Phosphorus in organic synthesis—XI Amino acids and peptides—XXI : Reaction of diethyl phosphorocyanidate(DEPC) with carboxylic acids. A new synthesis of carboxylic esters and amides

Diethyl phosphorocyanidate(DEPC) reacts with carboxylic acids in the presence of triethylamine leading to transient formation of acyl cyanides, but in presence of alcohols or amines, carboxylic esters or amides are produced. DEPC is especially effective for the synthesis of amides and peptides, and showed a satisfactory result on the Young racemization test.     

Immobilized urokinase column as part of a specific detection system for plasminogen species separated by high-performance affinity chromatography

Immobilized urokinase was used as part of a post-column reactor for the specific detection of human plasminogen species which were fractionated using a high-performance affinity column. After on-line activation of each peak, plasmin activity was measured by mixing the eluate with a specific fluorogenic substrate and the product was detected by a fluorescence monitor. This detection system gave linear calibration graphs for both purified plasminogens (0.1-50 micrograms) and plasminogens contained in plasma (25-100 microliters). Relative standard deviations for the determination of plasminogens in plasma were 6.1-6.6% (n = 12), showing good reproducibility. The detection limit was as low as 0.1 micrograms of plasminogen. Immobilized urokinase was very stable and no appreciable decrease in activity was found after 100 cycles of operation. In combination with an immobilized benzamidine column, this system made it possible to separate and detect Glu-plasminogen and Lys-plasminogen contained in human plasma samples as small as 100 microliters without any pretreatment.     

BLUE LIGHT PHOTORECEPTION IN Neurospora ORCADIAN RHYTHM: EVIDENCE FOR INVOLVEMENT OF THE FLAVIN TRIPLET STATE

The mechanism of the photoreceptor acting on the circadian conidiation rhythm of Neurospora crassa was studied, with the following results: (1) the efficiency of 8-haloflavins as sensitizers increased with their triplet yields. (2) Phase shifts were not abolished by removal of oxygen prior to illumination. (3) Oxygen inhibited phase shifts when introduced into the cultures after light treatment. It is proposed that the blue light photoreceptor for the circadian clock of Neurospora crassa acts (1) from its triplet state, but (2) not via singlet oxygen; (3) signal transduction involves (an) oxygen-sensitive intermediate(s).     

Liquid-liquid extraction separation and sequential determination of plutonium and americium in environmental samples by alpha-spectrometry

A procedure is described by which plutonium and americium can be determined in environmental samples. The sample is leached with nitric acid and hydrogen peroxide, and the two elements are co-precipitated with ferric hydroxide and calcium oxalate. The calcium oxalate is incinerated at 450 degrees and the ash is dissolved in nitric acid. Plutonium is extracted with tri-n-octylamine solution in xylene from 4M nitric acid and stripped with ammonium iodide/hydrochloric acid. Americium is extracted with thenoyltrifluoroacetone solution in xylene at pH 4 together with rare-earth elements and stripped with 1M nitric acid. Americium and the rare-earth elements thus separated are sorbed on Dowex 1 x 4 resin from 1M nitric acid in 93% methanol, the rare-earth elements are eluted with 0.1M hydrochloric acid/0.5M ammonium thiocyanate/80% methanol and the americium is finally eluted with 1.5M hydrochloric acid in 86% methanol. Plutonium and americium in each fraction are electro-deposited and determined by alpha-spectrometry. Overall average recoveries are 81% for plutonium and 59% for americium.     

Preparation of adsorbents for affinity chromatography using TSKgel Tresyl-Toyopearl 650M

The optimum conditions for the coupling of proteins were investigated using TSKgel Tresyl-Toyopearl 650M. They were dependent on the proteins coupled. For example, when soybean trypsin inhibitor was coupled at pH 8 the coupling was completed within 1 h and the subsequent adsorption capacity for trypsin was maximal. Longer coupling times decreased the adsorption capacity due to multi-point attachment. The adsorbents obtained were successfully used for affinity chromatography in a short time.     

A Nickel Complex Containing a Pyramidalized,Ambiphilic Pincer Germylene Ligand

Reaction of N-heterocyclic carbene (NHC)-stabilized PGeP-type germylene Ge{o-(PiPr₂)C₆H₄}₂⋅^MeIiPr ( 1 ) (^MeIiPr=1,3-diisopropyl-4,5-dimethylimidazol-2-ylidene) with Ni(cod)₂ gave pincer germylene complex Ni[Ge{o-(PiPr₂)C₆H₄}₂](^MeIiPr) ( 2 ), in which the Ge center of 2 is significantly pyramidalized. Theoretical calculation on 2 predicted the ambiphilicity of the germanium center, which was confirmed by reactivity studies. Thus, complex 2 reacted with both Lewis base ^MeIMe (^MeIMe=1,3,4,5-tetramethylimidazol-2-ylidene) and Lewis acid BH₃⋅SMe₂ at the germanium center to afford the adducts Ni[Ge{o-(PiPr₂)C₆H₄}₂⋅^MeIMe](^MeIiPr) ( 3 ) and Ni[Ge{o-(PiPr₂)C₆H₄}₂⋅BH₃](^MeIiPr) ( 4 ), respectively. Furthermore, the former was slowly converted to dinuclear complex Ni₂[Ge{o-(PiPr₂)C₆H₄}₂]₂(^MeIMe)₂ ( 5 ) at room temperature. Complex 5 can be regarded as a dimer of the ^MeIMe analog of 2 with a Ni-Ge-Ge-Ni linkage.
