Production and properties of 3-cyanopyridine hydratase inRhodococcus equi SHB-121

A strain ofRhodococcus equi SHB-121 forming 3-cyanopyridine hydratase was screened from nitrile-polluted soil. The optimum conditions for the formation of 3-cyanopyridine hydratase by the strain SHB-121 have been studied. Under the optimum conditions, the specific activity of the enzyme reached 5.32 U/mg of dry cell, 95 times higher than that cultured in screening medium. In addition, the activity of coexistent amidase was very low. 3-Cyanopyridine hydratase was purified from methylacrylamide-induced cells ofRh. equi SHB-121 by procedures including ultrasonic oscillation, ammonium sulfate precipitation, and column chromatographies on DEAE-cellulose DE52, hydroxyapatite, and Sephadex G-25. The overall purification was 31-fold. The molecular weight of the enzyme was about 30 kDA by SDS-PAGE. The pI value was 4.1. The transition temperature and pH were 7.0°C and 6.0, respectively, resulting from the differential spectra. The optimum pH and temperature for the enzyme reaction were 8.0 and 30°C. The enzyme activity was strongly inhibited by Ag⁺, Hg²⁺, Cu²⁺, and NH₄ ⁺, whereas it was enhanced by Fe³⁺ slightly. The enzyme catalyzed the hydration of 3-cyanopyridine to nicotinamide, and itsKm value was 0.1 mol/L. Uncompetitive inhibitor sodium cyanide has a K, value of 5 mmol/L.

Determination of enterostatin in human cerebrospinal fluid by capillary electrophoresis with laser induced fluorescence detection

A capillary electrophoresis (CE) method with laser induced fluorescence (LIF) detection is described for quantification of enterostatin (Val-Pro-Asp-Pro-Arg), a pentapeptide involved in appetite regulation and insulin secretion. Enterostatin and two other pentapeptides belonging to the enterostatin family (i.e. Ala-Pro-Gly-Pro-Arg and Val-Pro-Gly-Pro-Arg) were well separated from each other. The peptides were fluorescently tagged with naphthalene-2,3- dicarboxaldehyde (NDA) and separated by micellar electrokinetic chromatography (MEKC) in the presence of methanol as an organic modifier. Coupled with LIF detection, the method had a detection limit of 4.8 x 10(-6) M for enterostatin. The relative standard deviation was to be 4.0% from five determinations of enterostatin at 37.2 microM in a human cerebrospinal fluid (CSF) sample. Twenty-three human CSF samples were analyzed. The level of enterostatin ranged from 24 microM to 51 microM with a mean (+/- SEM) value of 41.7 +/- 2.0 microM.     

Noble gas-transition metal complexes: coordination of ScO+ by multiple Ar, Kr, and Xe atoms in noble gas matrixes

The combination of matrix isolation infrared spectroscopic and density functional calculation results provides strong evidence that the transition metal monoxide cation, ScO+, coordinates five noble gas atoms in forming the [ScO(Ng)5]+ (Ng = Ar, Kr, or Xe) complexes in noble gas matrixes.

Macrocyclic polycther sulfide syntheses. The preparation of thia-crown-5, 6 and 7 compounds

Macroeyclic polyether sulfides have been prepared by reacting an oligoethylene glycol dichloride with a dimercaptan or sodium sulfide as reported in a previous paper (6). The following new compounds were prepared: 1,4,7-trithia(15-crown-5) (1); 1,4,10-trithia(15-crown-5) (II): 1,4,7,10-tetrathia(15-crown-5) (III); I-thia(18-crown-6) (IV); 1,4-dithia(18-crown-6) (V); 1,7-dithia(18-crown-6) (VI); 1,4,7-trithia(18-crown-6) (VII); 1,7-dithia(21-crown-7) (VIII); and 1,4,7-trithia(21-crown-7) (IX). The melting points of these and previously reported thia-crown compounds correlate with their structures. X-ray analyses of two thia-crown compounds indicate that the large sulfur atoms are directed away from the center of the ring.     

氯丙嗪在离子液体BMIMPF6修饰玻碳电极上的伏安行为

对氯丙嗪(CPZ)在离子液体BMIMPF6修饰玻碳电极(BMIMPF6/GC)上的伏安行为进行了研究.发现CPZ在该修饰电极上于065V左右产生氧化还原峰,峰的可逆性比玻碳电极(GC)有较大改进,其电极过程为吸附控制.在0050 mol/L磷酸缓冲溶液(pH 4)中,CPZ在BMIMPF6/GC上的氧化峰的峰电流约为GC上的2倍.在优化实验条件的基础上,采用微分脉冲伏安法对不同浓度CPZ溶液进行了测定.结果表明,CPZ在BMIMPF6/GC上的氧化峰电流与浓度在56×10-9～30 ×10-5mol/L范围内有线性关系.将此方法用于尿样的测定,其加入回收率为97％左右.用3种电极测定了CPZ的表观扩散系数,其大小为D(BMIMPF6/GC)＞DOMIMBF6/GC)＞D(GC),这与电极表面积变化及膜内扩散有一定关系.     

QuEChERS-气相色谱-串联质谱法测定枸杞中农药残留

建立了QuEChERS-气相色谱-串联质谱法测定枸杞中农药残留的分析方法。样品经乙腈提取，采用乙二胺-N-丙基硅烷（PSA）、C18和石墨化炭黑（GCB）3种吸附剂净化，在多反应监测模式（MRM）下进行检测，基质外标法定量。结果显示，检测项目在5~500 μg/L范围内呈良好线性（R²>0.99），在3个添加水平下测得回收率为71.5%~109.7%，相对标准偏差（RSD）为1.94%~9.87%（n=6）。该方法灵敏度高、净化效果好，可同时快速检测有机磷类、有机氯类农药。     

计算单向复合材料弹性模量的新公式

根据复合材料单层的实际构造，提出了串并联组合模型，采用微观力学与宏观力学相结合的方法，推导出一组计算单向复合材料弹性模量的新公式。其优点是：Ｅ１和ｖ（１２）、Ｅ２和Ｇ（１２）的计算式形式对称，结构简单，计算容易，物理意义明确。我们把由世界上许多著名学者提出的多组预测公式和本文导出式计算所得的结果与国内外已发表的硼／环氧、碳／环氧、玻璃／环氧复合材料的实验数据相比较，结果表明：导出式比其他公式精确、更有价值，可供二程设计使用。
