Electronic detection of single-base mismatches in DNA with ferrocene-modified probes.

Genotyping and gene-expression monitoring is critical to the study of the association between genetics and drug response (pharmacogenomics) and the association of sequence variation with heritable phenotypes. Recently, we developed an entirely electronic method for the detection of DNA hybridization events by the site-specific incorporation of ferrocenyl derivatives into DNA oligonucleotides. To perform rapid and accurate point mutation detection employing this methodology, two types of metal-containing signaling probes with varying redox potentials are required. In this report we describe a new ferrocene-containing phosphoramidite 9 that provides a range of detectable redox potentials. Using automated DNA/RNA synthesis techniques the two ferrocenyl complexes were inserted at various positions along oligonucleotide probes. Thermal stability analysis of these metal-containing DNA oligonucleotides indicates that incorporation of 9 resulted in no destabilization of the duplex. A mixture of oligonucleotides containing compounds 9 and I was analyzed by alternating current voltammetry (ACV) monitored at the 1st harmonic. The data demonstrate that the two ferrocenyl oligonucleotide derivatives can be distinguished electrochemically. A CMS-DNA array was prepared on an array of gold electrodes on a printed circuit board substrate with a self-assembled mixed monolayer, coupled to an electronic detection system. Experiments for the detection of a single-base match utilizing two signaling probes were carried out. The results demonstrate that rapid and accurate detection of a single-base mismatch can be achieved by using these dual-signaling probes on CMS-DNA chips.     

The chemical constituents from Schisandra propinqua var intermedia

A new cirmamic acid monoterpenoid ester, intermedin C (1), was isolated from the aerial parts of Schisandra propinqua vat. intermedia. The structure of 1 was elucidated by spectroscopic methods including extensive 1D and 2D NMR techniques.     

Ablation mechanism study on metallic materials with a 10 ps laser under high fluence

Single shot ablation of metallic materials of aluminium, titanium alloy (Ti6Al4V) and gold has been studied with 10 picoseconds (ps) laser pulses experimentally and theoretically. The ablation rate variation at high fluence was explained by a simplified predictive model based on critical-point phase separation (CPPS) theory. A comparison between experimental and numerical results inferred that CPPS may well be the dominant ablation mechanism for high fluence laser ablation at 10 ps laser duration.     

Synthesis of achiral PEG‐PANI rod‐coil block copolymers and their helical superstructures

Poly(ethylene glycol) (PEG) was modified with aniline groups at both the end, and then PEG‐PANI rod‐coil block polymers have been synthesized by polymerization of the aniline with the aniline‐modified PEG. FTIR, NMR, and elemental analysis provided the chemical strucutre of the as‐prepared polymers. The achiral rod‐coil copolymer could form different superstructures by means of self‐assembly when adding diethyl ether into its THF solution and the length of PANI segments is a key factor to the superstructures. AFM measurements revealed that they form spring‐like helical superstructures from the short PANI‐containing copolymers while these form fibrous helical superstructures from the longer PANI‐containing copolymer. A possible mechanism of the helical superstructures is suggested in this article and the driving force is believed the π–π stacking of the rigid segment of the copolymers. © 2007 Wiley Periodicals, Inc. Algorithm of orbit determination using one or two GPS satellites

The problem of orbit determination using one or two GPS satellites is discussed. Methods of getting initial values based on linear translation is presented; the Secant method and the descend Newton iterative procedure and the continuation algorithm are used synthetically to solve the nonlinear equations. Computer simulation shows that this algorithm can give preliminary state of satellite orbit with a certain precision in short time.     

Periodic Wave Solutions for （2＋1）-Dimensional Boussinesq Equation and （3＋ 1）-Dimensional Kadomtsev-Petviashvili Equation

For describing various complex nonlinear phenomena in the realistic world, the higher-dimensional nonlinear evolution equations appear more attractive in many fields of physical and engineering sciences. In this paper, by virtue of the Hirota bilinear method and Riemann theta functions, the periodic wave solutions for the （2＋1）-dimensional Boussinesq equation and （3＋1）-dimensional Kadomtsev Petviashvili （KP） equation are obtained. Furthermore, it is shown that the known soliton solutions for the two equations can be reduced from the periodic wave solutions.     

Adaptive finite element methods for the identification of distributed parameters in elliptic equation

In this paper, adaptive finite element method is developed for the estimation of distributed parameter in elliptic equation. Both upper and lower error bound are derived and used to improve the accuracy by appropriate mesh refinement. An efficient preconditioned project gradient algorithm is employed to solve the nonlinear least-squares problem arising in the context of parameter identification problem. The efficiency of our error estimators is demonstrated by some numerical experiments.
