This paper uses chemical shift surfaces to simulate experimental (13)C cross polarization magic angle spinning spectra for amorphous solid state disaccharides, paying particular attention to the glycosidic linkage atoms in trehalose, sucrose, and lactose. The combination of molecular mechanics with density functional theory/gauge invariant atomic orbital ab initio methods provides reliable structural information on the conformational distribution in the glass. The results are interpreted in terms of an enhanced flexibility that trehalose possesses in the amorphous solid state, at least on the time scale of (13)C nuclear magnetic resonance measurements. Implications of these findings for the fragility of trehalose glass and bioprotectant action are discussed.  相似文献

IceCube has become the first neutrino telescope with a sensitivity below the TeV neutrino flux predicted from gamma-ray bursts if gamma-ray bursts are responsible for the observed cosmic-ray flux above 10(18) eV. Two separate analyses using the half-complete IceCube detector, one a dedicated search for neutrinos from pγ interactions in the prompt phase of the gamma-ray burst fireball and the other a generic search for any neutrino emission from these sources over a wide range of energies and emission times, produced no evidence for neutrino emission, excluding prevailing models at 90% confidence.  相似文献

We demonstrate controlled high-order harmonic generation in gas using high-energy femtosecond pulses (50 fs-50 mJ on target) by performing spatial shaping of the terrawatt fundamental laser beam. We have developed a two optical paths mirror that can withstand high power and shape the pump beam into a quasi-flat-top profile (super Gaussian) near focus. We observe clear signatures of the spatial shaping on the harmonic beam in terms of profile, divergence, level of signal, and spectrum. The harmonic generation in neon with a quasi-flat-top beam results in a broadband extreme UV beam with extremely low divergence (~340 μrad).  相似文献

Essential oils extracted from leaves and fruits of Schinus areira (Anacardiaceae) were tested for their repellent, toxic and feeding deterrent properties against Tribolium castaneum (Coleoptera: Tenebrionidae) larvae and adults. A topical application assay was employed for the contact toxicity study and filter paper impregnation for the fumigant assay. A treated diet was also used to evaluate the repellent activity and a flour disk bioassay for the feeding deterrent action and nutritional index alteration. The essential oil of the leaves contained mainly monoterpenoids, with alpha-phellandrene, 3-carene and camphene predominant, whereas that from the fruits contained mainly alpha-phellandrene, 3-carene and beta-myrcene. The leaf essential oil showed repellent effects, whereas that from the fruit was an attractant. Both oils produced mortality against larvae in topical and fumigant bioassays, but fumigant toxicity was not found against adults. Moreover, both essential oils produced some alterations in nutritional index. These results show that the essential oils from S. areira could be applicable to the management of populations of Tribolium castaneum.  相似文献

We report on the optimization of nano‐LC gradient separations of proteomic samples that vary in complexity. The gradient performance limits were visualized by kinetic plots depicting the gradient time needed to achieve a certain peak capacity, while using the maximum system pressure of 80 MPa. The selection of the optimal particle size/column length combination and corresponding gradient steepness was based on scouting the performance of 75 μm id capillary columns packed with 2, 3, and 5 μm fully porous silica C18 particles. At optimal gradient conditions, peak capacities up to 500 can be obtained within a 120 min gradient using 2 μm particle‐packed capillary columns. Separations of proteomic samples including a cytochrome c tryptic digest, a bovine serum albumin tryptic digest, a six protein mix digest, and an Escherichia coli digest are demonstrated while operating at the kinetic‐performance limit, i.e. using 2‐μm packed columns, adjusting the column length and scaling the gradient steepness according to sample complexity. Finally, good run‐to‐run retention time stability (RSD values below 0.18%) was demonstrated applying ultra‐high pressure conditions.  相似文献

The microwave-assisted decomposition of DMF was thoroughly studied and the reaction conditions (temperature, solvent effect, and effect of additives, such as acids, bases, and salts) were optimized for its use in amination reactions. The one step synthesis of difluoromethylphosphonamidates from dialkylphosphonates is reported. The addition of lithiated amides onto dialkyl difluoromethylphosphonates afforded the corresponding phosphonamidates, as potential prodrug precursors. The remarkable high stability of these phosphonamidates in acidic medium was studied by 31P NMR.  相似文献

A composite In-Pb:carbon was successfully synthetized by a two-step mechanochemical synthesis in order to obtain an adequate particles size and structure to investigate the electrochemical reactivity of the In-Pb solid solution towards Mg.A potential synergetic coupling of electroactive elements In and Pb was examined using electrochemical and ex situ X-ray diffraction analyses.The potential profile of the solid solution indicates the formation of Mg2Pb and Mg In.However,the diffraction study suggests a peculiar electrochemically-driven amorphization of Mg In during the magnesiation,in strong contrast to Mg In crystallization in In-based and In Bi-based electrodes reported in the literature.Combining In and Pb favors the amorphization of Mg In and a high first magnesiation capacity of about 550 m Ah g-1,but is thereafter detrimental to the material’s reversibility.These results emphasize the possible influence of electrochemically-driven amorphization and crystallization processes on electrochemical performance of battery materials.  相似文献

The failure to produce and/or crystallize proteins is often due to their modular structure. There exists therefore considerable interest to develop strategies for tailoring proteins into crystallizable domains. In the framework of a Structural Genomics Project on soluble yeast proteins, we have tested the expression of numerous genetic constructs of our targets in order to produce and crystallize proteins and protein domains and solve their three-dimensional structure. In some cases, the choice of the domain boundaries was guided by prediction from sequence using various software packages, including Prelink, a home-made prediction method for detecting unfolded regions. In other cases, large numbers of constructs were generated using molecular biology or biochemical methods. In this paper, we analyze the results of the over-expression in E. coli and crystallization of these constructs, and compare these with the predictions that can be obtained from our software and from others.  相似文献

Two novel chromatographic methods both based on utilization of sub-2-micron particle columns were developed for the analysis of phenolic compounds in this work. An HPLC system was equipped with C(18) silica-based analytical column (50 mm x 4.6 mm, 1.8 microm) and a UPLC system with ethylene-bridged hybrid C(18) analytical column (100 mm x 2.1 mm, 1.7 microm). In total 34 phenolic substances were divided into groups of phenolic acids, flavonoids, catechins and coumarins and were analysed in sequence using different gradient methods. System suitability test data, including repeatability of retention time and peak area, mean values of asymmetry factor, resolution, peak capacity and the height equivalent of a theoretical plate were determined for each gradient method by 10 replicate injections. The developed methods were applied in the analysis of real samples (grape wines, teas).  相似文献
