Formic acid oxidatively cleaves 1,2,3,4-tetraaryl-2-butene-1,4-diones (1,2-aroylstilbenes) to 1,2-diaryl-1,2-ethanediones (benzils) under microwave irradiation. Mechanistic probing revealed that formic acid incorporates one of its oxygen atoms into benzil.  相似文献

The cycloaddition of α,β-epoxyaldehydes or ketones (2) with the ketene acetal MeHC=C(OMe)2 (1) gives epoxyoxetanes (3) in high yields. Without isolation they can be transformed into 4-hydroxy-5-(1-hydroxyalkyl)-γ-butyrolactones (6) via the epoxy esters 4 and trihydroxy esters (5). The lactones 6 appear to be valuable precursors for the synthesis of 5-(1-hydroxyalkyl)-3-methyl-2-5H-furanones (7) and 3-methyl-5-ylidene-2-5H-furanones (8)  相似文献

Macrophage infiltration into tumors has been correlated with poor clinical outcome in multiple cancer types. Therefore, tools to image tumor-associated macrophages could be valuable for diagnosis and prognosis of cancer. Herein, we describe the synthesis and characterization of a cathepsin S-directed, quenched activity-based probe (qABP), BMV083. This probe makes use of an optimized nonpeptidic scaffold leading to enhanced in vivo properties relative to previously reported peptide-based probes. In a syngeneic breast cancer model, BMV083 provides high tumor-specific fluorescence that can be visualized using noninvasive optical imaging methods. Furthermore, analysis of probe-labeled cells demonstrates that the probe primarily targets macrophages with an M2 phenotype. Thus, BMV083 is a potential valuable in vivo reporter for tumor-associated macrophages that could greatly facilitate the future studies of macrophage function in the process of tumorigenesis.  相似文献

A seven-step solid-phase synthesis of spirohydantoins and an eight-step solid-phase synthesis of spiro-2,5-diketopiperazines is reported. Key intermediate in the synthesis of both compound libraries is the resin-bound cyclic alpha,alpha-disubstituted alpha-amino ester, which can be obtained after selective homogeneous reduction of the aliphatic nitro ester using tin(II) chloride dihydrate. Nitro ester, in turn, is synthesized by a high-pressure-assisted [4 + 2] cycloaddition of resin-bound nitro alkene and butadiene, whereas nitro alkene is obtained by a Knoevenagel condensation of resin-bound nitro acetate with an imine. Novel spirohydantoins are obtained by isocyanate coupling with the resin-bound amino ester 5, followed by cyclization cleavage using a base. Novel spiro-2,5-diketopiperazines are obtained by PyBOP coupling of a Fmoc-protected amino acid with resin-bound amino ester, followed by Fmoc deprotection and an acid-assisted cyclization cleavage. After preparation of seven different resin-bound alpha,alpha-disubstituted alpha-amino esters, a 7 x 8 compound library of spirohydantoins was synthesized using eight different isocyanates, and a 7 x 8 compound library of spiro-2,5-diketopiperazines was synthesized using eight different Fmoc amino acids.  相似文献

We report on first measurements with polarized electrons stored in a medium-energy ring and with a polarized internal target. Polarized electrons were injected at 442 MeV (653 MeV), and a partial (full) Siberian snake was employed to preserve the polarization. Longitudinal polarization at the interaction point and polarization lifetime of the stored electrons were determined with laser backscattering. Spin observables were measured for electrodisintegration of polarized 3He, with simultaneous detection of scattered electrons, protons, neutrons, deuterons, and 3He nuclei, over a large phase space.  相似文献

Analytical methods by reversed-phase high-performance liquid chromatography (HPLC) and ultraviolet–visible spectrophotometry were developed and validated to determine doxorubicin in pH-sensitive chitosan nanoparticles. Chromatographic separation was performed on a reversed-phase C18 column, with ultraviolet detection at 254?nm and a mobile phase composed by 90% (v/v) acetonitrile in water and water pH 3.0 (33:67, v/v). The spectrophotometry method had the wavelength set at 480?nm and pH 3.0 water was used as a diluent. Calibration curves were linear from 1 to 30?µg/?mL (r?>?0.9995) and the specificity was demonstrated by verifying the absence of interferences from nanoparticle components. The values of accuracy and precision were within the acceptable limits, and robustness studies were performed by a two-level full factorial design. The validated methods were further tested to assess doxorubicin content in six different batches of pH-sensitive chitosan nanoparticles. The comparative analyses showed nonsignificant differences (p?>?0.05). Likewise, the HPLC method was successfully applied to determine the drug encapsulation efficiency as well as to measure doxorubicin during in vitro release assays and degradation kinetic studies under ultraviolet light C irradiation. Both methods fulfilled all validation parameters and were shown to be suitable for the characterization of doxorubicin-loaded pH-sensitive chitosan nanoparticles, without interferences from nanoparticle matrix.  相似文献

The controlled decomposition of Pt2(dba)3 (dba = dibenzylideneacetone) dispersed in 1-n-butyl-3-methylimidazolium tetrafluoroborate (BMI.BF4) and hexafluorophosphate (BMI.PF6) ionic liquids in the presence of cyclohexene by molecular hydrogen produces Pt0 nanoparticles. The formation of these nanoparticles follows the two-step [A --> B, A + B --> 2B (k1, k2)] autocatalytic mechanism. The catalytic activity in the hydrogenation of cyclohexene is influenced by the nature of the anion rather than the mean-diameter of the nanoparticles. Thus, higher catalytic activity was obtained with Pt0 dispersed in BMI.BF4 containing the less coordinating anion although these nanoparticles possess a larger mean diameter (3.4 nm) than those obtained in BMI.PF6 (2.3 nm). Similar mean diameter values were estimated from in situ XRD and SAXS. XPS analyses clearly show the interactions of the ionic liquid with the metal surface demonstrating the formation of an ionic liquid protective layer surrounding the platinum nanoparticles. SAXS analysis indicated the formation of a semi-organized ionic liquid layer surrounding the metal particles with an extended molecular length of around 2.8 nm in BMI.BF4 and 3.3 nm in BMI.PF6.  相似文献
