Three principal methods have been developed for measuring femtomoles of damage in nanogram quantities of non-radioactive DNA. Lesions which can be quantified include single and double strand breaks, alkali labile sites including apurinic and apyrimidinic sites, and pyrimidine dimers. The first in vitro method measures the conversion of supercoiled DNA to relaxed or linear molecules, and can detect up to four lesions per molecule. The second in vitro method (supercoil depletion) assesses the fraction of intact linear molecules of homogeneous length, and allows detection of 8 lesions/molecule. The third method, measurement of molecular length distributions of DNAs of heterogeneous length, reveals the extent of DNA damage and repair in vivo or in vitro.  相似文献

The complex formation of vanadium(IV) with 1,3,5-triamino-1,3,5-trideoxy-cis-inositol (taci) and 1,3,5-trideoxy-1,3,5-tris(dimethylamino)-cis-inositol (tdci) was studied in aqueous solution and in the solid state. The formation constants of [V(IV)O(taci)](2+), [V(IV)O(tdci)](2+), and [V(IV)(tdci)(2)](4+) and of the deprotonation product [V(IV)(tdci)(2)H(-)(1)](3+) were determined (25 degrees C, 0.1 M KNO(3)). Macro- or microquantities of gallium can be separated from iron by precipitating the latter with sodium hydroxide in the presence of EDTA. Nearly all the gallium remains in solution. After the separation of iron, gallium is extracted with tributylphosphate from 3 N hydrochloric acid medium, and then re-extracted into water. Gallium is finally determined by precipitation with cupferron and ignition to the oxide, or for trace amounts of gallium, by colorimetric determination with rhodamine B. N-Nosyl-alpha-amino acid methyl esters are methylated quantitatively with diazomethane. After proper deprotection of the amino function by treatment with the reagent system mercaptoacetic acid/sodium methoxide, the obtained N-methyl amino acid methyl esters are coupled with N-Fmoc amino acid chlorides to afford the corresponding dipeptides. The obtained products do not show any detectable extent of racemization by (1)H NMR and HPLC.  相似文献

Treatment of trans-[PtCl(4)(RCN)(2)] (R = Me, Et) with R'OH (R' = Me, Et, n-Pr, i-Pr, n-Bu) at 45 degrees C in all cases allowed the isolation of the trans-[PtCl(4)[(E)-NH=C(R)OR'](2)] imino ester complexes, while the reaction between cis-[PtCl(4)(RCN)(2)] and the least sterically hindered alcohols (methanol and ethanol) results in the formation of cis-[PtCl(4)[(E)-NH=C(R)OR'](2)] (R/R' = Me/Me) or trans-[PtCl(4)[(E)-NH=C(Et)OR'](2)] (R' = Me, Et), the latter being formed via thermal isomerization (ROH, reflux, 3 h) of the initially formed corresponding cis isomers. The reaction between alcohols R'OH and cis-[PtCl(4)(RCN)(2)] (R = Me, R' = Et, n-Pr, i-Pr, n-Bu; R = Et; R' = n-Pr, i-Pr, n-Bu), exhibiting greater R/R' steric congestion, allowed the isolation of cis-[PtCl(4)[(E)-NH=C(R)OR'][(Z)-NH=C(R)OR']] as the major products. The alcoholysis reactions of poorly soluble [PtCl(4)(RCN)(2)] (R = CH(2)Ph, Ph) performed under heterogeneous conditions, directly in the appropriate alcohol and for a prolonged time and, for R = Ph, with heating led to trans-[PtCl(4)[(E)-NH=C(R)OR'](2)] (R = CH(2)Ph, R' = Me, Et, n-Pr, i-Pr; R = Ph, R' = Me) isolated in moderate yields. In all of the cases, in contrast to platinum(II) systems, addition of R'OH to the organonitrile platinum(IV) complexes occurs under mild conditions and does not require a base as a catalyst. The formed isomerically pure (imino ester)Pt(IV) complexes can be reduced selectively, by Ph(3)P=CHCO(2)Me, to the corresponding isomers of (imino ester)Pt(II) species, exhibiting antitumor activity, without change in configuration of the imino ester ligands. Furthemore, the imino esters NH=C(R)OR' can be liberated from both platinum(IV) and platinum(II) complexes [PtCl(n)[H=C(R)OR'](2)] (n = 2, 4) by reaction with 1,2-bis(diphenylphosphino)ethane and pyridine, respectively. All of the prepared compounds were characterized by elemental analyses (C, H, N), FAB mass spectrometry, IR, and (1)H, (13)C[(1)H], and (195)Pt (metal complexes) NMR spectroscopies; the E and Z configurations of the imino ester ligands in solution were determined by observation of the nuclear Overhauser effect. X-ray structure determinations were performed for trans-[PtCl(4)[(E)-NH=C(Me)OEt](2)] (2), trans-[PtCl(4)[(E)-NH=C(Et)OEt](2)] (10), trans-[PtCl(4)[(E)-NH=C(Et)OPr-i](2)] (11), trans-[PtCl(4)[(E)-NH=C(Et)OPr-n](2)] (12), and cis-[PtCl(4)[(E)-NH=C(Et)OMe](2)] (14). Ab initio calculations have shown that the EE isomers are the most stable ones for both platinum(II) and platinum(IV) complexes, whereas the most stable configurations for the ZZ isomers are less stable than the respective EZ isomers, indicating an increase of the stability on moving from the ZZ to the EE configurations which is more pronounced for the Pt(IV) complexes than for the Pt(II) species.  相似文献

