Et4N)2[Re(CO)3Br3] and py2CO. Kinetic and catalytic aspects of DMT transesterification reaction, key step in the industrial production of polyethylenterephtalate (PET), have been deepened also through the use of model molecules such as CH3COO–Ph–X. These molecules give the same reactions with ethylene glycol occurring in the DMT transesterification too. The use of model molecules in the study of this reaction seems therefore to be promising in perspective. As we observed, however, the nature of the para-substituent X has a very strong influence on the reaction rate, when salts of bivalent metals such as Zn, Cd, Co, Mg, Mn, are used as catalysts, while this influence is much smaller when a tetravalent metal, as Ti, is used. This fact suggests that the transesterification mechanism operating in the two cases is different and this suggestion has been confirmed by applying Hammett's approach to the available kinetic data. Experimental kinetic runs were all performed at 193°C, by withdrawing small samples of the reaction mixture at different reaction times. These samples were gaschromatographically analyzed. We found volcano shaped curves of the reaction rates as a function of the metal ion acidities and we obtained different trends with a maximum of activity shifted from a metal to another for different substrates. A comparison of the kinetic results obtained, respectively, with the model molecules and DMT will be reported too. 相似文献

Summary A high-performance liquid chromatography (HPLC) assay for estriol in nonpregnancy urine is described. After Enzymic hydrolysis, the estriol is extracted from urine by the sorbent trap technique utilizing graphitized carbon black (Carbopack B). After some washing steps, estriol is desorbed by a suitable solvent system. After solvent removal, the sample is injected into an HPLC column for estriol quantification. Analytical recovery of estriol was 96.1%. The precision of the method was 2.6 and 4.9% respectively at 145 and 10.6ng/ml of urine. The limit of sensitivity was set at 0.8 ng/ml of urine. The mean contents of estriol in the follicular and luteal phases were respectively 11.3 and 38.8 ng/ml of urine.Dedicated to Prof. Dr. A. Liberti on the occasion of his 70th birthday. 相似文献
