Development of a 5-exo, 6-exo tandem radical cyclization to access bisabosqual A

Substrates were designed to undergo tandem radical cyclizations to afford the cis, cis-fused hexahydrobenzofurobenzopyran ring system of the bisabosquals. Studies with these revealed that different initiating systems gave different yields and somewhat different diastereomeric ratios. A photocatalytic cyclization was also successful with the bisabosqual substrate.     

Palladium-Catalyzed Synthesis of 2,3,4-Trisubstituted Furans via Cascade Reactions of Aryloxy-enynes with Aryl Halides

A highly efficient palladium-catalyzed cascade reactions of aryloxy-enynes with aryl halides under mild reaction conditions has been developed. This methodology offers rapid access to 2,3,4-trisubstituted furans in good to excellent yields in a regioselective manner.     

Ammosamide D, an oxidatively ring opened ammosamide analog from a marine-derived Streptomyces variabilis

Ammosamide D (1), an oxidized analog of the ammosamide family, was isolated from a marine-derived Streptomyces variabilis. Pyrroloquinoline containing alkaloids are a growing class of natural products, with 1 being the first example of an oxidized analog resulting in a 5,6-dioxo-5,6-dihydroquinoline ring system. Attempts at chemical conversion of ammosamide B to ammosamide D revealed that a strong chemical oxidant is required. Ammosamide D has modest cytotoxicity to the MIA PaCa-2 pancreatic cancer cell line.     

Pd/Cu-catalyzed cascade Sonogashira coupling/cyclization reactions to highly substituted 3-formyl furans

An efficient palladium/copper-catalyzed approach to the synthesis of highly substituted 3-formyl furans from the reactions of readily available α-bromoenaminones with terminal alkynes has been developed. This methodology was realized by the cascade reactions of Sonogashira coupling and the subsequent intramolecular cyclization.

Divergent Synthesis of Monosubstituted and Unsymmetrical 3,6-Disubstituted Tetrazines from Carboxylic Ester Precursors

Since tetrazines are important tools to the field of bioorthogonal chemistry, there is a need for new approaches to synthesize unsymmetrical and 3-monosubstituted tetrazines. Described here is a general, one-pot method for converting (3-methyloxetan-3-yl)methyl carboxylic esters into 3-thiomethyltetrazines. These versatile intermediates were applied to the synthesis of unsymmetrical tetrazines through Pd-catalyzed cross-coupling and in the first catalytic thioether reduction to access monosubstituted tetrazines. This method enables the development of new tetrazine compounds possessing a favorable combination of kinetics, small size, and hydrophilicity. It was applied to a broad range of aliphatic and aromatic ester precursors and to the synthesis of heterocycles including BODIPY fluorophores and biotin. In addition, a series of tetrazine probes for monoacylglycerol lipase (MAGL) were synthesized and the most reactive one was applied to the labeling of endogenous MAGL in live cells.     

An automated dynamic viscometer with air bearing and inductive transducers for angular displacement detection

A dynamic viscometer is described, with which the dynamic moduli in the frequency range from 2 × 10^–3 to 10 Hz can be determined for liquids with 10^–3 Pa< |G ^* | <10² Pa. Due to the application of an air bearing and inductive transducers for the detection of the angular displacement of both the drive and the measuring cylinder a sensitive apparatus has been made. Very small strains (₀ 10^–3) can be applied and only a small amount of sample (4 ml) is needed. The operation of the apparatus is fully computer-controlled, thus, long runs at various frequencies and temperatures are possible without operator intervention. The theoretical background, calibration procedure, and operation window are described. A presentation of some measurements on two polyisoprene/polystyrene triblock copolymer solutions concludes the work.
