A selective and specific high-performance liquid chromatography method for the determination of daclatasvir enantiomers has been developed and validated. Various immobilized polysaccharide-based chiral stationary phases were used to define a separation strategy utilizing normal-phase and polar organic chromatography modes. Excellent resolution between daclatasvir and its enantiomer was achieved on amylose tris (3-chlorophenylcarbamate) stationary phase, namely CHIRALPAK ID-3, using binary gradient containing acetonitrile:diethylamine and methanol:diethylamine as the mobile phase. The flow rate of the mobile phases was maintained at 1.0 mL min−1 while the column oven temperature was maintained at 40 °C. The column effluent was monitored by UV detection at 315 nm. In comparison with isocratic method, the binary gradient method offered excellent peak shape and improved resolution between daclatasvir and its enantiomer while maintaining the specificity with diastereomers. The method was found to be precise, accurate, and linear (R 2 > 0.999). Limit of detection and limit of quantitation of the enantiomer were found to be 0.083 µg mL−1 as and 0.25 µg mL−1, respectively. Recovery of the enantiomer was found to be in the range of 90 to 112 %.

In this article, we study a finite element approximation for a model free boundary plasma problem. Using a mixed approach (which resembles an optimal control problem with control constraints), we formulate a weak formulation and study the existence and uniqueness of a solution to the continuous model problem. Using the same setting, we formulate and analyze the discrete problem. We derive optimal order energy norm a priori error estimates proving the convergence of the method. Further, we derive a reliable and efficient a posteriori error estimator for the adaptive mesh refinement algorithm. Finally, we illustrate the theoretical results by some numerical examples.  相似文献

In this present study, biodegradable PBAT nanocomposites containing different weight percentages (1, 3, 5, 7, and 10% w/w) of TiO2 nanoparticles were prepared by using solvent casting technique, chloroform as a solvent. The microstructure and morphology of the as‐synthesized poly(butylene adipate‐co‐terephthalate) (PBAT)/TiO2 nanocomposite films were characterized by Fourier‐transform infrared, X‐ray diffraction, scanning electron microscopy, and transmission electron microscope. The thermal degradation of PBAT composites was studied by using thermogravimetric analysis. The mechanical strength of the films was improved by increasing TiO2 concentration. Tensile strength increased from 32.60 to 63.26 MPa, respectively. Barrier properties of the PBAT/TiO2 nanocomposites were investigated by using an oxygen permeability tester. The oxygen permeability (oxygen transmission rate) decreased with increasing the TiO2 nanoparticle concentrations. The PBAT/TiO2 nanocomposite films showed profound antimicrobial activity against both Gram‐positive and Gram‐negative foodborne pathogenic bacteria, namely, Escherichia coli and Staphylococcus aureus, to understand to the zone of inhibition. These results indicated that filler–polymer interaction is important and the role of the TiO2 as a reinforcement in the nanocomposites was evident. Copyright © 2017 John Wiley & Sons, Ltd.  相似文献

This paper analyzes the implicit upwind finite difference scheme on Shishkin-type meshes (including the classical piecewise-uniform Shishkin mesh and the Bakhalov-Shishkin mesh) for a class of singularly perturbed parabolic convection-diffusion problems exhibiting strong interior layers. Suitable conditions on the mesh-generating functions are derived and are found to be sufficient for the convergence of the method, uniformly with respect to the perturbation parameter. Utilizing these conditions, it is shown that the method converges uniformly in the discrete supremum norm with an optimal error bound. Numerical results are presented to validate the theoretical results.  相似文献

(1)H and (13)C NMR spectra were recorded for some novel picrate derivatives derived from some 3,3-methyl-2,6-diarylpiperidin-4-ones and 3-benzyl-2,6-diarylpiperidin-4-one. The difference in the chemical shift of equatorial methylene proton and axial methylene proton at C(5) [Δ = δ(eq)-δ(ax)] is highly negative which is in contrast to the value observed in the corresponding parent piperidin-4-ones and this is attributed to the syn 1,3-diaxial interaction between the axial N-H bond and axial hydrogen at C-5. The effect of protonation on the chemical shifts was studied in detail. The chemical shifts of the heterocyclic ring protons are influenced by the picrate anion. Solvatochromism of picrates were studied in detail. DFT calculations were carried out in order to find out the NBO analysis, HOMO-LUMO energies, MEP studies and hyperpolarisability behaviour. The electric dipole moment (μ) and the first-hyperpolarisability (β) value of the investigated molecules have been studied theoretically which reveal that the synthesized molecules have microscopic non-linear optical (NLO) behaviour with non-zero values.  相似文献

Novel bioactive imidazole derivatives were synthesized and characterized by NMR spectra, mass and CHN analysis. The interaction between the imidazole derivative and bovine serum albumin (BSA) was investigated by fluorescence and UV-vis absorption spectroscopy. The fluorescence quenching of BSA by the imidazole derivatives may be due to the formation of imidazole-BSA complex. The fluorescence quenching mechanism of BSA by imidazole was analyzed and the binding constant has been calculated. The binding distance between imidazole and BSA was obtained based on Forester's non-radiation energy transfer (FRET). The effect of some common ions on the binding constant between imidazole and BSA was also examined.  相似文献

A simple and efficient three components domino reaction of γ-butyrolactam (2-pyrrolidinone), aromatic aldehyde and substituted thiophenol catalyzed by elemental iodine resulted in the formation of 1-((phenylthio)(phenyl)methyl)pyrrolidin-2-one derivatives. The stability of the synthesized analogues was evaluated in stimulated gastric fluid (SGF) and bovine serum albumin (BSA). In vitro anticancer activity was investigated in the low micromolar range and a few analogues were found to possess good activity. This current protocol provides several advantages like shorter reaction time, excellent yield and convenient work-up.  相似文献

The cis/trans isomer ratios of the Xaa-Pyr (Pyr = pyrrolidine) 3° amide bonds are significantly high (∼90% cis) in the novel peptidomimetics where Pyr contains 1,3-oxazine (Oxa) or 1,3-thiazine (Thi) at its 2 position. We find that an unusual n→πi−1 interaction, selectively stabilizes the cis conformer and the n)(n repulsion destabilizes the trans conformer of these molecules. Both these electronic effects oppose the steric effects in the 3° amide bond. The structural requirements for manifestation of these electronic effects are determined.  相似文献

A marine-derived fungus of the genus Stachylidium was isolated from the sponge Callyspongia cf. C. flammea. Chemical investigation of the bioactive fungal extract led to the isolation of the novel phthalimidine derivatives marilines A(1) (1a), A(2) (1b), B (2), and C (3). The absolute configurations of the enantiomeric compounds 1a and 1b were assigned by a combination of experimental circular dichroism (CD) investigations and quantum chemical CD calculations. The skeleton of marilines is most unusual, and its biosynthesis is suggested to require uncommon biochemical reactions in fungal secondary metabolism. Both enantiomers, marilines A(1) (1a) and A(2) (1b), inhibited human leukocyte elastase (HLE) with an IC(50) value of 0.86 μM.  相似文献
