Instrumental neutron activation analysis was used for determination of vanadium mass fraction in crystals of incipient ferroelectric strontium titanate and ferroelectric barium titanate. In order to improve vanadium limit of detection, discriminating gamma-ray spectrometry was used by inserting an absorption filter between the samples and an HPGe detector. The use of the absorption lead filter 6-mm thick yielded improvement of the vanadium limit of detection by a factor of two. The vanadium mass fraction determined in a quality control sample, which was NIST standard reference material SRM 1648 Urban Particulate Matter, was in close agreement with the certified value.

The term immuno/bioassay refers to analytical techniques that utilize the specific molecular recognition between antibodies and antigens or between biomolecules and specific receptors. Features such as high selectivity and low sample consumption make them very useful for analysis of samples with complex matrices. However, immuno/bioassays involve time-consuming (multi-step) operations which usually consist of steps of multiple incubation and washing. These are tedious and may result in large errors. Automatted immuno/bioassay systems can ease and shorten these processes and thus are highly beneficial. The hyphenation of flow-based techniques (i.e. flow injection related techniques and micro-fluidic systems) with immuno/bioassay protocols paves a new way for performing such assays. Compared to conventional micro-plate formats, flow-based immuno/bioassays can reduce the time needed for analysis, the volumes of samples and reagents consumed, and the need for trained personnel. In order to transform immuno/bioassays from conventional to flow-based formats, the solid surfaces used for the immobilization step has to be changed in order to meet the specific requirements of flow systems. To further develop the on-site analytical systems in micro-fluidic platforms, improvements in detection methods are necessary for high-sensitivity and rapid measurement. This review overviews the advantages and disadvantages of flow-based immuno/bioassay formats, the various types of solid surfaces for immobilization, and the methods of detection. Trends to improve sensitivity, speed and robustness are emphasized.  相似文献

Coupling nucleic acid processing enzymes to nanoscale pores allows controlled movement of individual DNA or RNA strands that is reported as an ionic current/time series. Hundreds of individual enzyme complexes can be examined in single-file order at high bandwidth and spatial resolution. The bacteriophage phi29 DNA polymerase (phi29 DNAP) is an attractive candidate for this technology, due to its remarkable processivity and high affinity for DNA substrates. Here we show that phi29 DNAP-DNA complexes are stable when captured in an electric field across the α-hemolysin nanopore. DNA substrates were activated for replication at the nanopore orifice by exploiting the 3'-5' exonuclease activity of wild-type phi29 DNAP to excise a 3'-H terminal residue, yielding a primer strand 3'-OH. In the presence of deoxynucleoside triphosphates, DNA synthesis was initiated, allowing real-time detection of numerous sequential nucleotide additions that was limited only by DNA template length. Translocation of phi29 DNAP along DNA substrates was observed in real time at ?ngstrom-scale precision as the template strand was drawn through the nanopore lumen during replication.  相似文献

The ligand 6,6"bis(4-methoxyphenyl)-4'-phenyl-2,2':6',2"terpyridine (2) has been prepared and characterized; deprotection using pyridinium chloride leads to the formation of 6,6"bis(4-hydroxyphenyl)-4'-phenyl-2,2':6',2"terpyridinium chloride ([H3]Cl). Treatment of the latter with 3-(2-(2-bromoethoxy)ethoxy)prop-1-ene under basic conditions yields ligand 4 containing pendant, alkene-terminated chains. Whereas direct complexation of 4 with ruthenium(II) proved problematical, the homoleptic complexes [Fe(2)(2)][PF(6)](2) and [Ru(2)(2)][PF(6)](2) were prepared in good to moderate yields. In the solid state, both complexes exhibit multiple face-to-face π-stacking of arene and pyridine rings which influences the coordination geometry about the metal ion. Consequential weakening of the ligand field results in [Fe(2)(2)][PF(6)](2) being high-spin. Variable temperature solution (1)H NMR spectroscopic studies confirm the iron(ii) centre remains high-spin between 200 and 295 K. The paramagnetically shifted (1)H NMR spectrum exhibits signals in the range δ 109.7 to -66.5 ppm and has been fully assigned. Paramagnetic relaxation enhancement (PRE) has been used to correlate the observed proton line-widths to the distances of the protons from the metal centre and these are in good agreement with the Fe···H separations observed in the solid state. The [Fe(2)(2)](2+) ion undergoes two dynamic processes (i) rotation of the pendant phenyl rings which is fast on the NMR timescale at 200 K, and (ii) twisting and sliding of the aromatic rings of the tpy and anisyl units which interconverts the two enantiomers of [Fe(2)(2)](2+) at 295 K.  相似文献

A method for the fast determination of yohimbine, a potent adrenoreceptor antagonist used for the treatment of sexual dysfunctions, is proposed in this article. MEKC under basic and acidic conditions (sodium borate, pH 9.5 and sodium phosphate, pH 2.5) with SDS was developed. The effect of the experimental parameters, e.g. pH, SDS concentration and injection time, on yohimbine migration was also studied. Both methods were validated in terms of linearity, limits of detection and quantification, accuracy, and precision using caffeine as an internal standard. The application for the determination of yohimbine in hand-made medicaments is also investigated in this study.   相似文献

This paper examines four acoustic properties (duration F0, F1, and F2) of the monophthongal vowels of Iberian Spanish (IS) from Madrid and Peruvian Spanish (PS) from Lima in various consonantal contexts (/s/, /f/, /t/, /p/, and /k/) and in various phrasal contexts (in isolated words and sentence-internally). Acoustic measurements on 39 speakers, balanced by dialect and gender, can be generalized to the following differences between the two dialects. The vowel /a/ has a lower first formant in PS than in IS by 6.3%. The vowels /e/ and /o/ have more peripheral second-formant (F2) values in PS than in IS by about 4%. The consonant /s/ causes more centralization of the F2 of neighboring vowels in IS than in PS. No dialectal differences are found for the effect of phrasal context. Next to the between-dialect differences in the vowels, the present study finds that /s/ has a higher spectral center of gravity in PS than in IS by about 10%, that PS speakers speak slower than IS speakers by about 9%, and that Spanish-speaking women speak slower than Spanish-speaking men by about 5% (irrespective of dialect).  相似文献

Many objects studied in biology, medicine or material sciences create spatial formations of random shape in which we can observe mutual interactions among those objects. In order to analyse the data composed of such patterns, we use the methods of spatial statistics. Recently, extended random-disc Quermass-interaction process was studied, simulated and consequently statistically analysed using MCMC maximum likelihood method (MCMC MLE). However, this analysis brought some problems. First, it was quite time-consuming, secondly, in some special cases, the parameter estimates may undervalue the real parameter values.
