A continuous flow, microwave-assisted, parallel-capillary microreactor has been developed. Libraries of drug candidates were prepared on the milligram scale with this reactor by injecting plugs of reagents from separate syringes into common reaction capillaries, thereby producing discrete compounds in excellent yield and purity. Microwave irradiation provides the necessary energy that existing room-temperature microreactor technology lacks for higher activation barrier transformations, producing the required amounts of desired compounds in minutes or less.  相似文献

A major focus of current efforts in genomics is to elucidate the genetic variations extent within the human population, and to study the effects of these variations upon the human system. The most common type of genetic variations are the single nucleotide polymorphisms (SNPs), which occur every 500-1000 nt in the genome. Large-scale population association studies to study the biological or medical significance of such variations may require the analysis of hundreds of thousands of SNPs on thousands of individuals. We are pursuing development of an approach to large-scale SNP analysis that combines the specificity of invasive cleavage reactions with the parallelism of high density DNA arrays. A surface-immobilized probe oligonucleotide is specifically cleaved in the presence of a complementary target sequence in unamplified human genomic DNA, yielding a 5' phosphate group. High sensitivity detection of this reaction product on the surface is achieved by the use of rolling circle amplification, with an approximate concentration detection limit of 10 fM target DNA. This combination of very specific surface cleavage and highly sensitive surface detection will make possible the rapid and parallel analysis of genetic variations across large populations.  相似文献

The reaction of [Os3(CO)12] with tetramethylthiourea in the presence of a methanolic solution of Me3NO·2H2O at 60° yields the compounds [Os3(CO)11{η 1-SC(NMe2)2}] (1) in 56% yield and [Os3(CO)9(μ-OH)(μ-MeOCO){η 1-SC(NMe2)2}] (2) in 10% yield in which the tetramethylthiourea ligand is coordinatedvia the sulfur atom at an equatorial position. Compound2 is a 50 e? cluster with two metal-metal bonds and the hydroxy and methoxycarbonyl ligands bridging the open metal-metal edge. In contrast, the analogous reaction of [Os3(CO)12] with thiourea gives the compounts [(μ-H)Os3(CO)10{μ-NHC(S)NH2}] (3) in 8% yield and [(μ-H)Os3(CO)9{3-NHC(S)NH2}] (4) in 30% yield. In3, the thioureato ligand bridges two osmium atomsvia the sulfur atom, whereas in4 in addition to the sulfur bridge, one of the nitrogen atoms of thioureato moiety bonds to the remaining osmium atom. The decacarbonyl compounds 3 can also be obtained in 50% yield from the reaction of [Os3(CO)10(MeCN)2] with thiourea at ambient temperature. Compound3 converts to4 (65%) photochemically. Compound1 reacts with PPh3 and acetonitrile at ambient temperature to give the simple substitution products [Os3(CO)11(PPh3)] and [Os3(CO)11(MeCN)], respectively, while with pyridine, the oxidative addition product [(μ-H)Os3(CO)10(μ-NC5H4] is formed at 80°C. All the new compounds are characterized by IR,1-H-NMR and elemental analysis together with the X-ray crystal structures of1,2 and4. Compound1 crystallizes in the triclinic space group P $P\bar 1$ with unit cell parametersa = 8.626(3) Å,b = 11.639(3) Å,c = 12.568(3_ Å,α = 84.67(2)°,β = 75.36(2)°,γ = 79.49(3)°,V = 1199(1) Å3, andZ = 2. Least-squares refinement of 4585 reflections gave a final agreement factor ofR = 0.0766 (R w = 0.0823). Compound2 crystallizes in the monoclinic space group P21/n with unit cell parametersa = 9.149(5) Å,b = 17.483(5) Å,c = 15.094(4) Å,β = 91.75(2)°,V = 2413(2) Å3, andZ = 4. Least-squares refinement of 3632 reflections gave a final agreement factor ofR = 0.0603 (R w = 0.0802). Compound4 crystallizes in the monoclinic space group C2/c with unit cell parametersa = 13.915(7) Å,b = 14.718(6) Å,c = 17.109(6) Å,β = 100.44(3)°,V = 3446(5) Å3, andZ = 8. Lipid membranes composed of phosphatidyl choline and cholesterol were interfaced to polyacrylamine hydrogen by Langmuir—Blodgett thin-film deposition. The extent of lipid adsorption to the gel surface was critically dependent on the hydration of the polymer as determined by contact angle measurements. Some electrochemical transducers incorporating the deposited membrane/gel structure responded positively to phloretin and valinomycin. Limitations to the construction of this device are discussed.  相似文献

