Antibiotic resistant infections are projected to cause over 10 million deaths by 2050, yet the development of new antibiotics has slowed. This points to an urgent need for methodologies for the rapid development of antibiotics against emerging drug resistant pathogens. We report on a generalizable combined computational and synthetic approach, called antibody-recruiting protein-catalyzed capture agents (AR-PCCs), to address this challenge. We applied the combinatorial protein catalyzed capture agent (PCC) technology to identify macrocyclic peptide ligands against highly conserved surface protein epitopes of carbapenem-resistant Klebsiella pneumoniae, an opportunistic Gram-negative pathogen with drug resistant strains. Multi-omic data combined with bioinformatic analyses identified epitopes of the highly expressed MrkA surface protein of K. pneumoniae for targeting in PCC screens. The top-performing ligand exhibited high-affinity (EC50 ∼50 nM) to full-length MrkA, and selectively bound to MrkA-expressing K. pneumoniae, but not to other pathogenic bacterial species. AR-PCCs that bear a hapten moiety promoted antibody recruitment to K. pneumoniae, leading to enhanced phagocytosis and phagocytic killing by macrophages. The rapid development of this highly targeted antibiotic implies that the integrated computational and synthetic toolkit described here can be used for the accelerated production of antibiotics against drug resistant bacteria.

Diced Roma tomatoes were treated with gamma irradiation and evaluated for changes in microbial, physical, chemical and sensory properties. Dosages for Trial 1 were 0.0, 0.39, 0.56 and 1.82 kGy and in Trial 2, 0.0, 0.50, 1.24 and 3.70 kGy. Irradiation at 3.70 kGy resulted in no aerobic populations through day 12 and significantly fewer colonies through day 15 whereas yeast and mold populations experienced a 2 log reduction through day 12. Color, titratable acidity, and °Brix were not significantly affected by irradiation. Tissue firmness decreased with increasing dose but not with storage time. Treatment with 3.7 kGy decreased firmness by 50% and 20% with 0.5 kGy, however, the reduced firmness induced by 0.50 kGy was undetected by a 9 member trained sensory panel. A significant (p0.05) inverse correlation between changes in texture and water-soluble pectin (WSP) was determined while total pectin remained relatively constant and oxalate soluble pectin content decreased slightly with irradiation dose. The significant inverse correlation between the loss of firmness and WSP indicates that the changes in WSP play an important role in the tissue softening of tomatoes, This study indicates that irradiation at 0.5 kGy can reduce microbial counts substantially to improve microbial shelf life without adverse effects on sensory qualities.  相似文献

An intercrosslinked network of cyanate ester (CE)-bismaleimide (BMI) modified epoxy matrix system was made by using epoxy resin, 1,3-dicyanatobenzene and bismaleimide (N,N-bismaleimido-4,4-diphenyl methane) with diaminodiphenylmethane as curing agent. BMI-CE-epoxy matrices were characterised using differential scanning calorimetry (DSC), thermogravimetric analysis (TGA) and heat deflection temperature (HDT) analysis. The matrices, in the form of castings, were characterised for their mechanical properties such as tensile strength, flexural strength and unnotched Izod impact test as per ASTM methods. Mechanical studies indicated that the introduction of cyanate ester into epoxy resin improves the toughness and flexural strength with reduction in tensile strength and glass transition temperature, whereas the incorporation of bismaleimide into epoxy resin influences the mechanical and thermal properties according to its percentage content. DSC thermograms of cyanate ester as well as BMI modified epoxy resin show an unimodal reaction exotherm. Electrical properties were studied as per ASTM method and the morphology of the BMI modified epoxy and CE-epoxy systems were studied by scanning electron microscope.  相似文献

Different concentrations of Ho3+-doped lead borate titanate aluminum fluoride (LBTAFHo) glasses with chemical composition of PbO-H3BO3-TiO2-AlF3-Ho2O3 were prepared by the melt quenching method. The spectral properties were investigated using the absorption, emission and decay measurements. The experimental oscillator strengths were calculated from the area under the absorption bands. Applying Judd-Ofelt theory, the intensity parameters (Ωλ=2, 4, 6) were calculated, by the least square fit approach from which the radiative transition rates, luminescence branching ratios and radiative decay times were determined. The photoluminescence spectra revealed the quenching of luminescence intensity beyond 1.0 mol% of Ho3+ ion concentration. To investigate the luminescence potentiality of 5F45I8 emission level, the effective bandwidth and the stimulated emission cross-section were determined. The quenching in experimental decay time is attributed to the resonance energy transfer among the excited Ho3+ ions.  相似文献

A detailed investigation on the effect of solvent polarity, temperature, and microwave irradiation on periselectivity in cycloaddition reactions of pentafulvenes with 3-oxidopyrylium betaine is described. The base catalyzed generation of 3-oxidopyrylium betaine in CHCl3 resulted in the exclusive formation of [6+3] adducts. With increase in solvent polarity and temperature, mixtures of [6+3] and [3+2] adducts were formed, where as under microwave irradiation, [3+2] adducts were formed exclusively. The composite hydrogel of a nanoscale metal–organic framework (NMOF) and nanoclay has emerged as a new soft-material with advanced properties and applications. Herein, we report a facile synthesis of a hydrogel nanocomposite by charge-assisted self-assembly of Pd@ZIF-8 nanoparticles with Laponite® nanoclay which coat the surface of Pd@ZIF-8 nanoparticles. Such surface coating significantly enhanced the thermal stability of the ZIF-8 compared to the pristine framework. Further, the Pd@ZIF-8+LP hydrogel nanocomposite shows better size-selective catalytic hydrogenation of olefins than Pd@ZIF-8 nanoparticles based on selective diffusion of the substrate.  相似文献

High surface area silicon carbide-derived carbons (Si-CDCs) synthesized by chlorination of beta silicon carbide (βSiC) with two different particle sizes (6 μm and 50 nm) show different porosities with graphitic structure. Transmission electron microscopy, Raman spectroscopy and argon (Ar) and carbon dioxide (CO2) sorption analyses are used to examine the textural properties of the Si-CDCs. The results show that the particle size of the precursor affects the surface area and porosity of carbons. Furthermore, an additional heat treatment of the Si-CDC with 50-nm particle size for 24 h at 1,000 °C results in a collapse of the pore structure and reduces the surface area. The capacitive behaviours are investigated in H2SO4 and in tetraethyl ammonium tetrafluoroborate (TEABF4)/acetonitrile (AN). The electrochemical performance of the Si-CDCs is influenced by the particle size, surface area, pore volume and pore size distribution. The Si-CDCs exhibit capacitances in 1 M H2SO4 of up to 179 F g?1 and very stable charge–discharge performance over 5,000 cycles. This study shows the crucial importance of ultramicropores less than 1 nm combined with nanosized particles for achieving high capacitance in aqueous electrolyte. Moreover, the graphitic degree at the surface of the Si-CDCs enhances considerably the rate capability and stability in both electrolytes.  相似文献
