The 1,3-dipolar cycloaddition of the C-t-butyloxycarbonyl-N -mannosyl-nitrone 5 , formed in situ from the partially protected D -mannose-oxime 3 and the glyoxylate 4 , to ethylene gave preferentially the (3S)-N-glycosyl-isoxazolidine 6 which was transformed into the 3-isoxazolidine-carboxylate (L -5-oxaproline ester) 12 and into some derivatives thereof. The (S)-configuration of 12 was proved by chemical correlation with a derivative of L -asparagine. The D -5-oxaproline ester was obtained from the corresponding N-ribosyl-nitrone 24 . Two protected dipeptides containing either C-terminal- ( 28 ) or N-terminal-5-oxaproline (= Opro) ( 30 ) were synthesized. Starting from 12 , the analogue 1 of captopril® ( 2 ) was prepared and its activity as an inhibitor of the angiotensin-converting-enzyme (ACE) was examined.  相似文献

Wastewater treatment plants dealing with industrial wastes are often susceptible to overload of toxic influent that can partially or completely destroy treatment for extended periods. An obvious candidate for monitoring toxicity in such wastewater systems is bioluminescent bacteria. However, the natural bioluminescent bacteria can be particularly sensitive to some industrial wastes and therefore their response to normal operational conditions does not reflect the status of the microbial community responsible for treatment. Moreover, the salt dependence of the marine bioluminescent bacteria, and the temperature sensitivity of some strains, further complicate their use. Here we describe the construction of whole cell genetically modified bioluminescent biosensors and their immobilisation for use in monitoring the toxicity of a complex industrial wastewater containing phenolic materials. A hand-held luminometer was designed for laboratory or field use, and the immobilisation system designed with several things in mind: the geometry of the instrument; the need for containment of GM bacteria; the maximisation of the bioavailability of the wastewater to the biosensor. The performance of a candidate GM sensor was compared with Vibrio fischeri in liquid culture and after immobilisation in thin films of poly(vinyl alcohol) (PVA) cryogels. The biosensors were tested against pure phenol and 3-chlorophenol as a reference toxic chemical known to be much more toxic to bacteria than phenol. The biosensors were then tested with the phenolics-containing industrial wastewater. The immobilisation system proved to operate predictably with pure toxicants, and was able to discriminate toxicity of various zones within the wastewater treatment plant.  相似文献

Development of new generations of membranes with high degrees of permeabilities and controllable mass transport properties requires a fundamental understanding of the relationship between molecular structures and permeabilities. Initiation of interdisciplinary research in biology, biophysics, polymer and colloid chemistry is proposed to provide the insight to membrane transport processes at the molecular level. Mother nature's most talented transporter — the biological membrane — should inspire this endeavor. Following a survey of the properties of, and recognized transport mechanisms in, biomembranes, membrane-mimetic chemistry is introduced to serve as a bridge between biological and polymeric membranes. Surfactant aggregates — micelles, monolayers, organized multilayers (Langmuir—Blodgett films), bilayer lipid membranes (BLMs), vesicles and polymerized vesicles — are shown to be the media in membrane-mimetic chemistry. Properties of these organized surfactant assemblies are summarized. Emphasis is placed on the control of molecular transport in membrane-mimetic systems. Perspectives and prospectives of biomimetic membranology are discussed.  相似文献

The structure of an ordered, ultrathin theta-Al(2)O(3) film grown on a NiAl(100) single-crystal surface was studied by Auger electron spectroscopy (AES), X-ray photoelectron spectroscopy (XPS), and low-energy electron diffraction (LEED), and its interaction with water was investigated with temperature programmed desorption (TPD) and XPS. Our results indicate that H(2)O adsorption on the theta-Al(2)O(3)/NiAl(100) surface is predominantly molecular rather than dissociative. For theta(H)()2(O) < 1 ML (ML = monolayer), H(2)O molecules were found to populate Al(3+) cation sites to form isolated H(2)O species aligned in a row along the cation sites on the oxide surface with a repulsive interaction between them. For theta(H)()2(O) > 1 ML, three-dimensional ice multilayers were observed to form, which then desorb during TPD with approximate zero-order kinetics as expected. A small extent of H(2)O dissociation was observed to occur on the theta-Al(2)O(3)/NiAl(100) surface, which was attributed to the presence of a low concentration of oxygen atom vacancies. Titration of these defect sites with adsorbed H(2)O molecules revealed an estimated defect density of 0.05 ML for the theta-Al(2)O(3)/NiAl(100) system consistent with the ordered nature of the synthesized oxide film.  相似文献

