Methacrylanilides, ArN(CH3)COC(CH2LG)=CH2, with allylic leaving groups (LG(-) = BocAla, PhCO2(-), PhCH2CO2(-), PhO(-)) undergo photochemical electrocyclic ring closure to produce a zwitterionic intermediate. Further reaction of the intermediate results in expulsion of the leaving group to give an alpha-methylene lactam as the major product. In addition, a lactam product that retains the leaving group is formed via a 1,5-H shift in the intermediate. Elimination of the leaving group is generally preferred, even for LG(-) = PhO(-), although in benzene as the solvent the lactam retaining the phenolate group becomes the sole photoproduct. The electrocyclic ring closure occurs in the singlet excited-state for the para-COPh-substituted anilide derivative and is not quenched by 0.15 M piperylene or 0.01 M sodium 2-naphthalenesulfonate (2-NPS) as triplet quenchers. Comparable concentrations of 2-NPS strongly quench the transient absorption of the triplet excited state observed at 450-700 nm according to laser flash photolysis experiments. In aqueous media, quantum yields for total products are insensitive to leaving group ability, and Phi(tot)(para-CO2CH3) = 0.04-0.06 at 310 nm and Phi(tot)(para-COPh) = 0.08-0.1 at 365 nm, for which Phi(isc) = 0.15.  相似文献

Gas-phase infrared photodissociation spectroscopy is reported for the microsolvated [Mn(ClO4)(H2O) n ]+ and [Mn2(ClO4)3(H2O) n ]+ complexes from n = 2 to 5. Electrosprayed ions are isolated in an ion-trap where they are photodissociated. The 2600–3800 cm−1 spectral region associated with the OH stretching mode is scanned with a relatively low-power infrared table-top laser, which is used in combination with a CO2 laser to enhance the photofragmentation yield of these strongly bound ions. Hydrogen bonding is evidenced by a relatively broad band red-shifted from the free OH region. Band assignment based on quantum chemical calculations suggest that there is formation of water—perchlorate hydrogen bond within the first coordination shell of high-spin Mn(II). Although the observed spectral features are also compatible with the formation of structures with double-acceptor water in the second shell, these structures are found relatively high in energy compared with structures with all water directly bound to manganese. Using the highly intense IR beam of the free electron laser CLIO in the 800–1700 cm−1, we were also able to characterize the coordination mode (η2) of perchlorate for two clusters. The comparison of experimental and calculated spectra suggests that the perchlorate Cl—O stretches are unexpectedly underestimated at the B3LYP level, while they are correctly described at the MP2 level allowing for spectral assignment.  相似文献

We discuss families of meromorphic functions f h obtained from single functions f by the re-scaling process f h (z) = h −α f (h + h −β z), generalising Yosida’s process f h (z) = f (h + z). The main objective is to obtain information about the value distribution of the generating functions f themselves. Among the most prominent (generalised) Yosida functions are the elliptic functions and also some first, second and fourth Painlevé transcendents. Laulimalide and peloruside A are microtubule‐stabilizing agents (MSAs), the mechanism of action on microtubules of which is poorly defined. Here, using X‐ray crystallography it is shown that laulimalide and peloruside A bind to a unique non‐taxane site on β‐tubulin and use their respective macrolide core structures to interact with a second tubulin dimer across protofilaments. At the same time, they allosterically stabilize the taxane‐site M‐loop that establishes lateral tubulin contacts in microtubules. Structures of ternary complexes of tubulin with laulimalide/peloruside A and epothilone A are also solved, and a crosstalk between the laulimalide/peloruside and taxane sites via the M‐loop of β‐tubulin is found. Together, the data define the mechanism of action of laulimalide and peloruside A on tubulin and microtubules. The data further provide a structural framework for understanding the synergy observed between two classes of MSAs in tubulin assembly and the inhibition of cancer cell growth.  相似文献

Owing to their ability to be genetically expressed in live cells, fluorescent proteins have become indispensable markers in cellular and biochemical studies. These proteins can undergo a number of covalent chemical modifications that may affect their photophysical properties. Among other mechanisms, such covalent modifications may be induced by reactive oxygen species (ROS), as generated along a variety of biological pathways or through the action of ionizing radiations. In a previous report [1], we showed that the exposure of cyan fluorescent protein (ECFP) to amounts of ?OH that mimic the conditions of intracellular oxidative bursts (associated with intense ROS production) leads to observable changes in its photophysical properties in the absence of any direct oxidation of the ECFP chromophore. In the present work, we analyzed the associated structural modifications of the protein in depth. Following the quantified production of ?OH, we devised a complete analytical workflow based on chromatography and mass spectrometry that allowed us to fully characterize the oxidation events. While methionine, tyrosine, and phenylalanine were the only amino acids that were found to be oxidized, semi-quantitative assessment of their oxidation levels showed that the protein is preferentially oxidized at eight residue positions. To account for the preferred oxidation of a few, poorly accessible methionine residues, we propose a multi-step reaction pathway supported by data from pulsed radiolysis experiments. The described experimental workflow is widely generalizable to other fluorescent proteins, and opens the door to the identification of crucial covalent modifications that affect their photophysics.
