Echogenic liposomes (ELIP) encapsulate gas bubbles and drugs within lipid vesicles, but the mechanisms of ultrasound-mediated drug release from ELIP are not well understood. The effect of cavitation activity on drug release from ELIP was investigated in flowing solutions using two fluorescent molecules: a lipophilic drug (rosiglitazone) and a hydrophilic drug substitute (calcein). ELIP samples were exposed to pulsed Doppler ultrasound from a clinical diagnostic ultrasound scanner at pressures above and below the inertial and stable cavitation thresholds. Control samples were exposed to a surfactant, Triton X-100 (positive control), or to flow alone (negative control). Fluorescence techniques were used to detect release. Encapsulated microbubbles reduced the measured fluorescence intensity and this effect should be considered when assessing drug release from ELIP. The origin of this effect is not specific to ELIP. Release of rosiglitazone or calcein compared to the negative control was only observed with detergent treatment, but not with ultrasound exposure, despite the presence of stable and inertial cavitation activity. Release of rosiglitazone or calcein from ELIP exposed to diagnostic ultrasound was not observed, even in the presence of cavitation activity. Ultrasound-mediated drug delivery strategies with ELIP will thus rely on passage of the drug-loaded liposomes to target tissues.  相似文献

Green synthesized silver nanoparticles (AgNPs) have enormous applications. Hence, there is an increasing demand to explore diverse bioresources for AgNP fabrication to make the process more cost‐effective and rapid as possible. Due to the abundantly present hydroxyl groups of rice starch, it provides ideal sites for metal ion complexation and thereby synthesis of nanoparticles with promising activity. So the study was designed to develop rapid, eco‐friendly and cost‐effective method for green AgNP synthesis using boiled rice water starch in the presence of sunlight irradiation. The starch‐capped nanoparticles (sAgNPs) formed in the study were found to have the surface plasmon absorbance at 439 nm. The study showed optimum yield of sAgNPs when 25% rice starch was treated with aqueous 1 mM AgNO3 for 15 min in the presence of sunlight. Fourier transform infrared spectroscopy analysis provided mechanistic insight into the role of –OH groups of starch in the reduction of AgNO3 to sAgNPs. On further characterization by X‐ray diffraction analysis, the sAgNPs were identified to have FCC crystal structure. At the same time, high‐resolution transmission electron microscopic analysis showed majority of sAgNPs to have spherical morphology, and dynamic light scattering study revealed the average particle size as 36.3 nm. Further confirmation on presence of AgNPs was carried out by energy‐dispersive X‐ray spectroscopy. Moreover, the sAgNPs exhibited promising antibacterial activity against foodborne pathogens, Salmonella Typhimurium and Staphylococcus aureus.  相似文献

The energy transfer mechanism in C 540–rhodamine 6G and C 540–rhodamine B donor–acceptor (D–A) pairs is investigated. The good spectral overlap and the proximity of D–A pair molecules, which is around 60 Å, denote the possibility of resonance energy transfer between the D–A pairs. Energy transfer is studied in two solvent environments. C 540–Rh 6G and C 540–RhB D–A pairs behave in quite different manner in different solvent environments.  相似文献

Lempel-Ziv complexity (LZ) [J. Ziv, A. Lempel, On the complexity of finite sequences, IEEE Trans. Inform. Theory 22 (1976) 75-81] and its variants have been used widely to identify non-random patterns in biomedical signals obtained across distinct physiological states. Non-random signatures of the complexity measure can occur under nonlinear deterministic as well as non-deterministic settings. Surrogate data testing have also been encouraged in the past in conjunction with complexity estimates to make a finer distinction between various classes of processes. In this brief letter, we make two important observations (1) Non-Gaussian noise at the dynamical level can elude existing surrogate algorithms namely: Phase-randomized surrogates (FT) amplitude-adjusted Fourier transform (AAFT) and iterated amplitude-adjusted Fourier transform (IAAFT). Thus any inference nonlinear determinism as an explanation for the non-randomness is incomplete (2) Decrease in complexity can be observed even across two linear processes with identical auto-correlation functions. The results are illustrated with a second-order auto-regressive process with Gaussian and non-Gaussian innovations. AR(2) processes have been used widely to model several physiological phenomena, hence their choice. The results presented encourage cautious interpretation of non-random signatures in experimental signals using complexity measures.  相似文献

