Basic evaluation of highly sensitive thyroid stimulating hormone immunoradiometric assay kits

Usefulness of three kinds of TSH kits by immunoradiometric assay (IRMA) was evaluated. They were able to measure low levels (less than 0.1 microIU/ml) in serum thyroid stimulating hormone (TSH) with incubation of short time (4 hours). In particular, RIABEAD II kit had a highly specific affinity for TSH and the normal range (+/- 2 S.D.) using it showed from 0.20 to 3.50 microIU/ml in 150 normal subjects. In patients with hyperthyroidism and in patients with hypothyroidism, the values of TSH were lower and higher than those of normal subjects, respectively. Another kits showed similar results. These results indicate that these TSH-IRMA kits are useful to evaluate serum TSH levels exactly.     

Solid-state optical properties of a chiral supramolecular fluorophore consisting of chiral (1R,2R)-1,2-diphenylethylenediamine and fluorescent carboxylic acid derivatives

By using (1R,2R)-1,2-diphenylethylenediamine as a chiral molecule and 2-anthracenecarboxylic acid as a fluorescent molecule, we created a chiral supramolecular organic fluorophore having circularly polarized luminescence properties in the solid-state.     

Preparation of phosphorus-containing polymers—XVI. Synthesis of phosphorus-containing polyanhydride-imides

Phosphorus-containing polyanhydride-imides were synthesized from N,N-bis(4-carboxyphthalimido)-3,3′-diphenylalkylphosphine oxide, 3,3′-[N,N′-bis(4-carboxyphthalimido)]benzophenone and their mixtures in two steps via the diacetyl derivatives of the bisimide-carboxylic acids. The resulting polymers have reduced viscosities of 0.06–0.14 dl/g and are soluble in polar aprotic solvents such as DMA, DMF and DMSO, and conc. H₂SO₄ etc. They have good hydrolytic stability for moisture and water. Phosphorus-containing polymers have little heat-resistance and poor flame-retardance. The benefit of incorporating phosphorus in the polymers is small.     

13C nuclear magnetic resonance studies of cellulose acetate

¹³C-NMR spectra of ring carbons and O-acetyl carbonyl carbons of cellulose acetate (CA) in dimethyl sulfoxide-d₆ were analyzed. The CA samples with the degree of substitution (DS) ranging from 0.84 and 1.91 were prepared by homogeneous acetylation of cellulose with acetic anhydride in a 10% LiCl/dimethyl acetamide solvent. It was found that the use of these low DS samples permitted easier assignments not only of the ring carbon but also of the O-acetyl carbonyl carbon signals. The assignments were confirmed by comparing with the ¹H-NMR spectra of the samples obtained by complete acetylation of the corresponding CA samples with acetyl-d₃ chloride. Two methods for determining the distribution of O-acetyl groups of CA, i.e., the relative DS at the three different types of hydroxyl groups, were developed. One is based on the measurements of the relative intensities of the signals for the ring carbons and the other is based on the measurements of the relative intensities of the signals for the O-acetyl carbonyl carbons.     

Medium-sized cyclophanes—XVII : Tetra-, hexa-, octa- and decahydropyrenes from [2.2]metacyclophane. Transannular dehydrogenation,isomerization, and disproportionation reactions by means of benzoyl peroxide,metal salts and sulfuric acids

[2.2]Metacyclophane (1) undergoes a variety of reactions according to the reagents and conditions. These include (1) substitution (path a), (2) transannular dehydrogenation (path b and c), (3) cycloisomerization (path d) and (4) transannular hydrogenation. A brief summary of these reactions is presented.The diversity of the reactions of 1 is further explored using benzoyl peroxide (BPO), cupric chloride, aluminum chloride, other metal salts, H₂SO₄ and FSO₃H. With BPO or cupric chloride, one-electron transfer mechanism is postulated. This involves a tautomeric ion pair formed by the intramolecular arylation with an aryl cation radical. A supporting evidence in favor of the mechanism is presented from experiments using various metal salts in different solvents.On the contrary, the reaction with aluminum chloride gives decahydropyrene (8) and octahydropyrene (11) together with cycloisomerization product 5 and dehydrogenation products 2, 3 and 4. When treated with AlCl₃HCl 1 gives similar products as above but the product ratios are quite different. The major product is 5 but only a trace amount of 8 is formed. The reaction with H₂SO₄ or FSO₃H also produce 2, 4, 5 and 11. Some mechanistic evidence in favor of the disproportionation reaction is presented.     

