New pyridine, pyrazoloyridine, and furopyridine derivatives substituted with naphthyl and thienyl moieties were designed and synthesized starting from 6-(naphthalen-2-yl)-2-oxo-4-(thiophen-2-yl)-1,2-dihydropyridine-3-carbonitrile (1). The chloro, methoxy, cholroacetoxy, imidazolyl, azide, and arylamino derivatives were prepared to obtain the pyridine-C2 functionalized derivatives. The derived pyrazolpyridine-N-glycosides were synthesized via heterocyclization of the C2-thioxopyridine derivative followed by glycosylation using glucose and galactose. The furopyridine derivative 14 and the tricyclic pyrido[3′,2′:4,5]furo[3,2-d]pyrimidine 15 were prepared via heterocyclization of the ester derivative followed by a reaction with formamide. The newly synthesized compounds were evaluated for their ability to in vitro inhibit the CDK2 enzyme. In addition, the cytotoxicity of the compounds was tested against four different human cancer cell lines (HCT-116, MCF-7, HepG2, and A549). The CDK2/cyclin A2 enzyme inhibitory results revealed that pyridone 1, 2-chloro-6-(naphthalen-2-yl)-4-(thiophen-2-yl)nicotinonitrile (4), 6-(naphthalen-2-yl)-4-(thiophen-2-yl)-1H-pyrazolo[3,4-b]pyridin-3-amine (8), S-(3-cyano-6-(naphthaen-2-yl)-4-(thiophen-2-yl)pyridin-2-yl) 2-chloroethanethioate (11), and ethyl 3-amino-6-(naphthalen-2-yl)-4-(thiophen-2-yl)furo[2,3-b]pyridine-2-carboxylate (14) are among the most active inhibitors with IC50 values of 0.57, 0.24, 0.65, 0.50, and 0.93 µM, respectively, compared to roscovitine (IC50 0.394 μM). Most compounds showed significant inhibition on different human cancer cell lines (HCT-116, MCF-7, HepG2, and A549) with IC50 ranges of 31.3–49.0, 19.3–55.5, 22.7–44.8, and 36.8–70.7 μM, respectively compared to doxorubicin (IC50 40.0, 64.8, 24.7 and 58.1 µM, respectively). Furthermore, a molecular docking study suggests that most of the target compounds have a similar binding mode as a reference compound in the active site of the CDK2 enzyme. The structural requirements controlling the CDK2 inhibitory activity were determined through the generation of a statistically significant 2D-QSAR model.  相似文献

The aim of this work is to the study the influence of the isomer structures of butyl acrylate monomer on the single‐electron transfer/degenerative chain transfer mediated living radical polymerization (SET‐DTLRP). The kinetic of isobutyl acrylate is determined for the first time by SET‐DTLRP in water catalyzed by sodium dithionite. The plots of number‐average molecular weight versus conversion and ln([M]0/[M]) versus time are linear, demonstrating a controlled polymerization. The influence of the isomer t‐butyl, i‐butyl, and n‐butyl on the kinetics, properties, and stereochemistry of the reactions was assessed. To the best of our knowledge, there is no previous report dealing with the synthesis of PiBA by any LRP approach in aqueous medium. The results presented in this work suggest that the stability provided by the acrylate side group has an important influence in the polymerization process. © 2008 Wiley Periodicals, Inc. The mechanism of alkaline earth metal tungstate formation during soft solution processing was studied by cyclic voltammetry, electrochemical impedance spectroscopy and by direct in situ observation of the surface changes using atomic force microscopy. The electrochemical oxidation of W to WO3 was followed by dissolution of WO3 and, with some delay, by precipitation of tungstates at the metal surface. The same Tafel slopes observed in Li+, Ba2+, Sr2+ and Ca2+ containing solutions indicate that the course of the oxidation process is independent of the cation present in solution. The observed differences in the current-voltage curves outside the Tafel region are accounted for by the different film-forming tendencies of the various alkaline earth metal cations. The growth of tungstate layers at the W substrate decreases the electrochemically active area and limits the production of WO4 2– at later stages of deposition. At low potentials (E<0.2 V) the oxidation of W is the rate-controlling step. At higher potentials, however, the dissolution process slows down due to a relative decrease of the pH in the electrode vicinity and dissolution becomes the rate-limiting step. Adsorption dynamics of chlorobenzene vapors on a 5% V2O5/Al2O3 catalyst has been investigated using the frontal chromatography technique. The uptakes of chlorobenzene have been measured as a function of vapor concentration and adsorption equilibrium has been found to follow formally the Langmuir isotherm. The breakthrough time proved to be a linear function of the column length as expected. Breakthrough profiles have been reported for different experimental conditions and quantitatively fitted by a reduced lumped diffusion model. This model provides an analytical solution that facilitates engineering calculations. Model parameters show complex behavior as functions of stream characteristics and depend on column length. When empirical expressions relating model mass transfer coefficients with influencing variables are found the model demonstrates good accuracy in predicting column performance.  相似文献