High fidelity polymerases are efficient catalysts of phosphodiester bond formation during DNA replication or repair. We interpret molecular dynamics simulations of a polymerase bound to its substrate DNA and incoming nucleotide using a quasiharmonic model to study the effect of external forces applied to the bound DNA on the kinetics of phosphoryl transfer. The origin of the force dependence is shown to be an intriguing coupling between slow, delocalized polymerase-DNA modes and fast catalytic site motions. Using noncognate DNA substrates we show that the force dependence is context specific.  相似文献

Salvia aethiopis is a perennial plant native to Eurasia and known by the common names Mediterranean sage or African sage. This plant has been used in Iranian medicine as a carminative and tonic. The ethanolic extract of aerial part of S. aethiopis exhibited moderate to weak activity towards delta and kappa opioid receptors (46.3 and 45.3% displacement, respectively). Chromatographic purification of the ethanolic extract on silica gel column led to isolation of one new: 3α-hydroxy-8α-acetoxy-13,14,15,16-tetranorlabdan-12-oic acid (I) and three known compounds: sesquiterpene spathulenol (II), β-sitosterol (III) and β-sitosterol-3-O-glucoside (IV). The structures of all isolated compounds were elucidated by their NMR (1D and 2D) and MS spectral data. All the fractions and isolated compounds were tested for cannabinoid and opioid receptor binding assays.  相似文献

Cichorium intybus L. (Asteraceae family) is a world-wide grown plant known as chicory. In traditional medicine, this plant is used as diuretic, anti-inflammatory, digestive, cardiotonic and liver tonic. Chromatographic purification of the supercritical fluid extract of aerial parts of C. intybus on silica gel column led to isolation of three compounds: new compound, 28β-hydroxytaraxasterol (I), and two known compounds usnic acid (II) and β-sitosterol (III). Purification of the ethanolic extract of aerial parts of this plant on silica gel column chromatography yielded four compounds: 1,3-dioleylglycerate (IV), sitoindoside II (V), 11β-13-dihydrolactucin (VI) and β-sitosterol-3-O-glucoside (VII). The structures of the isolated compounds were determined by their 1D, 2D NMR and MS spectral data. All the fractions and isolated compounds were tested for cannabinoid and opioid receptor binding, as well as antibacterial, antifungal and antimalarial activities. Compound I showed moderate activity (60.5% displacement) towards CB1 receptor.  相似文献

Titanium oxide thin films have been deposited by an ion plating technique under rf glow. The structure of the deposited film has been found to be amorphous and the composition has been analysed by IR spectrum. Aging and annealing studies of the titanium oxide capacitors have been made. The dielectric constant of the film at 1 kHz has been estimated to be 12.4. The dependence of the capacitance and dielectric loss on frequency and temperature have been studied, and the results are discussed. The temperature coefficient of the capacitance has also been calculated.  相似文献

A method for the removal of anionic (sulfonated) dyes from aqueous dye solutions using the chemical interaction of dye molecules with polyaniline is reported. Polyaniline (PANI) emeraldine salt was synthesized by chemical oxidation. Sulfonated dyes undergo chemical interactions with the charged backbone of PANI, leading to significant adsorption of the dyes. This phenomenon of selective adsorption of the dyes by PANI is reported for the first time and promises a green method for removal of sulfonated organics from wastewater. The experimental observations from UV-vis spectroscopy, X-ray diffraction, and conductivity measurements rule out the possibility of secondary doping of polyaniline salt by sulfonated dye molecules. A possible mechanism for the chemical interaction between the polymer and the sulfonated dye molecules is proposed. The kinetic parameters for the adsorption of sulfonated dyes on PANI are also reported.  相似文献
