Some of the most active scientific research fronts of the past decade are centered on ionic liquids. These fluids present characteristic surface behavior and distinctive trends of their surface tension versus temperature. One way to explore and understand their unique nature is to study their surface properties. The structure of psychrophilic chitinase (CHI II) from Glaciozyma antarctica PI12 has yet to be studied in detail. Due to its low sequence identity (<30?%), the structural prediction of CHI II is a challenge. A 3D model of CHI II was built by first using a threading approach to search for a suitable template and to generate an optimum target-template alignment, followed by model building using MODELLER9v7. Analysis of the catalytic insertion domain structure in CHI II revealed an increase in the number of aromatic residues and longer loops compared to mesophilic and thermophilic chitinases. A molecular dynamics simulation was used to examine the stability of the CHI II structure at 273, 288 and 300?K. Structural analysis of the substrate-binding cleft revealed a few exposed aromatic residues. Substitutions of certain amino acids in the surface and loop regions of CHI II conferred an increased flexibility to the enzyme, allowing for an adaptation to cold temperatures. A substrate binding comparison of CHI II with the mesophilic chitinase from Coccidioides immitis, 1D2K, suggested that the psychrophilic adaptation and catalytic activity at low temperatures were achieved through a reduction in the number of salt bridges, fewer hydrogen bonds and an increase in the exposure of the hydrophobic side chains to the solvent.  相似文献

Production of high performance conductive textile yarn fibers for different electronic applications has become a prominent area of many research groups throughout the world. We have used oxidative chemical vapor deposition (OCVD) technique to coat flexible and high strength polyester yarns with conjugated polymer, poly(3,4‐ethylenedioxythiophene) (PEDOT) in presence of ferric (III) chloride (FeCl3) oxidant. OCVD is an efficient solvent free technique used to get uniform, thin, and highly conductive polymer layers on different substrates. In this paper, PEDOT‐coated polyester (PET) yarns were prepared under specific reaction conditions, and the electrical, mechanical and thermal properties were compared to previously studied PEDOT‐coated viscose yarns. Scanning electron microscopy (SEM) and FT‐IR analysis revealed that polymerization of PEDOT on the surface of the polyester yarns has been taken place successfully and structural analysis showed that PEDOT has strong interactions with viscose yarns as compared to PET yarns. The voltage–current (VI) characteristics showed that PET yarns are more conductive than PEDOT‐coated viscose yarns. The variation in the conductivity of PEDOT‐coated yarns and the heat generation properties during the flow of current through coated yarns for longer period of time, was studied by time–current (tI) characteristics. Thermogravimeteric analysis (TGA) was employed to investigate the thermal properties and the amount of PEDOT in PEDOT‐coated PET yarns compared to PEDOT‐coated viscose. The effect of PEDOT coating and ferric (III) chloride concentration on the mechanical properties of coated yarns was evaluated by tensile testing. Pilot plant trials were conducted in a corn wet mill with a 7000-L membrane recycle bioreactor (MRB) that integrated ceramic microfiltration membranes in a semi-closed loop configuration with a stirred-tank reactor. Residence times of 7.5–10 h with ethanol outputs of 10–11.5% (v/v) were obtained when the cell concentration was 60–100 g/L drywt of yeast, equivalent to about 109−1010 cells/mL. The performance of the membrane was dependent on the startup mode and pressure management techniques. A steady flux of 70 L/(m2·h) could be maintained for several days before cleaning was necessary. The benefits of the MRB include better productivity; a clear productstream containing no particulates or yeast cells, which should improve subsequent stripping and distillation operations; and substantially reduced stillage handling. The capital cost of the MRB is $21–$34/(m3·yr) ($0.08–$0.13/[gal·yr]) of ethanol capacity. Operating cost, including depreciation, energy, membrane replacement, maintenance, labor, and cleaning, is $4.5–9/m3 ($0.017–$0.034/gal) of ethanol.  相似文献

Abstract

The olive (Olea europaea L.) is a worldwide famous tree crop having valuable commercial role due to nutritional as well as therapeutic benefits obtained from its edible oil. Eight varieties of Olea europaea L. fruits were obtained from three Agricultural research institutes located in different areas of Pakistan including Quetta, Lorallia and Zhob to extract and determine their oil contents by conventional method as solvent extraction (soxhlet apparatus). Statistical analysis showed significant (p?=?0.002) difference in oil composition of collected plants. Highest amount (percentage) of oil was obtained from Gemlik variety (65%) from Lorallia and lowest oil content from Dolece-agogia variety (17.5%) collected from Quetta. Further research is under consideration to rule out best suitable factors including some soil and environmental factors to obtained more potential cultivars of Olea europaea in Pakistan.  相似文献

A practical, operationally simple preparation of 2-(trimethylsilyl)methyl-2-propen-1-ol derivatives is described. The cobalt catalyzed coupling of a protected vinyl halide with trimethylsilylmethylmagnesium chloride shows excellent functional group tolerance and provides these synthetically useful allyl silanes in good overall yield. By this method, the use of highly concentrated organolithium reagents, complex reaction protocols, and expensive starting materials is avoided.  相似文献

B physics at LEP

The Z factory at LEP is like a mini B factory. About 20% of the time a Z decays to a pair of b flavoured hadrons. As a result, this has provided a major boost to B physics studies. The four sophisticated LEP detectors, with millions of Z events, have provided a variety of interesting results. Only a snapshot of the major developments can be covered in one review. We have considered the results related to coupling, in particular, the measurements of Rbbhad the forward backward charge asymmetry. The b hadron properties, like inclusive and exclusive lifetime measurements are summarised briefly. Measurements of oscillation parameter as well as the evidence for time dependent evolution have been discussed. Studies of interesting decay modes have been mentioned.  相似文献
