Cytochromes P450 are a ubiquitous group of hemoproteins that perform vital cellular reactions in all lifeforms. Until recently, it was thought that P450s contained non-covalently bound heme. However, it was established that covalent linkage of the heme macrocycle occurs naturally in one major group of the P450 superfamily. The reaction involves heme linkage to a conserved amino acid and is autocatalytic, occurring as a consequence of P450 turnover. This finding presents opportunities to engineer biotechnologically important P450s to covalently link the heme, in order to stabilize cofactor binding and to enhance operational stability of these P450s. This opportunity has been taken in studies on two important bacterial P450s and has produced variants with intriguingly different properties. In this article we survey the developments in the field, the relationships with heme macrocycle ligations in other proteins and the important impact that recent studies of heme ligation have had on our general appreciation of P450 structure and mechanism.  相似文献

Inclusion of a polymer cushion between a lipid bilayer membrane and a solid surface has been suggested as a means to provide a soft, deformable layer that will allow for transmembrane protein insertion and mobility. In this study, mobile, tethered lipid bilayers were formed on a poly(ethylene glycol) (PEG) support via a two-step adsorption process. The PEG films were prepared by coadsorbing a heterofunctional, telechelic PEG lipopolymer (1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-poly(ethylene glycol)-2000-N-[3-(2-(pyridyldithio)propionate]) (DSPE-PEG-PDP) and a nonlipid functionalized PEG-PDP from an ethanol/water mixture, as described in a previous paper (Munro, J. C.; Frank, C. W. Langmuir 2004, 20, 3339-3349). Then a two-step lipid adsorption strategy was used. First, lipids were adsorbed onto the PEG support from a hexane solution. Second, vesicles were adsorbed and fused on the surface to create a bilayer in an aqueous environment. Fluorescence recovery after photobleaching experiments show that this process results in mobile bilayers with diffusion coefficients on the order of 2 microm2/s. The mobility of the bilayers is decreased slightly by increasing the density of tethered lipids. The formation of bilayers, and not multilayer structures, is also confirmed by surface plasmon resonance, which was used to determine in situ film thickness, and by fluorimetry, which was used to determine quantitatively the fluorescence intensity for each 18 by 18 mm sample. Unfortunately, fluorescence microscopy also shows that there are large defects on the samples, which limits the utility of this system.  相似文献

ESCA has been used to characterize films deposited from perfluorobenzene and perfluorobenzene/benzene vapors during plasma polymerization and irradiation with vacuum ultraviolet light. The films deposited by both methods are shown to be essentially the same. This indicates that similar electronically excited states are involved in the formation of the precursors to deposition in both methods and that reaction mechanisms involving ions do not need to be considered for the plasma polymerization of the monomers investigated.  相似文献

A liquid chromatographic method for the determination of sulfachloropyridazine, sulfadiazine, sulfadimethoxine, sulfadoxine, sulfaethoxypyridazine, sulfamethazine, sulfaquinoxaline, and sulfathiazole residues in the muscle, liver, and kidney of food animals using sulfapyridine as internal standard is reported. Tissues are extracted using a modified version of AOAC Official Method 983.31 (Sulfonamide Residues in Animal Tissues). The sample extract is reconstituted in pH 3.0 buffer-acetonitrile (60 + 40) and filtered into an autosampler vial. Using a programmable autosampler of a liquid chromatograph, a portion of the sample is derivatized precolumn with fluorescamine. The sulfonamide derivatives are separated by liquid chromatography using a C18 column with a mobile phase of 0.02M phosphoric acid-acetonitrile (60.5 + 39.5) and detected by fluorescence (excitation, 405 nm; emission, 495 nm). The method was applied to swine and cattle muscle, liver, and kidney; sheep and horse muscle and kidney; and chicken muscle and liver. The mean values for samples fortified with sulfonamides at levels between 0.05 and 0.2 microg/g agreed within 96-99% of spiked levels, with coefficients of variation ranging from 4-10%. The limit of detection (LOD) for all sulfonamides was 0.01 microg/g, with the exception of sulfaquinoxaline, for which the LOD was 0.015 microg/g.  相似文献

This review covers the literature published in 2003 for marine natural products, with 619 citations (413 for the period January to December 2003) referring to compounds isolated from marine microorganisms and phytoplankton, green algae, brown algae, red algae, sponges, coelenterates, bryozoans, molluscs, tunicates and echinoderms. The emphasis is on new compounds (656 for 2003), together with their relevant biological activities, source organisms and country or origin. Biosynthetic studies or syntheses that lead to the revision of structures or stereochemistries have been included (78), including any first total syntheses of a marine natural product.  相似文献

5-Geranoxypsoralen, commonly called bergamottin, a major furocoumarin contained in bergamot oil, is reported in vitro as a highly photoreactive psoralen. In ethanol, it exhibits quite a high triplet state quantum yield (approximately 0.37). The triplet state is involved in subsequent photochemistry which depends on the initial concentration and on the presence of oxygen. In contrast to most psoralens, absorption and fluorescence data suggest that 5-geranoxypsoralen does not interact with DNA in the dark. No UVA-induced interstrand cross-links in DNA were shown.  相似文献

We report the X-ray crystal structures of two different iron(III) porphyrinates: [Fe(OEP)(NO3)] and [Fe(TPP)(NO3)]. The first complex has the nitrate ion coordinated by a single oxygen atom while the second derivative has the nitrate coordinated in a symmetric bidentate fashion. This latter structure is a redetermination that shows some differences from an earlier structure; the difference appears to be the result of an unrecognized nitrate ion disorder in the earlier structure determination. Changes in physical properties of three species [Fe(TPivP)(NO3)], [Fe(OEP)(NO3)], and [Fe-(TPP)-(NO3)] as a function of coordination mode were examined by Mössbauer and EPR spectroscopies; EPR spectra appear to be most sensitive to the change in coordination mode.  相似文献

When {2,2′‐[(2‐methyl‐2‐nitropropane‐1,3‐diyl)diimino]diacetato}copper(II), [Cu(C8H13N3O6)], (I), was crystallized from a binary mixture of methanol and water, a monoclinic two‐dimensional water‐ and methanol‐solvated metal–organic framework (MOF) structure, distinctly different from the known orthorhombic one‐dimensional coordination polymer of (I), was isolated, namely catena‐poly[[copper(II)‐μ3‐2,2′‐[(2‐methyl‐2‐nitropropane‐1,3‐diyl)diimino]diacetato] methanol 0.45‐solvate 0.55‐hydrate], {[Cu(C8H13N3O6)]·0.45CH3OH·0.55H2O}n, (II). The monoclinic structure of (II) comprises centrosymmetric dimers stabilized by a dative covalent Cu2O2 core and intramolecular N—H...O hydrogen bonds. Each dimer is linked to four neighbouring dimers via symmetry‐related (opposing) pairs of bridging carboxylate O atoms to generate a `diamondoid' net or two‐dimensional coordination network. Tight voids of 166 Å3 are located between these two‐dimensional MOF sheets and contain a mixture of water and methanol with fractional occupancies of 0.55 and 0.45, respectively. The two‐dimensional MOF sheets have nanometre‐scale spacings (11.2 Å) in the crystal structure. Hydrogen‐bonding between the methanol/water hydroxy groups and a Cu‐bound bridging carboxylate O atom apparently negates thermal desolvation of the structure below 358 K in an uncrushed crystal of (II).  相似文献
