Capillary zone electrophoresis with field enhanced sample stacking as a tool for targeted metabolome analysis of adenine nucleotides and coenzymes in Paracoccus denitrificans

The main aim of this work was to demonstrate the applicability of capillary zone electrophoresis in combination with field enhanced sample stacking in targeted metabolome analyses of adenine nucleotides – AMP, ADP, ATP, coenzymes NAD⁺, NADP⁺ and their reduced forms in Paracoccus denitrificans. Sodium carbonate/hydrogencarbonate buffer (100 mM, pH 9.6) with the addition of β‐CD at a concentration of 10 mM was found to be an effective BGE for their separation within 20 min. Besides this, special attention was paid to the development of the procedure for the extraction of specific metabolites from the bacterium P. denitrificans. This procedure was not only optimised to achieve the highest metabolite yields but also to obtain a sample that was fully compatible with the online preconcetration strategy used. The developed methodology was finally applied in a study of the bacterium P. denitrificans at various stages of the active respiratory chain.     

Die Hohlkugel bei einem nichtlinearen Elastizitätsgesetz

Zusammenfassung Mit einem allgemeinen nichtlinearen. Elastizitätsgeseta für kleine Verzerrungen werden die Spannungen in einer dickwandigen Hohlkugel berechnet. In allen Zahlenbeispielen zeigt es sich, daß die infolge der Nichtlinearität des Elastizitätsgesetzes errechnete Spannungsverteilung erhebliche Abweichungen gegenüber den Werten der linearen Theorie ergibt, und zwar insbesondere für die Randwerte. Die Ergebnisse legen somit die Richtigkeit der Vermutung nahe, wonach bereits kleine, bei den meisten Werkstoffen vorkommende Abweichungen vom Hookeschen Gesetz einen starken Abbau der Spannungsspitzen zur Folge haben.Zum Schluß möchte ich Herrn Prof. Dr. R. Evaluation of Antiproliferative Palladium(II) Complexes of Synthetic Bisdemethoxycurcumin towards In Vitro Cytotoxicity and Molecular Docking on DNA Sequence

Metallodrugs form a large family of therapeutic agents against cancer, among which is cisplatin, a paradigmatic member. Therapeutic resistance and undesired side effects to Pt(II) related drugs, prompts research on different metal–ligand combinations with potentially enhanced biological activity. We present the synthesis and biological tests of novel palladium(II) complexes containing bisdemethoxycurcumin (BDMC) 1 and 2. Complexes were fully characterized and their structures were determined by X-ray diffraction. Their biological activity was assessed for several selected human tumor cell lines: Jurkat (human leukaemic T-cell lymphoma), HCT-116 (human colorectal carcinoma), HeLa (human cervix epitheloid carcinoma), MCF-7 (human breast adenocarcinoma), MDA-MB-231 (human mammary gland adenocarcinoma), A549 (human alveolar adenocarcinoma), Caco-2 (human colorectal carcinoma), and for non-cancerous 3T3 cells (murine fibroblasts). The cytotoxicity of 1 is comparable to that of cisplatin, and superior to that of 2 in all cell lines. It is a correlation between IC₅₀ values of 1 and 2 in the eight studied cell types, promising a potential use as anti-proliferative drugs. Moreover, for Jurkat cell line, complexes 1 and 2, show an enhanced activity. DFT and docking calculations on the NF-κB protein, Human Serum Albumin (HSA), and DNA were performed for 1 and 2 to correlate with their biological activities.     

Methyl 5-Deoxy-α and β-D-Xylofuranosides

Abstract

Synthesized from D-xylose, methyl 5-deoxy-α-D-xylofuranoside (1) and methyl 5-deoxy-β-D-xylofuranoside (2) were obtained in overall yields of 24 and 26 %, respectively. The key step in the synthesis was the separation of an anomeric mixture on a strong anion exchanger in OH^? form. NMR data and mass spectra of title compounds 1, 2, methyl 2,3-di-O-acetyl-5-deoxy-α-D-xylofuranoside (3), and methyl 2,3-di-O-acetyl-5-deoxy-β-D-xylofuranoside (4) are discussed. The conformations of 1 and 2 were established from the best fit between calculated and experimental coupling constants using Karplus equation.

o-Methoxycarbonyl-phenyl isocyanate reacts with mono-aminoalkyl orthophosphoric and sulfuric acids to yield the corresponding 1,2,3,4-tetrahydroquinazoline-2,4-dione-3-ethyl phosphoric (or sulfuric) monoesters which are isolated as sodium salts. 1,2,3,4-Tetrahydroquinazoline-2,4-dione-3-ethyl phosphoric acid has been isolated by adding hydrochloric acid to an aquous solution of its sodium salt.     

Dynamic Jahn-teller effect in a Fe-N6 cage molecule

Mössbauer spectroscopic studies of [Fe(di(amH)-sar)](NO₃)_4·H₂O where di(amH)-sar represents 1,8 diamino 3,6,10,13,16,19-hexaza bicyclo [6,6,6] icosane in the temperature range of 4.2 to 300 K suggest that it undergoes a dynamic Jahn-Teller effect as revealed from the plots of temperature dependent quadrupole coupling constant, chemical shift and line width values. The spectrum down to 4.2 K, shows a quadrupole doublet with no magnetic hyperfine splitting.

Aminoalkyl sulfuric monoesters with primary or secondary amino groups – with the exception of colaminesulfuric monoester – react with isothiocyanates in the presence of one equivalent of a base to yield the corresponding hydrosoluble salts of [N-aryl(aralkyl or alkyl)thiocarbamoyl]-aminoalkyl sulfates.
