The aim of this study was to evaluate the effects of the laser radiation (685 nm) associated with photosensitizers on viability of different species of Candida genus. Suspensions of Candida albicans, Candida dubliniensis, Candida krusei and Candida tropicalis, containing 10(6) viable cells per milliliter were obtained with the aid of a Neubauer's chamber. From each species, 10 samples of the cell suspension were irradiated with diode laser (685 nm) with 28 J/cm2 in the presence of methylene blue (0.1 mg/ml), 10 samples were only treated with methylene blue, 10 samples were irradiated with laser in the absence of the dye, 10 samples were treated with the dye and irradiated with laser light and 10 samples were exposed to neither the laser light nor to the methylene blue dye. From each sample, serial dilutions of 10(-2) and 10(-3) were obtained and aliquots of 0.1 ml of each dilution were plated in duplicate on Sabouraud dextrose agar. After incubation at 37 degrees C for 48 h, the number of colony-forming units (CFU/ml) was obtained and data were submitted to ANOVA and Tukey's test (p<0.05). Laser radiation in the presence of methylene blue reduced the number of CFU/ml in 88.6% for C. albicans, 84.8% for C. dubliniensis, 91.6% for C. krusei and 82.3% for C. tropicalis. Despite this, only laser radiation or methylene blue did not reduce significantly the number of CFU/ml of Candida samples, except for C. tropicalis. It could be concluded that the photo activation of methylene blue by the red laser radiation at 685 nm presented fungicide effect on all Candida species studied. 相似文献

A method for light and heavy crude oil digestion using microwave-induced combustion (MIC) in closed vessels is described for further determination of Ag, As, Ba, Bi, Ca, Cd, Cr, Fe, K, Mg, Li, Mn, Mo, Ni, Pb, Rb, Se, Sr, Tl, V, and Zn by inductively coupled plasma mass spectrometry (ICP-MS). Conventional microwave-assisted acid digestion (MW-AD) in pressurized vessels and analyte determination by inductively coupled plasma optical emission spectrometry (ICP OES) were also used for results comparison. For MIC procedure, samples were wrapped in polyethylene films and combusted using 20 bar of oxygen and 50 µl of 6 mol l− 1 ammonium nitrate as aid for ignition. The concentration of nitric acid used as absorbing solution was evaluated (1, 2, 4, 7, 10 and 14 mol l− 1) using an additional reflux step after combustion. Accuracy was evaluated for As, Ba, Ni, Se V, and Zn using certified reference material (CRM) with similar matrix composition and for Cr, Fe, K, Mg, Mn, and Mo by neutron activation analysis (NAA). Recovery tests were also performed for all the analytes by MIC and they were better than 97% using 2 mol l− 1 nitric acid as absorbing solution (with reflux step). Agreement with certified values and NAA results was better than 95%. Using MIC it was possible to obtain lower limits of detection (LODs) by ICP-MS and also by ICP OES in comparison with those obtained by MW-AD. In spite of both sample preparation techniques were apparently suitable for crude oil digestion, MIC was preferable in view of the possibility of using diluted nitric acid as absorbing solution that is an important aspect to minimize interferences by ICP-MS and ICP OES. In order to avoid polyatomic interferences on 52Cr and 56Fe determinations by ICP-MS, a dynamic reaction cell with ammonia gas was used. Residual carbon content in digests obtained by MW-AD and MIC was 15% and < 1%, respectively. Using MIC the simultaneous digestion of 8 samples was possible in less than 30 min. 相似文献

Skin exposure to UVB radiation has been reported to produce both a significant inflammatory response and marked immunosuppression. This work was aimed to evaluate whether the response of murine skin to an acute UVB dose was modified by pre-exposure to chronic UVB irradiation and by topical treatment with naproxen, a nonsteroidal anti-inflammatory drug. Moreover, the effect of naproxen on the incidence of UV-induced skin tumors was studied. Prostaglandin E2 (PGE2 ) and tumor necrosis factor alpha (TNF-α) levels were increased 96 h post-UVB in acutely irradiated animals and both mediators were modified by topical naproxen application—PGE2 was decreased while TNF-α was increased. Such inflammatory response was suppressed when mice were chronically irradiated. Naproxen application on chronically irradiated mice reduced the incidence of tumor lesions. Taken together, our data suggest that chronic UVB irradiation generates an immunosuppressive state that prevents skin cells from responding normally to an acute irradiation challenge, thus impairing the protective effect of TNF-α against skin tumor development. Furthermore, reduction in the incidence of tumor lesions by naproxen may be due to its ability to increase TNF-α levels as well as to decrease PGE2 . 相似文献

