The peak intensities obtained when 2,5-dihydroxybenzoic acid (DHB) was used as a 'classic' matrix were measured using substance P (SP) and betacyclodextrin (BCD) as analytes. Enhancements in peak intensities were observed going from 1:1 MeOH/H2O to dimethylforamide (DMF) as matrix solvents. Also non-covalent interactions between SP and solvent and DHB were observed suggesting close interactions between matrix, solvent and analyte in the gas-phase. Peak enhancements were previously reported with 'superDHB' (DHB and 2-hydroxy-5-methoxybenzoic acid at 10% v/v). Co-addition of structural analogues and their respective absorption coefficients were determined. It was found that other analogues used as co-matrices could give analyte peak enhancement similar to reported for sDHB with the additional benefit that some analogues could act as matrices with DHB addition. No direct correlation was observed between absorption coefficient and the ability of the molecule to act as a 'good' UV MALDI matrix.  相似文献

Production of high performance conductive textile yarn fibers for different electronic applications has become a prominent area of many research groups throughout the world. We have used oxidative chemical vapor deposition (OCVD) technique to coat flexible and high strength polyester yarns with conjugated polymer, poly(3,4‐ethylenedioxythiophene) (PEDOT) in presence of ferric (III) chloride (FeCl3) oxidant. OCVD is an efficient solvent free technique used to get uniform, thin, and highly conductive polymer layers on different substrates. In this paper, PEDOT‐coated polyester (PET) yarns were prepared under specific reaction conditions, and the electrical, mechanical and thermal properties were compared to previously studied PEDOT‐coated viscose yarns. Scanning electron microscopy (SEM) and FT‐IR analysis revealed that polymerization of PEDOT on the surface of the polyester yarns has been taken place successfully and structural analysis showed that PEDOT has strong interactions with viscose yarns as compared to PET yarns. The voltage–current (VI) characteristics showed that PET yarns are more conductive than PEDOT‐coated viscose yarns. The variation in the conductivity of PEDOT‐coated yarns and the heat generation properties during the flow of current through coated yarns for longer period of time, was studied by time–current (tI) characteristics. Thermogravimeteric analysis (TGA) was employed to investigate the thermal properties and the amount of PEDOT in PEDOT‐coated PET yarns compared to PEDOT‐coated viscose. The effect of PEDOT coating and ferric (III) chloride concentration on the mechanical properties of coated yarns was evaluated by tensile testing. A new class of unsaturated polyesters based on diethylketone have been prepared by interfacial polymerization of 2,4-bis(4-hydroxybenzylidene)-3-pentanone(I) and 2,4-bis(4-hydroxy-3-methoxybenzylidene)-3-pentanone(II) with 4,4’-azodibenzoyl chloride and 3,3’-azodibenzoyl chloride at ambient temperature. The model compounds were synthesized by reaction of(I) and(II) with benzoyl chloride. The new monomers, model compounds and polyesters have been characterized by different spectral analyses. The polyesters have inherent viscosity of 0.55-0.80 d L/g and moderate number average molecular weight(Mn) in the range of 6150-7400 g/mol. Most of the compounds exhibited their solubility in aprotic solvents while partial solubility in various halogenated organic solvents was observed. The temperatures of 10% weight loss were high(225-330 °C) in nitrogen, indicating that these polyesters have excellent thermal stability. Doping with iodine dramatically raised the conductivity and produced brown colored semiconductive polymers with a maximum conductivity of 2.7 × 10-6 --1cm-1. Three new pseudoguaiane-type sesquiterpenes, dichrocepholides A-C, and two new pseudoguaiane-type sesquiterpene dimers, dichrocepholides D and E, were isolated from the aerial part of Dichrocephala integrifolia. Their stereostructures were determined on the basis of chemical and physicochemical evidence. In addition, the extract and its principal sesquiterpene constituent, parthenin, showed an inhibitory activity on nitric oxide (NO) production and on induction of inducible NO synthase.  相似文献

