Process optimisation for the biosynthesis of cellulase by Bacillus PC-BC6 and its mutant derivative Bacillus N3 using submerged fermentation

This study deals with optimisation of cultural conditions for enhanced production of cellulase by Bacillus PC-BC6 and its mutant derivative Bacillus N3. Influence of different variables including incubation time, temperature, inoculum size, pH, nitrogen sources and metal ions has been studied. The optimum conditions for cellulase production were incubation period of 72 h, inoculum size 4% incubation temperature 37°C, pH 7, 0.25% ammonium sulphate, 0.2% peptone as inorganic and organic nitrogen source in case of Bacillus PC-BC6. In case of mutant Bacillus N3, optimal conditions were incubation period of 48 h, incubation temperature 37°C, inoculum size 3%, pH 7, 0.2% ammonium chloride and 0.15% yeast extract. Presence of MnSO₄ and CaCl₂ enhances the enzyme production by Bacillus PC-BC6 and mutant Bacillus N3, respectively. This study was innovative and successful in producing cellulase economically by using cheap indigenous substrate Saccharum spontaneum.     

Polymerization of SU-8 under irradiation stimulated by temperature increase

X-ray irradiation of thick polymer layers (a deep X-ray lithography procedure) results in a temperature distribution over the exposed area. The induced inhomogeneous temperature increase can reach several tens of degrees and it affects direct SU-8 polymerization depending on the microstructure size and area.     

Ion population fraction calculations using improved screened hydrogenic model with l-splitting

Ion population fraction(IPF) calculations are very important to understand the radiative spectrum emitted from the hot dense matter. IPF calculations require detailed knowledge of all the ions and correlation interactions between the electrons of an ion which are present in a plasma environment. The average atom models, e.g., screened hydrogenic model with l-splitting(SHML), now have the capabilities for such calculations and are becoming more popular for in line plasma calculations. In our previous work [Ali A, Shabbir Naz G, Shahzad M S, Kouser R, Rehman A and Nasim M H 2018 High Energy Density Phys. 26 48], we have improved the continuum lowering model and included the exchange and correlation effects in SHML. This study presents the calculation of IPF using classical theory of fluctuation for our improved screened hydrogenic model with l-splitting(I-SHML) under local thermodynamic equilibrium conditions for iron and aluminum plasma over a wide range of densities and temperatures. We have compared our results with other models and have found a very good agreement among them.     

Design,synthesis and X‐ray structural studies of novel [acetonitrile‐benzyl‐3‐N‐(2, 4 dihydroxyphenylmethylene) hydrazinecarbodithioato‐κ3‐N′, S,O] nickel(ll) complex that potently inhibit cell proliferation through regulation of apoptosis related genes

Nickel is a fundamental element for healthy life for human and higher animals. For biological importance, its complexation with bioactive ligand is worth to be studied with the aim to understand its function. Using mouse peritoneal cancer model, MTT colorimetric assay and anticancer activity analysis, we examined the role of nickel(ll) complex in growth inhibition of cancer cells. A novel nickel(ll) complex was synthesized and characterized using physico‐chemical and spectroscopic techniques. The study indicated that both the ligand and complex were capable of inhibiting Ehrlich Ascites Carcinoma (EAC) cells growth by 28.21% and 44.52%, respectively, when administered 0.3 mg/kg/day body weight intraperitoneally for five consecutive days in Swiss Webstar mice. Determination the LD₅₀ of the complex (55 mg/kg) allowed adjusting the dose as 2.75 mg/kg and upon administration, inhibition increased to 69.36%. The ligand and complex have shown an inhibitory effect in the range of 4.86%–67.3% and 6.1%‐ 89.37%, respectively, against EAC cells (concentration range of 31.25–500 μg/ml) in RPMI‐1640 medium as determined by MTT colorimetric assay. Apoptotic cell morphological alteration was determined through optical and fluorescence microscopy. Up regulation of P⁵³, Bax, Cas‐8, Cas‐3 and Fas and down regulation of NF‐kB and Bcl‐2 gene expression were observed in the cells treated with the nickel(ll) complex for five consecutive days. In conclusion, the newly synthesized nickel(ll) complex has shown anti‐proliferative activity and can further be optimized to be used as a lead molecule for anticancer drug.

One-Pot Hydrothermal Synthesis of Ternary 1T-MoS2/Hexa-WO3/Graphene Composites for High-Performance Supercapacitors

A new ternary composite of 1T-molybdenum disulfide, hexagonal tungsten trioxide, and reduced graphene oxide (M-W-rGO) is synthesized by using a one-pot hydrothermal process. The synergetic effect of 1T-MoS₂ and hexa-WO₃ nanoflowers improves the electrochemical performance for supercapacitors by inducing additional active sites and hexagonal tunnels, respectively, which lead to high storage capacity and easy transfer of electrolyte ions. The ternary M-W-rGO composite has a high specific capacitance of 836 F g⁻¹ at 1 A g⁻¹, which is nearly twice that of binary composites of M-rGO and W-rGO with high capacitance retention of 86.35 % after 3000 cycles at a high current density of 5 A g⁻¹. This study provides a new ternary composite that can be used as an electrode material for high-performance supercapacitors.

Validated Method for the Simultaneous Determination of Gemifloxacin and H2‐receptor Antagonists in Bulk,Pharmaceutical Formulations and Human Serum by RP‐HPLC; In‐vitro Applications

Simple, isocratic and rapid RP‐HPLC method has been developed for the simultaneous analysis of gemifloxacin and H₂‐receptor antagonists i.e. Cimetidine, Famotidine and Ranitidine, in bulk, pharmaceutical formulation and human serum. Separation was achieved on the RP‐Mediterranea column [C18 (250 × 4.6 mm, 5 μ)] at ambient temperature using mobile phase consisting of acetonitrile: methanol: water (20:28:52 v/v/v pH 2.8 adjusted by phosphoric acid). Flow rate was 1.0 mL/min with an average operating pressure of 180 kg/cm². Gatifloxacin (GATI) was used as an internal standard (IS). Quantitation was achieved with UV detection at 221, 256 and 267 nm, respectively. Linear calibration curves, at concentration ranges of 0.05‐37.5 μgmL^‐L with a correlation coefficient of ±0.9994. The detection and quantification limits were in the ranges of 0.023‐0.250 μgmL^‐L and 0.071‐0.756 μgmL^‐L, respectively. Friedman's and Student's t‐test were applied to correlate these results. Method was validated in terms of selectivity, linearity, precision, robustness, recovery, limits of detection and quantitation and is applicable to the routine analysis of GFX and H₂‐receptor antagonists, alone or in combination.