The temperature dependence of limiting apparent molal volumes ° in water for some alcohols (methanol, ethanol, 1-propanol, 2-butanol, 3-pentanol, 3-hexanol, 2,5-hexanediol, cyclopentanol, cyclohexanol, cycloheptanol, and 1,4-cyclohexanediol) and ethers (trimethylene oxide, tetrahydrofuran, tetrahydropyran, 1,3-dioxolane, 1,3-dioxane, 1,4-dioxane, 1,3-dioxepane, 1,3,5-trioxane, dimethoxymethane, 1,2-dimethoxyethane, diethoxymethane, and diethyl ether) has been studied in the temperature range 10–50°C by means of an automatic digital-readout dilatometer. Values of the thermal expansion coefficient * = (1/°)(°/T)p have been obtained at several temperatures and discussed together with literature data on expansibilities of related compounds. The data show a wide spectrum of values of * at low temperatures which is narrowed at the higher ones. The expansibilities of monofunctional alcohols increase with increasing temperature; the opposite effect is observed for polyhydric alcohols. The *'s of ethers are very slightly temperature dependent and are much higher, at low temperature, than those of alcohols having the same ratio ofn o/nc. These results are discussed in terms of solutewater interactions, and a possible interpretation is put forward.  相似文献

A method based on cloud point extraction (CPE) separation/preconcentration of trace cadmium as a prior step to its determination by graphite furnace atomic absorption spectrometry (GFAAS) has been developed. If the system temperature is higher than the cloud point temperature (CPT) of the nonionic surfactant of p-octyl polyethyleneglycolphenyether (Triton X-100), the complex of Cd2+ with 1-(2-pyridylazo)-2-naphthol (PAN) could be extracted into surfactant-rich phase. The chemical variables affecting CPE were evaluated and optimized. Under the optimum conditions, preconcentrating 10.0 mL of water samples permitted a limit of detection of 5.9 ng · L−1 (3σ) for cadmium with an enhancement factor of 50 and a relative standard deviation of 2.1% (n = 11, c = 2.0 ng · mL−1). The method was applied to the determination of cadmium in reference material and water samples with satisfactory results.  相似文献

The conformational stability of individual DNA topoisomers depends on the concentration of DNA intercalating drugs. To study the DNA-drug interaction, we used ethidium bromide (EtBr) and negative supercoiled pUC19 as a model system. The effects of two anthracyclines widely used in cancer therapy, daunorubicin (Dau) and doxorubicin (Doxo), and EtBr were compared. In spite of their different chemical structures and intercalation mode, all intercalating agents show similar effects on the conformational stability of supercoiled DNA. Our observations show that the studied intercalators have at least two main effects on the supercoiled DNA: (i) they decrease the level of negative supercoiling and, at certain concentrations, they may induce positive supercoiling in DNA; (ii) a temperature increase can cause a recovery of negative supercoiling in DNA. The conformational stability of plasmid DNA-drug complexes has been investigated by temperature gradient gel electrophoresis (TGGE). We demonstrate the suitability of TGGE for this purpose, because it offers a global view on DNA-drug complexes over a continuous range of temperature. Images of DNA plasmids adsorbed onto a substrate at different temperatures and drug concentrations were acquired by atomic force microscopy (AFM), allowing us to distinguish directly the conformation chirality assumed by the plasmid under different conditions confirming TGGE results. Our detection system allows to characterize unknown drugs and to determine their intercalating properties.  相似文献

The effect of monochromatic UVB (280–315 nm) irradiation on the photosynthetic activity of the marine green alga Dunaliella salina was investigated by monitoring the rate of oxygen exchange. Samples were irradiated with narrow bands centered at different UV wavelengths (281, 290, 300, 310 and 322 nm). In a first set of experiments the samples were preirradiated keeping constant the irradiance and varying exposure time. By increasing UVB fluence, a wavelength-dependent decrease in the rate of oxygen production was observed. In a second set of experiments oxygen exchange was monitored simultaneously to UVB irradiation. The decline in the rate of oxygen production was sharper at short wavelengths and faster with increasing UVB photon flux density. The photon fluence action spectra derived from the two sets of experiments are in good agreement and indicate a decrease in the UVB-inhibiting effectiveness of two orders of magnitude from about 0.2 (mmol m-2)-1 at 281 nm to about 0.003 (mmol m-2)-1 at 322 nm. The photon flux density action spectrum derived from the second set of experiments indicates a smoother decrease from about 0.2 (pmol s-l m-2)-1 at 281 nm to about 0.05 (pmol s-l m-z)-l at 310 nm.  相似文献

A dozen Iris species (Iridaceae) are considered traditional remedies in Kurdistan, especially for treating inflammations. Phytochemical studies are still scarce. The information reported in the literature about Iris species growing in Kurdistan has been summarized in the first part of this paper, although, except for Iris persica, investigations have been performed on vegetal samples collected in countries different from Kurdistan. In the second part of the work, we have investigated, for the first time, the contents of the methanolic extracts of Iris postii aerial parts and rhizomes that were collected in Kurdistan. Both extracts exhibited a significant dose-dependent free radical scavenging and total antioxidant activities, comparable to those of ascorbic acid. Medium-pressure liquid chromatographic separations of the two extracts afforded l-tryptophan, androsin, isovitexin, swertisin, and 2″-O-α-l-rhamnopyranosyl swertisin from the aerial parts, whereas ε-viniferin, trans-resveratrol 3,4′-O-di-β-d-glucopyranoside, and isotectorigenin were isolated from the rhizomes. This is the first finding of the last three metabolites from an Iris species. The various remarkable biological activities of isolated compounds scientifically sustain the traditional use of I. postii as a medicinal plant.  相似文献
