Nonlinear harmonic radiation was observed using the VISA self-amplified, spontaneous emission (SASE) free-electron laser (FEL) at saturation. The gain lengths, spectra, and energies of the three lowest SASE FEL modes were experimentally characterized. The measured nonlinear harmonic gain lengths and center spectral wavelengths decrease with harmonic number, n, which is consistent with nonlinear harmonic theory. Both the second and third nonlinear harmonics energies are about 1% of the fundamental energy. These experimental results demonstrate for the first time the feasibility of using nonlinear harmonic SASE FEL radiation to produce coherent, femtosecond x rays.  相似文献

Observation of ultrawide bandwidth, up to 15% full-width, high-gain operation of a self-amplified spontaneous emission free-election laser (SASE FEL) is reported. This type of lasing is obtained with a strongly chirped beam (deltaE/E approximately 1.7%) emitted from the accelerator. Because of nonlinear pulse compression during transport, a short, high current bunch with strong mismatch errors is injected into the undulator, giving high FEL gain. Start-to-end simulations reproduce key features of the measurements and provide insight into mechanisms, such as angular spread in emitted photon and electron trajectory distributions, which yield novel features in the radiation spectrum.  相似文献

A laser-based double beam absorption detection system for aggregation immunoassays has been developed. The assay was based on the aggregation of gold nanoparticles that are coated with protein antigens in the presence of their corresponding antibodies. The aggregation of the gold nanoparticles results in an absorption change that is monitored at 635 nm using the double beam spectrometer. The noise level of the spectrometer is 1x10(-6)arbitrary units. This corresponds to a tenfold improvement in comparison to commercial absorption detectors and is comparable with previously reported more complicated laser-based absorption spectrometers. The dye Nile-Blue-A was used to test the analytical performance of the system. A limit of detection of 3x10(-8 )M Nile-Blue-A was observed. The relative standard deviation between consecutive measurements was lower than 1.5%. The system is suitable for field applications of aggregation-based immunoassays.  相似文献

Particulate methane monooxygenase (pMMO) is one of the few enzymes that can activate methane. The metal content of this enzyme has been highly controversial, with suggestions of a dinuclear Fe site or mono‐, di‐, or trinuclear Cu sites. Crystal structures have shown a mono‐ or dinuclear Cu site, but the resolution was low and the geometry of the dinuclear site unusual. We have employed quantum refinement (crystallographic refinement enhanced with quantum‐mechanical calculations) to improve the structure of the active site. We compared a number of different mono‐ and dinuclear geometries, in some cases enhanced with more protein ligands or one or two water molecules, to determine which structure fits two sets of crystallographic raw data best. In all cases, the best results were obtained with mononuclear Cu sites, occasionally with an extra water molecule. Thus, we conclude that there is no crystallographic support for a dinuclear Cu site in pMMO.  相似文献

While RF photoinjectors are an excellent source of high brightness electron beams, there are constraints to tying together the expected emittance and peak current performance of a given photoinjector system. These constraints, which arise from the complicated dynamics of the electrons due to the interplay of RF and space-charge forces within the photoinjector, tend to favor lower peak current operation. For some ultimate uses of photoinjector beams, such as linear collider test beams, wakefield accelerators, and free-electron lasers (FEL's), one may desire much higher peak currents. In this case, an inexpensive and reliable method for producing extremely short high-current electron bunches is to use magnetic compression. We examine this scheme analytically and by computer simulation. Many applications are illustrated, including the TESLA Test Facility/FEL injector, ultra-high current beams for plasma wakefields and generation of femtosecond electron pulses for injection into short wavelength laser-based accelerators. It is shown that the injection timing jitter associated with the laser can be nearly eliminated using this scheme, making it an indispensable component in many of the advanced accelerator injectors we consider  相似文献
