Proteolytic enzymes play a central role in the biochemical mechanism of germination. The present study reported the presence of Zn2+-dependent endoproteases in the dry seeds of grass pea (Lathyrus sativus L.) with maximum caseinolytic activity observed at pH 8.0. Studies with class-specific inhibitors (specific for cysteine, serine, aspartate, and metalloproteases) on crude extract identified the inhibitory effect of 1,10-phenanthroline. This inhibitory effect was overcome by addition of Zn2+, not with Fe, Ca, Cu, Mg, or Co and indicates that the protease is Zn2+ dependent. This metalloprotease was further characterized by attempting gelatin-PAGE zymography and observed three distinct zones of proteolytic activity with higher mobility. The protease fraction consisted of three isoforms as evidenced by the appearance of three different bands on gelatin-PAGE zymogram. We also purified these proteases to 110-fold by a three-step procedure comprising crude extract from dry seeds, (NH4)2SO4 fractionation, and casein–alginate affinity chromatography. The molecular mass of isoforms of metalloproteases is 25, 18, and 14 kDa.  相似文献

Ternary copper(II) complexes [Cu(l-pro)(B)(H2O)](NO3) (1, 2) where l-pro = l-proline, B is a N,N-donor heterocyclic base, viz. 2,2′-bipyridine (bpy, 1), 1,10-phenanthroline (phen, 2), are synthesized, characterized, and their DNA binding and cleavage activity studied. The bpy complex (1) is structurally characterized by single-crystal X-ray crystallography. The complexes show the presence of a distorted square-pyramidal (4 + 1) CuN3O2 coordination geometry. Complex [Cu(l-pro)(bpy)(H2O)](NO3) (1) crystallizes in the triclinic space group P1 with unit cell parameters: a = 7.082(3) Å, b = 10.483(5) Å, c = 11.581(5) Å, α = 89.700(7)°, β = 83.488(8)°, γ = 84.109(8)° and V = 849.7(7) Å3. The one-electron paramagnetic complexes display a d–d band near 600 nm in water and show a cyclic voltammetric response due to Cu(II)/Cu(I) couple near 0.1 V (versus SCE) in Tris–HCl buffer–0.1 M KCl. Binding interactions of the complexes with calf thymus (CT) DNA have been investigated by emission, absorption, viscosity and DNA thermal denaturation studies. The phen complex displays significant binding propensity to the CT DNA giving an order: 2 (phen)  1 (bpy). The bpy complex does not show any apparent binding to the DNA and hence poor cleavage efficiency. Complex 2 shows efficient oxidative cleavage of SC-DNA in the presence of 3-mercaptopropionic acid (MPA) involving hydroxyl radical species as evidenced from the control data showing inhibition of DNA cleavage in the presence of DMSO and catalase.  相似文献

Steady-state fluorescence and time-resolved absorption measurements in pico- and femtosecond time domain have been used to investigate the dynamics of hydrogen bond in the excited singlet (S(1)) state of fluorenone in alcoholic solvents. A comparison of the features of the steady-state fluorescence spectra of fluorenone in various kinds of media demonstrates that two spectroscopically distinct forms of fluorenone in the S(1) state, namely the non-hydrogen-bonded (or free) molecule as well as the hydrogen-bonded complex, are responsible for the dual-fluorescence behavior of fluorenone in solutions of normal alcoholic solvents at room temperature (298 K). However, in 2,2,2-trifluoroethanol (TFE), a strong hydrogen bond donating solvent, emission from only the hydrogen-bonded complex is observed. Significant differences have also been observed in the temporal evolution of the absorption spectroscopic properties of the S(1) state of fluorenone in protic and aprotic solvents following photoexcitation using 400 nm laser pulses. An ultrafast component representing the solvent-induced vibrational energy relaxation (VER) process has been associated with the dynamics of the S(1) state of fluorenone in all kinds of solvents. However, in protic solvents, in addition to the VER process, further evolution of the spectroscopic and dynamical properties of the S(1) state have been observed because of repositioning of the hydrogen bonds around the carbonyl group. In normal alcohols, two different kinds of hydrogen-bonded complex of the fluorenone-alcohol system with different orientations of the hydrogen bond with respect to the carbonyl group and the molecular plane of fluorenone have been predicted. On the other hand, in TFE, formation of only one kind of hydrogen-bonded complex has been observed. These observations have been supported by theoretical calculations of the geometries of the hydrogen-bonded complexes in the ground and the excited states of fluorenone. Linear correlation between the lifetimes of the equilibration process occurring because of repositioning of the hydrogen bonds and Debye or longitudinal relaxation times of the normal alcoholic solvents establish the fact that, in weakly hydrogen bond donating solvents, the hydrogen bond dynamics can be described as merely a solvation process. Whereas, in TFE, hydrogen bond dynamics is better described by a process of conversion between two distinct excited states, namely, the non-hydrogen-bonded form and the hydrogen-bonded complex.  相似文献