The suitability of three different retention models to predict the retention times of poly(ethylene glycol)s (PEGs) in gradient and isocratic chromatography was investigated. The models investigated were the linear (LSSM) and the quadratic solvent strength model (QSSM). In addition, a model describing the retention behaviour of polymers was extended to account for gradient elution (PM). It was found that all models are suited to properly predict gradient retention volumes provided the extraction of the analyte specific parameters is performed from gradient experiments as well. The LSSM and QSSM on principle cannot describe retention behaviour under critical or SEC conditions. Since the PM is designed to cover all three modes of polymer chromatography, it is therefore superior to the other models. However, the determination of the analyte specific parameters, which are needed to calibrate the retention behaviour, strongly depend on the suitable selection of initial experiments. A useful strategy for a purposeful selection of these calibration experiments is proposed.  相似文献

Fundamental research has been carried out to define optimal "green" synthesis conditions for the production of titania (TiO(2)) and silver (Ag) nanocomposites (TANCs) ranging from 12.7-22.8 nm in diameter. A bottom-up colloidal approach was employed to accurately control TANC monodispersity and composition. TANCs were found to be effective at inactivating Escherichia coli (E. coli) in water. The presence of Ag in the nanocomposites induced a decrease in TiO(2) band gap energy, which favoured valence to conduction band electron transfer and allowed for electron excitation using visible light. Aggregation of ultra-fine particles was prevented through the use of a long-chain polymer as evidenced by electrophoretic mobility studies. The TANCs catalyzed oxidation of bacterial membranes and cell death or disinfection. Theoretically, the TANC mode of E. coli disinfection is via water photolysis, which results in production of hydroxyl radicals and hydrogen peroxide. These interact with the outer membrane polysaccharides and lipids, leading to lipid peroxidation, membrane weakening and resulted in cell death. Our overarching goals were to optimize the variables involved in TANC "green" synthesis and to characterize its nanostructure. High resolution (HR) transmission and scanning electron microscopic (TEM and SEM) studies demonstrated that TANCs were highly crystalline and mono-dispersive. Elemental composition of Ag and Ti, as measured by X-ray energy dispersive (EDS) and X-ray photoelectron spectroscopy (XPS) confirmed sample purity. Ultraviolet-visible (UV-VIS) spectroscopy showed that the energy band-gap of Ag modified TiO(2) was in the visible range.  相似文献

Cell-based microfluidic devices have attracted interest for a wide range of applications. While optical cell counting and flow cytometry-type devices have been reported extensively, sensitive and efficient non-optical methods to detect and quantify cells attached over large surface areas within microdevices are generally lacking. We describe an electrical method for counting cells based on the measurement of changes in conductivity of the surrounding medium due to ions released from surface-immobilized cells within a microfluidic channel. Immobilized cells are lysed using a low conductivity, hypotonic media and the resulting change in impedance is measured using surface patterned electrodes to detect and quantify the number of cells. We found that the bulk solution conductance increases linearly with the number of isolated cells contributing to solution ion concentration. The method of cell lysate impedance spectroscopy is sensitive enough to detect 20 cells microL(-1), and offers a simple and efficient method for detecting and enumerating cells within microfluidic devices for many applications including measurement of CD4 cell counts in HIV patients in resource-limited settings. To our knowledge, this is the most sensitive approach using non-optical setups to enumerate immobilized cells. The microfluidic device, capable of isolating specific cell types from a complex bio-fluidic and quantifying cell number, can serve as a single use cartridge for a hand-held instrument to provide simple, fast and affordable cell counting in point-of-care settings.  相似文献

Calmodulin is an EF hand calcium binding protein. Its binding affinities to various protein/peptide targets often depend on the conformational changes induced by the binding of calcium. One such target is melittin, which binds tightly to calmodulin in the presence of calcium, and inhibits its function. Chemical cross-linking combined with Fourier transform ion cyclotron resonance mass spectrometry has been employed to investigate the coordination of calmodulin and melittin in the complex at different concentrations of calcium. This methodology can be used to monitor structural changes of proteins induced by ligand binding, and study the effects these changes have on non- covalent interactions between proteins. Cross-linking results indicate that the binding place of the first melittin in the calcium free calmodulin form is the same as in the calcium loaded calmodulin/melittin complex.  相似文献
