The synthesis and structure-activity-relationship (SAR) for a series of N-substituted piperazinyl carbamoyl 7-15 and piperazinyl acetyl 18-26 derivatives of tetrahydropapaverine have been carried out. The general synthetic methods of carbamoyl tetrahydropapaverine analogues involve N-substituted piperazines and carbamoyl imidazole tetrahydropapaverine as starting materials. Another route for synthesizing these compounds, involving the formation of carbamoyl imidazole piperazine has also been explored. Acylation of tetrahydropapaverine followed by substitution with various piperazinyl moities afforded the acetyl tetrahydropapaverine derivatives. Variously substituted piperazines have been used to monitor the effect of electron releasing and electron withdrawing substituents upon the antispasmodic activity of the molecules. Effect of varying electron densities on the antispasmodic activity, by altering the position of these groups on the benzene ring has also been monitored. Pharmacological methods involve the in vitro antispasmodic activity studies on a freshly removed guinea pig ileum using a force displacement transducer amplifier connected to a physiograph. Among the analogues synthesized in the present study, a promising compound 7, a potent muscle relaxant as compared to papaverine has been obtained.  相似文献

Abstract

One of the highly emerging and an important aspect of organic chemistry is the metal catalyzed synthesis of heterocycles. The methods used earlier for the synthesis of heterocycles were significant in the organic synthesis and developing cost-effective, improved and facial methods were beneficial to construct the complex architectures. For the both stereoselective and regioselective synthesis of six-membered nitrogen containing heterocycles, cyclic reactions that are Ag-mediated have known to be very efficient. NMR spectroscopy has become a vital tool for studies of protein conformational changes and dynamics. Oxidized Fe(III)cytochromes c are a particularly attractive target for NMR analysis because their paramagnetism (S = (1)/(2)) leads to high (1)H chemical shift dispersion, even for unfolded or otherwise disordered states. In addition, analysis of shifts induced by the hyperfine interaction reveals details of the structure of the heme and its ligands for native and nonnative protein conformational states. The use of NMR spectroscopy to investigate the folding and dynamics of paramagnetic cytochromes c is reviewed here. Studies of nonnative conformations formed by denaturation and by anomalous in vivo maturation (heme attachment) are facilitated by the paramagnetic, low-spin nature of native and nonnative forms of cytochromes c. Investigation of the dynamics of folded cytochromes c also are aided by their paramagnetism. As an example of this analysis, the expression in Escherichia coli of cytochrome c(552) from Nitrosomonas europaea is reported here, along with analysis of its unusual heme hyperfine shifts. The results are suggestive of heme axial methionine fluxion in N. europaea ferricytochrome c(552). The application of NMR spectroscopy to investigate paramagnetic cytochrome c folding and dynamics has advanced our understanding of the structure and dynamics of both native and nonnative states of heme proteins.  相似文献

Thermal decomposition of ammonium tris (malonato) ferrate (III) tetrahydrate, i. e. (NH4)3[Fe(CH2C2O4)3]·4H2O has been studied up to 973 K in static air atmosphere employing Mössbauer and infrared spectroscopies, and non-isothermal techniques (TG, DTG, DTA). The anhydrous complex decomposes into an iron (II) intermediate at 453 K. The iron (II) species on further heating is reoxidized to -Fe2O3 as the final thermolysis product. An increase in particle size of -Fe2O3 with increasing decomposition temperature has been observed. The results are compared with the analogous oxalate complex.  相似文献

Thermal decomposition of some hydroxy iron(III) carboxylates, i.e., iron(III) lactate, Fe(CH3CHOHCOO)3, iron(III) tartrate, Fe2(C4H4O6)3 and iron(III) citrate, Fe(C6H5O7) · 5H2O has been studied in static air atmosphere in the temperature range 298–773 K employing Mössbauer, infrared spectroscopies and themogravimetric methods. The compounds directly decompose to -Fe2O3 without undergoing reduction to iron(II) intermediates. An increase in particle size of -Fe2O3 has been observed with increasing decomposition temperature. The thermal stability follows the sequence: iron(III) tartrate > iron(III)citrate > iron(III)lactate.  相似文献