The reaction between AuCl(PPh3 and [Fe(μ3-HCNBut)(CO)9]? gives AuFe33-HCNBut)(CO)9(PPh3), crystals of which are triclinic, space group P1, with a 12.815(3), b 16.265(4), c 19.106(3) Å, α 67.15(3), β 73.46(2), γ 73.12(2)° and Z = 4. The comlex contains an AuFe3 “butterfly” cluster, the Fe3 face of which is bridged by the HCNBut ligand bonded in the (2σ + π) mode on the side opposite to the Au(PPh3) moiety, which, contrary to expectation based on the analogy with H, bridges the two Fe atoms σ-bonded to N, and π-bonded to the CN group. The AuFe2/Fe3 dihedral angles in the two independent molecules differ significantly, with values of 110.9 and 132.1°.  相似文献

As it is shown by crystal structure determinations { 1 : ?90°C; I 4 2d; a = 1017,3(4); c = 3738,0(26) pm; Z = 8; R w = 0,065; 2 : + 20°C; P2 1 /c; a = 680,9(4); b = 1704,8(13); c = 1197,9 (10) pm; β = 119,46(6)°; Z = 2; R w = 0,084} both compounds form chains which in the case of bismuthide 1 are built up as screws of alternating bismuth and lithium atoms; bonding further to two trimethylsilyl groups or to the chelating DME ligand both atoms gain coordination number 4 {Li? Bi 292(3); Bi? Si 263.3(14) pm; Bi? Li? Bi 132(1); Li? Bi? Li 148(1); φ(Li? Bi? Li? Bi) 83°}. In the case of dibismuthane 2 the centrosymmetric molecules are strung, their Bi-Bi groups forming nearly linear zigzag chains with shortened intermolecular contact distances {Bi-Bi 303.5(3); Bi …? Bi 380.4(3); Bi? Si 268 pm; Bi? Bi …? Bi 169; Bi? Bi? Si 97.4(5) and 92.0(5)°}.     Abstract— The difference (in cm−1) in absorption maxima between the protonated Schiff base of retinals and the pigment derived therefrom has been defined as the opsin shift. It represents the influence of the opsin binding site on the chromophore. The analysis of the opsin shifts of a series of dihydrobacteriorhodopsins has led to the external point-charge model, which in addition to a counter anion near the Schiff base ammonium, carries another negative charge in the vicinity of the β-ionone ring. This is in striking contrast to the external point-charge model proposed earlier for the bovine visual pigment. The absorption maxima of rhodopsins formed from bromo- and phenyl retinals support the two models. A retinal carrying a photoaffinity label has yielded a nonbleachable bacteriorhodopsin.  相似文献

    A study of the effect of the various solution components on the kinetics of the polymerization of acrylamide in water/oil (w/o) microemulsions has been performed. For the polymerizations with toluene as the continuous phase, both the rate of polymerization, Rp, and the molecular weight of the polyacrylamide were found to be first order in monomer concentration. Furthermore, for the low temperatures (10°C) involved in these experiments, nondegradative chain transfer to monomer appears to be insignificant. When the continuous-phase solvent was changed, an exponential dependence, X, of Rp on the incident light intensity in the order of toluene (X = 1.06) > heptane (X = 0.73) > benzene (X = 0.55) was found. Thus, the monoradical termination found in the toluene microemulsions is likely due to degradative transfer to toluene, forming a stable benzyl radical, while polymerization in benzene (no labile hydrogen atoms) leads to biradical termination  相似文献

    The surface compositions of various polymeric films, grown electrochemically on platinum foils, have been investigated by energy-dispersive x-ray analysis in conjunction with scanning electron microscopy (SEM/EDS). Comparison of the relative area ratios of peaks for the C and N Kemission lines show that the EDS may be used to study the surface composition of polymers. The evidence presented strongly suggests that there is limited structural degradation and the elemental composition is not changed under the electron beam at relatively low accelerating voltages. This technique statistically samples the repeat units of the polymer. For samples grown in both aqueous and nonaqueous solutions. SEM/EDS provides evidence for extensive contamination with oxygen.  相似文献