Eggs of Psorophora albigenu were analyzed by light and scanning electron microscopy. Eggs were laid singly and were black and elliptical in outline. A length of approximately 586.4 ± 10.83 μm, a width of 172.3 ± 8.09 μm (n=30), and an egg index (length/width ratio) of 3.33 were observed. The anterior extremity tapered abruptly from a width of 64.9 μm, while such tapering was more gradual at the posterior extremity from a width of 77.4 μm. The ventral surface of the chorionic coating presented cells with tubular aspect containing tubercles in rows with a density of 9-16 tubercles per cell. These tubercles presented two different sizes, with the largest measuring 7.87 ± 01.58 μm and the smallest measuring 0.71 ± 0.53 μm (n=30) in longitudinal diameter. The micropyle orifice was very evident, with a diameter of 3.36 μm. The morphometric characteristics of Ps. albigenu obtained in this study were distinct from those of other species of the Psorophora genus, including Psorophora varipes which has been considered for a long time to be synonymous with Ps. albigenu. 相似文献

Theoretical calculations at the M05‐2X/6‐31+G(d) level of theory have been carried out in order to explore the nature of the mechanism of the thermal decomposition reactions of the β‐hydroxy ketones, 4‐hydroxy‐2‐butanone, 4‐hydroxy‐2‐pentanone, and 4‐hydroxy‐2‐methyl‐2‐pentanone in gas phase and in m‐xylene solution. The mechanism proposed is a one‐step process proceeding through a six‐membered cyclic transition state. A reasonable agreement between experimental and calculated activation parameters and rate constants has been obtained, the tertiary : secondary : primary alcohol rate constant ratio being calculated, at T = 503.15 K, as 5.9:4.7:1.0 in m‐xylene solution and 44.1:5.0:1.0 in the gas phase, compared with the experimental values, 3.7:1.3:1.0 and 13.5:3.2:1.0, respectively. The progress of the thermal decomposition reactions of β‐hydroxy ketones has been followed by means of the Wiberg bond indices. The lengthening of the O1–C2 bond with the initial migration of the H6 atom from O5 to O1 can be seen as the driving force for the studied reactions. Calculated synchronicity values indicate that the mechanisms correspond to concerted and highly synchronous processes. Simulations of polymerization rate, molecular weight development and evolution of the concentrations of species participating in the reaction mechanism over a range of operating conditions, and a parameter sensitivity analysis showing the effects of temperature, activation/deactivation equilibrium constant and initial concentrations of controller and initiator (if present) on these variables are presented for the nitroxide‐mediated radical polymerization of styrene. The simulations were performed with a computer program based on a detailed reaction mechanism. The simulated profiles of conversion, number average molecular weight ( ), and polydispersity agree well with experimental data. Previously unknown activation energies for reactions involved in the mechanism are estimated. The temperature dependence of the kinetic rate constants obtained in this study will be useful for future modeling and optimization studies.

Two series of segmented polyurethanes were prepared with systematic variation in soft and hard segment length. The soft segment was constituted by polycaprolactone (PCL) blocks of molecular masses 530 or 2000 and the hard segment (HS) by urethane blocks, in a concentration that varied from 12% to 44% in weight of the whole polyurethane. Morphological analyses indicated that the amount of crystallinity of copolymers was strongly dependent on the PCL molar mass and hard segment content. The copolymers with longer PCL soft segments (Mn=2000) were semicrystalline, but those with shorter PCL segment (Mn=530) were unable to crystallize. The primary factor affectingthe biodegradability of copolymers as evaluated by Sturm tests was the extent of the phase separation, and that the segmental blending of the less biodegradable polyurethane (HS) blocks with PCL in the amorphous phase had a critical unfavorable consequence, which may be attributed to the size of the accessible area by microorganisms. 相似文献