The reaction of copper(I) iodide with tri-m-tolylphosphine (m-tolyl(3)P) in acetonitrile yielded the cluster [Cu(6)(mu2-I)(mu3-I)4(mu4-I)(m-tolyl(3)P)4(CH(3)CN)2] (1), with a bicapped adamantoid geometry. In this compound, four Cu atoms are coordinated to four terminally bonded m-tolyl(3)P ligands, two Cu atoms are bonded to two CH(3)CN ligands, and iodide ligands have mu2-I, mu3-I, and mu4-I bonding modes. This compound has four CuI(3)P and two CuI(3)N cores, and geometry around each Cu center is distorted tetrahedral.The polarizable iodide ligand and the position of the methyl group in the phenyl ring attached to the P atom appear to have played the pivotal role in the formation of monomeric bicapped adamantoid geometry, which is unique in copper chemistry.  相似文献

The use of a new electrospray qQq Fourier transform ion cyclotron mass spectrometer (qQq-FTICR MS) instrument for biologic applications is described. This qQq-FTICR mass spectrometer was designed for the study of post-translationally modified proteins and for top-down analysis of biologically relevant protein samples. The utility of the instrument for the analysis of phosphorylation, a common and important post-translational modification, was investigated. Phosphorylation was chosen as an example because it is ubiquitous and challenging to analyze. In addition, the use of the instrument for top-down sequencing of proteins was explored since this instrument offers particular advantages to this approach. Top-down sequencing was performed on different proteins, including commercially available proteins and biologically derived samples such as the human E2 ubiquitin conjugating enzyme, UbCH10. A good sequence tag was obtained for the human UbCH10, allowing the unambiguous identification of the protein. The instrument was built with a commercially produced front end: a focusing rf-only quadrupole (Q0), followed by a resolving quadrupole (Q1), and a LINAC quadrupole collision cell (Q2), in combination with an FTICR mass analyzer. It has utility in the analysis of samples found in substoichiometric concentrations, as ions can be isolated in the mass resolving Q1 and accumulated in Q2 before analysis in the ICR cell. The speed and efficacy of the Q2 cooling and fragmentation was demonstrated on an LCMS-compatible time scale, and detection limits for phosphopeptides in the 10 amol/muL range (pM) were demonstrated. The instrument was designed to make several fragmentation methods available, including nozzle-skimmer fragmentation, Q2 collisionally activated dissociation (Q2 CAD), multipole storage assisted dissociation (MSAD), electron capture dissociation (ECD), infrared multiphoton induced dissociation (IRMPD), and sustained off resonance irradiation (SORI) CAD, thus allowing a variety of MS(n) experiments. A particularly useful aspect of the system was the use of Q1 to isolate ions from complex mixtures with narrow windows of isolation less than 1 m/z. These features enable top-down protein analysis experiments as well structural characterization of minor components of complex mixtures.  相似文献

A sensitive and inexpensive method of spectrophotometric determination of chromium(VI), based on the absorbance of its complex with malachite green and acetic acid at pH 2.5 is reported. The complex shows a molar absorptivity of 8 x 10(4) l.mole(-1) cm(-1) at 560 nm, using malachite green and acetic acid as reference solution. The effect of time, temperature, pH and reagent concentration is studied and optimum operating conditions are established. Beer's law is applicable in the concentration range 2.0-22.8 mug/ml chromium(VI). The resin beads act as a catalyst and as little as 1.6 mug of chromium(VI) is detected in the resin phase as compared to 4.1 mug in the solution phase. The standard deviation in the determinations is +/-0.40 mug/ml for a 10.35 mug/ml solution.  相似文献