PEGylated Nb2O5 surfaces were obtained by the adsorption of poly(L-lysine)-g-poly(ethylene glycol) (PLL-g-PEG) copolymers, allowing control of the PEG surface density, as well as the surface charge. PEG (MW 2 kDa) surface densities between 0 and 0.5 nm(-2) were obtained by changing the PEG to lysine-mer ratio in the PLL-g-PEG polymer, resulting in net positive, negative and neutral surfaces. Colloid probe atomic force microscopy (AFM) was used to characterize the interfacial forces associated with the different surfaces. The AFM force analysis revealed interplay between electrical double layer and steric interactions, thus providing information on the surface charge and on the PEG layer thickness as a function of copolymer architecture. Adsorption of the model proteins lysozyme, alpha-lactalbumin, and myoglobin onto the various PEGylated surfaces was performed to investigate the effect of protein charge. In addition, adsorption experiments were performed over a range of ionic strengths, to study the role of electrostatic forces between surface charges and proteins acting through the PEG layer. The adsorbed mass of protein, measured by optical waveguide lightmode spectroscopy (OWLS), was shown to depend on a combination of surface charge, protein charge, PEG thickness, and grafting density. At high grafting density and high ionic strength, the steric barrier properties of PEG determine the net interfacial force. At low ionic strength, however, the electrical double layer thickness exceeds the thickness of the PEG layer, and surface charges "shining through" the PEG layer contribute to protein interactions with PLL-g-PEG coated surfaces. The combination of AFM surface force measurements and protein adsorption experiments provides insights into the interfacial forces associated with various PEGylated surfaces and the mechanisms of protein resistance.  相似文献

3-Alkyl-1-benzoxepin-5-one derivatives and 2-alkyl-1,4-naphtoquinones from 2-acylaryl propargyl ethers. It was found that 3-alkyl-1-benzoxepin-5(2H)-ones of type B can be synthesized by treating 2-acylaryl propargyl ethers of type A with sodium methylsulfinyl methide (NaMSM, dimesyl sodium) (Scheme 13). Oxepinone derivatives of type B undergo ring contraction with base (also NaMSM) to yield the quinol derivatives C which, oxidize (during work-up), if R2 = H, to the 1,4-naphthoquinones D (Scheme 13). The propargyl ethers used are listed in Scheme 1. The naphthalene derivatives 1 and 3 give oxepinones (E- 9 and a mixture of 14/15 respectively), whereas the expected oxepinone from 2 is transformed directly into the quinone 11 (Scheme 2, 3 and 5). Isomerizations of 2-acetylphenyl propargyl ethers ( 4, 5 and 6 ) (Schemes 6, 7 and 8) are less successful because of side reactions. If however the acetyl group is replaced by a propionyl or substituted propionyl group (as in ethers 7 and 8 ) oxepinones are obtained again in good yield (Scheme 9). The mechanistic pathway for the transformation of naphthyl propargyl ethers (and phenyl derivatives) under influence of NaMSM is shown in Scheme 10. The base-catalysed conversion of 4-phenyl-l-benzoxepin-5(2H)-one,benzo[f]furo[2,3-c](10 H)-oxepin-4-oncsand 3-methoxy-G,11- dihydro-dibenzo[b, e]loxepin-11-oneinto thc corresponding quinones has been reported [13] [20] [21]. The conversion of 2-acylaryl propargyl others via the isolable benzoxepin-5-one derivativcs or directly into the specifically substituted 1,4-naphthoquinone derivatives is of synthetic interest.  相似文献

A new program has recently been implemented with the aim of extending quasi-particle (QP) band structure calculations to polymers with larger unit cells. The theoretical background is briefly reviewed and the new algorithm described, which has been optimized for machines with vector processors. To illustrate the usefulness of this technique, calculations have been performed on polyparaphenylene (PPP) using a double-zeta basis set. The calculated QP band gap between the valence and conduction bands is 2.3 eV, which compares favorably with the experimental value of 2.8 eV. The self-consistent field (SCF) result with the same double-zeta basis set is 8.7 eV. © 1994 by John Wiley & Sons, Inc.  相似文献

A stochastic variational inequality is proposed to model a white noise excited elasto-plastic oscillator. The solution of this inequality is essentially a continuous diffusion process for which a governing diffusion equation is obtained to study the evolution in time of its probability distribution. The diffusion equation is degenerate, but using the fact that the degeneracy occurs on a bounded region we are able to show the existence of a unique solution satisfying the desired properties. We prove the ergodic properties of the process and characterize the invariant measure. Our approach relies on extending Khasminskii’s method (Stochastic Stability of Differential Equations, Sijthoff and Noordhoff, 1980), which in the present context leads to the study of degenerate Dirichlet problems with nonlocal boundary conditions.
