Evaluation and validation of the ABI 3700, ABI 3100, and the MegaBACE 1000 capillary array electrophoresis instruments for use with short tandem repeat microsatellite typing in a forensic environment

The demand for high-throughput DNA profiling has increased with the introduction of national DNA databases and has led to the development of automated methods of short tandem repeat (STR) profile production; however, a potential bottleneck still exists at the gel electrophoresis stage. Capillary electrophoresis sequencers capable of processing 96 samples with considerably reduced manual intervention are now available. In this paper, we compare the ABI Prism 377 slab-gel sequencer currently used by the Forensic Science Service with three leading capillary array electrophoresis instruments: the ABI Prism 3700, the Amersham MegaBACE 1000 and the 16-capillary ABI Prism 3100. We describe the experiments used to evaluate and validate these platforms for forensic use with the STR multiplex Ampf/STR SGMplus [1, 2], along with comparative data from the ABI Prism 377 sequencer.     

Magnetoelectric compounds with two sets of magnetic sublattices: UCrO4 and NdCrTiO5

Using magnetic and magnetoelectric (ME) powder susceptibility measurements, the low temperature magnetic properties of antiferromagnetic UCrO₄ and NdCrTiO₅ have been studied. Their Néel temperatures T_N are 44.5 and 20.5°K, respectively, the Cr³⁺ spin systems of both materials ordering cooperatively at T_N. Below T_N, the U⁵⁺ and Nd³⁺ moments are polarized due to their exchange interaction with the ordered Cr³⁺ spins. It is argued that, for both compounds, each of the two spin systems contributes to the ME susceptibilities. They are thus the first known ME materials possessing two distinct magnetic sublattices. The effective magnetic moments calculated from the magnetic susceptibilities are in good agreement with those previously reported by neutron diffraction studies.     

High-resolution 17O NMR spectroscopy of wadsleyite (beta-Mg2SiO4)

The sensitivity of high-resolution 17O (I = 5/2) NMR spectroscopy of solids has advanced significantly in recent years. Here, we show that excellent results are now obtainable from milligram quantities of 17O-enriched materials, thereby allowing the technique to be applied to silicate phases synthesized under very high pressures in a multiple-anvil apparatus. We report the first 17O NMR study of beta-Mg2SiO4 (9.6 mg of 35% 17O-enriched material, synthesized at p = 16 GPa and T = 1873 K), a dense phase believed to have a significant role in the Earth's mantle. Using STMAS at magnetic fields of B0 = 9.4 and 11.7 T and MQMAS at B0 = 18.8 T, we have resolved and assigned all four crystallographically distinct O sites and determined their chemical shift and quadrupolar parameters.     

Molecular composites in thermosets

The synthesis of molecular composites where rigid polymer molecules are reinforcing elements in a thermoset bisimide matrix has been investigated. The approach has been designed to avoid phase separation by selecting systems where reaction of amine-terminated rigid and semiflexible oligomers with maleimide unsaturation occurs prior to crosslinking of the thermoset. This objective has been met for some compositions. The concentration and molecular weight of the rigid oligomers have been varied. The structure of the reinforcing polymer, the reactivity of the maleimide and the conditions for composite synthesis are variables of critical importance, and further work must determine the promise and limitations of this approach.     

Conformational analysis—II: A lanthanide-induced shift nmr and theoretical study of bicyclo [3.1.0] hexan-3-one and adamantanone

The solution conformation of bicyclo [3.1.0] hexan-3-one has been obtained by the use of the shifts induced in its ¹H and ¹³C NMR by Yb(fod)₃. Refinement of the angle of pucker, ∝, of the 5-membered ring indicates that the molecule adopts a flattened boat conformation with ∝ = 195°. This prediction is supported by ab initio, STO-3G, calculations on the isolated molecule. Use of a two- or four-site model for lanthanide-substrate complexing adequately reproduces the experimental data whereas a one-site binding model is unsatisfactory. The importance of multi-site binding is further emphasised by results for the C_2v-symmetric ketone, adamantanone, where only a four-site model gives satisfactory agreement between observed and calculated lanthanide-induced shifts.     

The formation of propylene fumarate oligomers for use in bioerodible bone cement composites

Poly(propylene fumarate) (PPF) oligomers were synthesized by step polymerization using bis(2-hydroxypropyl fumarate) or propylene bis(hydrogen maleate) as starting materials. Oligomers possessing identical degrees of polymerization (DP), but varying in their end group character (either hydroxyl or carboxyl) were first prepared and characterized, then used as part of a bone cement preparation consisting of oligomer, tricalcium phosphate, calcium carbonate, and methyl methacrylate. Compressive strength of the resulting composite appeared to be dependent on both the degree of polymerization of the PPF, and the nature of the oligomers' end groups.     

Micromethod for the analysis of the four commonly used antiepileptic drugs using a jet tube column

We describe a simple method for the determination of phenobarbital, diphenylhydantoin, carbamazepine, and primidone in serum, by use of gas-liquid chromatography with temperature programming. The methylated derivatives of these anticonvulsants were well resolved, as was 5-(p-methylphenyl)-5-phenylhydantoin, the internal standard. In this procedure we used the jet tube for separation of the drug from the serum. The proposed procedure requires only 100 μl of serum and can be done in less than 30 min. The lower limit of detection for each of the drugs is 0.5 mg/liter. Analytical recoveries of drug from serum were excellent and peak height and concentration were linearly related up to twice the toxic concentration for serum.