Analysis of transient membrane protein interactions by single-molecule diffusional mobility shift assay

Various repertoires of membrane protein interactions determine cellular responses to diverse environments around cells dynamically in space and time. Current assays, however, have limitations in unraveling these interactions in the physiological states in a living cell due to the lack of capability to probe the transient nature of these interactions on the crowded membrane. Here, we present a simple and robust assay that enables the investigation of transient protein interactions in living cells by using the single-molecule diffusional mobility shift assay (smDIMSA). Utilizing smDIMSA, we uncovered the interaction profile of EGFR with various membrane proteins and demonstrated the promiscuity of these interactions depending on the cancer cell line. The transient interaction profile obtained by smDIMSA will provide critical information to comprehend the crosstalk among various receptors on the plasma membrane.Subject terms: Fluorescence imaging, Super-resolution microscopy, Single-molecule biophysics     

Dipole moments of (styrene-p-chlorostyrene) triblock copolymers in solutions

(Styrene-p-chlorostyrene) triblock copolymers of the ABA and BAB types (A—polystyrene; B—poly-p-chlorostyrene) were prepared by anionic polymerization and their conformational behaviours in solutions were studied from measurements of the dipole moments. Two solvents, toluene and cumene, were used for the study; toluene is a good solvent for both polystyrene and poly-p-chlorostyrene whereas cumene is a selective solvent, good for polystyrene but poor for poly-p-chlorostyrene. It was found that the dipole moments of the block copolymers measured in toluene are the same for the ABA and BAB copolymers; in cumene however the dipole moment of the BAB copolymer is smaller than that of the ABA copolymer. The results give an additional support to our previous conclusion that the conformation of the block copolymers in a good solvent such as toluene could be approximated with a pseudo-random coil form; in a selective solvent, however, some anomalies take place in the conformation of the block copolymers, as deduced from intrinsic viscosity, osmotic pressure and light scattering measurements.     

Photoactivation of pheophorbide a induces a mitochondrial-mediated apoptosis in Jurkat leukaemia cells

The mechanism of cell death by pheophorbide a (Pba) which has been established to be a potential photosensitizer was examined in experimental photodynamic therapy (PDT) on Jurkat cells, a human lymphoid tumor cell line. In 30-60 min after irradiation, Pba treated cells exhibited apoptotic features including membrane blebbing and DNA fragmentation. Pba/PDT caused a rapid release of cytochrome c from mitochondria into the cytosol. Sequentially, activation of caspase-3 and the cleavage of poly ADP-ribose polymerase (PARP) were followed. Meanwhile, no evidence of activation of caspase-8 was indicated in the cells. In experiments with caspase inhibitors, it was found that caspase-3 alone was sufficient initiator for the Pba-induced apoptosis of the cells. Pba specific emission spectra were confirmed in the mitochondrial fraction and the light irradiation caused a rapid change in its membrane potential. Thus, mitochondria were entailed as the crucial targets for Pba as well as a responsible component for the cytochrome c release to initiate apoptotic pathways. Taken together, it was concluded that the mode of Jurkat cell death by Pba/PDT is an apoptosis, which is initiated by mitochondrial cytochrome c release and caspase-3-pathways.     

Synthesis of new azonia-helicenes with a quaternary nitrogen at the inner helix skeleton

The 16c-azonia[6]helicene salts, in which a carbon atom at the inner helix skeleton of [6]helicene is replaced by a quaternary nitrogen atom, have been synthesized by photo-induced intramolecular quater-nization of 2-[2-(1-chloronaphthyl)vinyl]benzo[h]quinolines in acetonitrile.     

Synthesis of aromatic polyamides by 1-hydroxybenzotriazole-catalyzed polycondensation of active aromatic diesters with aromatic diamines

Catalytic actions of various additives were studied in the polycondensation of di(4-nitrophenyl) isophthalate with bis(4-aminophenyl) ether in N-methyl-2-pyrrolidone, and 1-hydroxybenzotriazole (HOBt) was found to be a highly effective catalyst that yielded high-molecular-weight polyamide. In addition to the polycondensation of the 4-nitrophenyl ester, the polymerization of negatively substituted phenyl esters like di(2,4,5-trichlorophenyl) isophthalate was also accelerated by HOBt. For the HOBt-catalyzed aminolysis of esters a bifunctional concerted mechanism that involves an eight-membered transition state was proposed.
