The dynamics of RNA contributes to its biological functions such as ligand recognition and catalysis. Using quasielastic neutron scattering spectroscopy, we show that Mg(2+) greatly increases the picosecond to nanosecond dynamics of hydrated tRNA while stabilizing its folded structure. Analyses of the atomic mean-squared displacement, relaxation time, persistence length, and fraction of mobile atoms showed that unfolded tRNA is more rigid than folded tRNA. This same result was found for a sulfonated polystyrene, indicating that the increased dynamics in Mg(2+) arises from improved charge screening of the polyelectrolyte rather than specific interactions with the folded tRNA. These results are opposite to the relationship between structural compactness and internal dynamics for proteins in which the folded state is more rigid than the denatured state. We conclude that RNA dynamics are strongly influenced by the electrostatic environment, in addition to the motions of local waters.  相似文献

Based on the hits, 3,4-dihydroquinazoline-2-thione (1) and benzimidazole-2-thione (2), a series of indole-2-thione (3) and indole-2-thiol inhibitors (4) of melanogenesis were designed, synthesized and evaluated in melanoma B16 cells under the stimulant of α-melanocyte stimulating hormone (α-MSH). The indole-2-thione compounds (3a-g) exhibited an effective inhibitory activity on melanin synthesis. The Structure-Activity Relationship (SAR) studies of 2 have revealed that in potent inhibitor 3a (>100% inhibition at 30 μM, IC50=1.40 μM) the role of nitrogen (3-N) at 3-position is insignificance. In addition, the hydrophobic substituents of 3 were better than the hydrophilic one. However, conversion of thione (-C=S, 3) to thiol (-C-SH, 4) led to decrease in the potency.  相似文献

In this study we investigated the effects of graphite flake alignment on thermal emissivity by applying a magnetic field during coating of aluminum sheets with graphite. The coating paste was prepared by ball milling graphite flakes with an organic binder. The graphite flake content was 9.1, 13.0, 16.7, 20.0, or 23.1 wt.%. After coating of aluminum sheets with the paste by dipping, a magnetic field was applied vertically to the coated aluminum sheet by use of neodymium magnets. It was observed that the graphite flakes were aligned at an angle to the surface by application of the magnetic field. In contrast, in the absence of the magnetic field the graphite flakes were aligned horizontally on the aluminum sheets. The surface roughness of specimens prepared by use of a magnetic field (MF; R a = 10.172–14.654 μm) was more than twofold that of specimens for which no magnetic field was applied (NMF; R a = 4.564 μm). The thermal emissivity of MF9 (9.1 wt.% graphite; ε = 0.80) was higher than that of NMF9 (9.1 wt.% graphite; ε = 0.77). The thermal emissivity of MF20 (20.0 wt.% graphite) was 0.91, the highest in this study. It was shown that flakes aligned at an angle to the surface contribute to enhanced thermal emissivity. Well aligned graphite flakes are therefore expected to enable high thermal dissipation from electronic components.  相似文献

Metagenomic DNA (gDNA) is an important research topic because uncultivated microorganisms represent an interesting reservoir of genes with potential biotechnological applications. Here we describe a novel Si-C-N chromatography approach of gDNA extraction from environmental samples. Amorphous Si-C-N ceramic was obtained by the pyrolysis of a polymer precursor (polycarbosilazane) at 1350 degrees C in Ar. The purified gDNA fragments were at least 12 kilo base pairs in size and were sufficiently free of contaminants, thus were applicable to both restriction enzyme digestion using five different enzymes and polymerase chain reaction amplification for detecting phylogenetic groups of native microorganisms in environmental samples. This Si-C-N material produces pure gDNA that can be utilized in some of the most common molecular biological procedures applied in the purification step.  相似文献

Herein, we prepared 1,3-dipalmitoyl-2-oleoyl glycerol (POP)-rich fats with reduced levels of diacylglycerols (DAGs), adversely affecting the tempering of chocolate, via two-step hexane fractionation of palm stearin. DAG content in the as-prepared fats was lower than that in POP-rich fats obtained by previously reported conventional two-step acetone fractionation. Cocoa butter equivalents (CBEs) were fabricated by blending the as-prepared fats with 1,3-distearoyl-2-oleoyl glycerol (SOS)-rich fats obtained by hexane fractionation of degummed shea butter. POP-rich fats achieved under the best conditions for the fractionation of palm stearin had a significantly lower DAG content (1.6 w/w%) than that in the counterpart (4.6 w/w%) prepared by the previously reported method. The CBEs fabricated by blending the POP- and SOS-rich fats in a weight ratio of 40:60 contained 63.7 w/w% total symmetric monounsaturated triacylglycerols, including 22.0 w/w% POP, 8.6 w/w% palmitoyl-2-oleoyl-3-stearoyl-rac-glycerol, 33.1 w/w% SOS, and 1.3 w/w% DAGs, which was not substantially different from the DAG content in cocoa butter (1.1 w/w%). Based on the solid-fat content results, it was concluded that, when these CBEs were used for chocolate manufacture, they blended with cocoa butter at levels up to 40 w/w%, without distinctively altering the hardness and melting behavior of cocoa butter.  相似文献

The alkylation of 1-substituted 1H-tetrazole-5-thiols and 4-substituted 4 H-1,2,4-triazole-3-thiols with alkyl halides or sulfonates lead to the formation of S-alkylated products regardless of the substituent on the heterocycle. In this work, we found that substituted 1H-tetrazole-5-thiols and 4 H-1,2,4-triazole-3-thiols readily reacted with vinyl ethers in the absence of a catalyst to exclusively form N-substituted 1H-tetrazole-5(4H)-thiones and 1H-1,2,4-triazole-5(4H)-thiones, respectively. Furthermore, the reactions of 5-substituted-1H-tetrazoles with vinyl ethers under the same conditions selectively yielded 2,5-disubstitued tetrazoles.  相似文献

Various spiroindenyl-2-oxindoles have been synthesized in a one-pot reaction from isatin-derived propargylic alcohols with sterically hindered and electron-rich arenes such as 2-phenylindole and 1,3,5-trimethoxybenzene. Thermoacoustic theory is extended to stacks made of random bulk media. Characteristics of the porous stack such as the tortuosity and dynamic shape factors are introduced into the thermoacoustic wave equation in the low reduced frequency approximation. Basic thermoacoustic equations for a bulk porous medium are formulated analogously to the equations for a single pore. Use of different dynamic shape factors for the viscous and thermal effects is adopted and scaling using the dynamic shape factors and tortuosity is demonstrated. Comparisons of the calculated and experimentally derived thermoacoustic properties of reticulated vitreous carbon and aluminum foam show good agreement. A consistent mathematical model of sound propagation in a random porous medium with an imposed temperature is developed. This treatment leads to an expression for the coefficient of the temperature gradient in terms of scaled cylindrical thermoviscous functions.  相似文献
