In stability studies on moisture-resistant Opalmon tablets in press-through-packages (PTP), which were placed in aluminum bags, we found that the degradation rate of the dextran formulation is faster than that of the lactose formulation. The fast degradation of the dextran formulation is attributed to residual moisture in the package because drying the tablets before packaging suppressed the degradation and there is a good correlation between the stability of the drug and the water-activity of the tablets. Therefore, we developed a new drying method for the tablets, i.e. the rotary vacuum drying method, and investigated the effects of the operating conditions such as heating temperature, rotation speed, and vacuum degree on the drying time, and the appearance of the tablets. Using the rotary vacuum drying method, the tablets were dried over a short time (30 min) on a mass production scale so that the water activity was less than 0.03. Furthermore, the tablets suffered negligible damage such as breaking and chipping during the drying process. These results indicate that the rotary vacuum drying method is useful for drying tablets on mass production scales. 相似文献

Ion-exclusion chromatography (IEC) finds applications in various different analytical separations of weak acids. Pure, deionized water or a diluted, aqueous solution of a strong mineral acid (such as, e.g., sulphuric acid) is used as the mobile phase, whereas a typical stationary phase is a strongly acidic resin in the H(+) form (e.g., the sulfonated polystyrene-divinylbenzene resin with a high ion-exchange capacity, provided by the sulfonic acid groups). When pure water is used as the mobile phase, then the characteristic leading (i.e., frontally tailing) peaks are obtained, and the retention depends mainly on the concentration of the analyte. An alternative technique is vacancy ion-exclusion chromatography (v-IEC), in which the column is equilibrated with the sample solution, flowing as the mobile phase through the system, and pure water is injected as the sample. In this case, the symmetrical vacant peaks are obtained. The aim of this paper is to describe the retention mechanism in IEC and v-IEC for the adsorptive and nonadsorptive acids in analytical and concentration overload conditions, with pure water and the diluted sulphuric acid solution as the two different mobile phases. The retention times and the peak shapes predicted by the derived equations remain in a good qualitative and quantitative agreement with the experimental data. The model proposed in this paper predicts the new features characteristic of IEC for the adsorptive acids. These are, namely, an increase in the retention time of the peak apexes (up to a certain level and concurring with an increase in the acid concentration), followed by a subsequent decrease of the retention time (with the further growth of the acid concentration in the eluent). Similar changes in the retention time observed for v-IEC in the specific adsorption conditions were also correctly predicted by the model. 相似文献

Direct observation of the unstable intermediate in the radical addition reaction of the oxime ether 1 mediated by triethylborane (Et(3)B) is described using (1)H and (11)B micro channeled cell for synthesis monitoring (MICCS), which was recently developed as an interfacing microchip for NMR. It was possible that the signal of the intermediate was observed as a result of using MICCS technique with a standard NMR instrument. This result supports the structure of the intermediate analyzed by diffusion-ordered spectroscopy (DOSY) NMR method in a previous paper. The procedure of micro channeled cell for synthesis monitoring-nuclear magnetic resonance (MICCS-NMR) was much easier than that of DOSY method. Photo-Fries rearrangement reactions of 1-naphthyl acetate (NA) in n-hexane and in cyclohexane were studied by the magnetic field effect probe (MFE probe) under magnetic fields (B) of 0 to 7 T. Transient absorptions of the 1-naphthoxyl radical, T-T absorption of NA, and a short-lifetime intermediate (τ = 24 ns) were observed by a nanosecond laser flash photolysis technique. In n-hexane, the yield of escaped 1-naphthoxyl radicals dropped dramatically upon application of a 3 mT field, but then the yield increased with increasing B for 3 mT < B≤ 7 T. These observed MFEs can be explained by the hyperfine coupling and the Δg mechanisms through the singlet radical pair. The fact that MFEs were observed for the present photo-Fries rearrangement reaction indicates the presence of a singlet radical pair intermediate with a lifetime as long as several tens of nanoseconds. 相似文献

The absolute configuration of a binaphthyl-azobenzene dyad 2b, which has a chiral axis and a chiral plane, was determined by comparing the experimental circular dichroism (CD) spectra with the theoretical CD spectra calculated by the time-dependent (TD)-DFT method. The CD signals of the trans-azobenzene moiety indicated that the two benzene rings of this moiety are twisted unidirectionally.