A rapid l-proline catalyzed direct aldol reaction between various aldehydes and acetone was achieved using PEG as the solvent with comparable enantioselectivities and yields to those obtained in other solvents. A new synthetic strategy for the synthesis of novel 3-(3-(3-methyl-4-nitroisoxazol-5-yl)-2-phenyl-1-(5,7-diaryl-7H-thiazolo[3,2-a]pyrimidin-3-yl)propyl)-5,7-diaryl-7H-thiazolo[3,2-a] pyrimidines(7a-i) analogues is described.Reaction of 3-(2(3-methyl-4-nitroisoxazole-5-yl)-1-phenylethyl)pentane-2,4-dione(3) with two moles of thiourea in presence of iodine and CuO afforded 4-(1-(2-aminothiazol-4-yl)-3-(3-methyl-4-nitroisoxazol-5-yl)-2-aryl propyl-thiazol-2-amine(5).Compound 5 on reaction with two moles of chalcone(6) furnished novel 3-(3-(3-methyl-4-nitroisoxazol-5-yl)-2-phenyl-1-(5,7-diaryl-7H-thiazolo[3,2a]pyrimidin-3-yl)propyl)-5,7-diaryl-7H-thiazolo[3,2-a] pyrimidines(7a-i).  相似文献

The use of reversible covalent bonding in a four-component assembly incorporating chiral alcohols was recently reported to give a method for determining the enantiomeric excess of the alcohols via CD spectroscopy. Experiments that probe the mechanism of this assembly, which consists of 2-formylpyridine (2-PA), dipicolylamine (DPA), Zn(ii) and alcohols to yield zinc complexes of tren-like ligands, are presented. The studies focus upon the mechanism of conversion of a hemi-aminal (1) to a hemi-aminal ether (3), thereby incorporating the fourth component. It was found that molecular sieves along with 3 to 4 equivalents of alcohol are required to drive the conversion of 1 to 3. Attempts to isolate an intermediate in this reaction via addition of strong Lewis acids led to the discovery of a five-membered ring pyridinium salt (5), but upon exposure to Zn(ii) and alcohols gave different products to the assembly. This was interpreted to support the intermediacy of an iminium species. Kinetic studies reveal that the conversion of 1 to 3 is zero-order in alcohol in large excesses of alcohol, supporting rate-determining formation of an intermediate prior to reaction with alcohol. Further, the magnitudes of the rate constants for interconversion of 1 and 3 are similar, supporting the notion that there are similar rate-determining steps (rds) for the forward and reverse reactions. Hammett plots show that the rds involves creation of a negative charge (interpreted as the loss of positive charge), supporting the notion that the decomplexation of Zn(ii) from the assemblies to generate apo-forms of 1 and 3 is rate-determining. The individual mechanistic conclusions are combined to create a qualitative reaction coordinate diagram for the interconversion of 1 and 3.  相似文献

The low temperature microwave sintered NiCuZn and MgCuZn ferrites with compositions Ni0.35Cu0.05Zn0.60Fe2O4 and Mg0.35Cu0.05Zn0.60Fe2O4 were synthesized by conventional mixed oxide method. NiCuZn and MgCuZn ferrite samples obtained showed better sintered densities at 950 and 900 °C, respectively. The scanning electron micrographs of both the ferrite samples appear to be very much similar. The magnitude of initial permeability of MgCuZn ferrite samples is found to be obviously higher than those of NiCuZn ferrite samples at all sintering temperatures. This is mainly due to the fact that MgCuZn ferrite has smaller magnetocrystalline anisotropy constant and magnetostrictive constant. NiCuZn ferrites have higher saturation magnetization than MgCuZn ferrites, which is due to the higher magnetic moment of NiCuZn ferrites. Our results indicate that the microwave sintering method seems to be a potential technique in the MLCI technology.  相似文献

A simple, sensitive and rapid high-performance liquid chromatography/negative ion electrospray tandem mass spectrometry method was developed and validated for the assay of fluvastatin in human plasma. Following solid-phase extraction, the analytes were separated using an isocratic mobile phase on a reversed-phase column and analyzed by mass spectrometry in the multiple reaction monitoring mode using the respective [M-H]- ions, m/z 410/348 for fluvastatin and m/z 480/418 for the internal standard. The assay exhibited a linear dynamic range of 2-500 ng/mL for fluvastatin in human plasma. The lower limit of quantification was 2 ng/mL with a relative standard deviation of less than 5%. Acceptable precision and accuracy were obtained for concentrations over the standard curve range. A run time of 1.5 min for each sample made it possible to analyze more than 400 human plasma samples per day. The validated method has been successfully used to analyze human plasma samples for application in pharmacokinetic, bioavailability or bioequivalence studies.  相似文献