The Nazarov reaction of pentadienyl cations generated by protonation of either dienones or alkoxytrienes has been examined in detail both experimentally and by DFT calculations. In particular, calculations at the B3LYP/6‐311G** level of theory accurately predicted, and accounted for, the outcome of the Brønsted acid catalyzed electrocyclization of 4π‐electron systems in which one of the double bonds involved in the process was embedded in N‐ and S‐heterocyclic rings. Calculations showed that both heteroatoms are capable of accelerating the ring closure by stabilizing the partial positive charge which develops at C‐6 (C‐2) in the transition state, with S‐heterocyclic derivatives being more reactive than the corresponding N‐containing compounds. In general, pentadienyl cations generated by protonation of alkoxytrienes were expected to react faster than those obtained by protonation of the corresponding dienones, as the latter were stabilized by a hydrogen bond. The presence of a substituent on the heterocyclic ring significantly affects the stereoselectivity (torquoselectivity) only in the case of the N‐heterocyclic derivatives, in which a 2‐alkyl group is axially oriented, providing the cis‐2,5‐disubstituted isomer only. Instead, with substituted S‐heterocyclic compounds, the anticipated torquoselectivity was very low and, in fact, a 3:1 diastereomeric mixture between the trans and cis products was experimentally found after ring closure. For this study, the synthesis of the appropriate N‐ and S‐containing dienones and alkoxytrienes was realized to evaluate the predictivity power of the DFT computations, which was very good in all of the cases examined, both in terms of reactivity and stereoselectivity. The consistency observed between computational and experimental results, therefore, shows the usefulness of DFT calculations at the B3LYP/6‐311G** level of theory as a robust instrument for the prediction of reactivity and stereoselectivity in the Nazarov electrocyclic reaction.  相似文献

Implementation of DART and DESI ionization on a fieldable mass spectrometer   总被引:1,自引:1,他引:0  
A recently developed prototype mobile laboratory mass spectrometer, incorporating an atmospheric pressure ionization (API) interface, is described. This system takes advantage of the small size, lower voltage requirements, and tandem MS abilities of the cylindrical ion trap mass analyzer. The prototype API MS uses small, low-power pumps to fit into a 0.1-m(3) self-contained package weighing <45 kg. This instrument has been adapted to allow rapid interfacing to electrospray ionization, desorption electrospray ionization, and direct analysis in real-time sources. Initial data indicate that these techniques provide rapid detection and identification of compounds for quality control, homeland security, and forensic applications. In addition, this instrument is self-contained and compact, making it ideally extensible to mobile laboratory and field analyses. Initial MS and MS/MS data for analyses of drugs, food, and explosives are presented herein.  相似文献

[Ge9]4? Zintl clusters are used as soluble germanium source for a bottom–up fabrication of Ge nanomorphologies such as inverse opal structures with tunable composition. The method is based on the assembly and oxidation of [Ge9]4? clusters in a template mold using SiCl4, GeCl4, and PCl3 leading to Si and P‐containing Ge phases as shown by X‐ray diffraction, Raman spectroscopy, and energy‐dispersive X‐ray analysis. [Ge9]4? clusters are retained using ethylenediamine (en) as a transfer medium to a mold after removal of the solvent if water is thoroughly excluded, but are oxidized to amorphous Ge in presence of water traces. 1H NMR spectroscopy reveals the oxidative deprotonation of en by [Ge9]4?. Subsequent annealing leads to crystalline Ge. As an example for wet‐chemical synthesis of complex Ge nanomorphologies, we describe the fabrication of undoped and P‐doped inverse opal‐structured Ge films with a rather low oxygen contents. The morphology of the films with regular volume porosity is characterized by SEM, TEM, and grazing incidence small‐angle X‐ray scattering.  相似文献

Dihalocarbenes add regioselectively to aryl-substituted benzocyclopropenes to produce dihalobenzocyclobutenes. The regioselectivity of addition is not due to steric effects but depends on the electronic donor or acceptor ability of the substituent. B3LYP/6-31G* calculations show preferential :CCl2 addition to substituted benzocyclopropene through electrophilic attack on the benzocyclopropene pi-system (Ea = 1.1-2.4 kcal/mol) rather than C-C sigma-bond insertion into the cyclopropenyl moiety (Ea = 5-24 kcal/mol). pi-Addition proceeds regioselectively through a single transition state to xylylene intermediates or directly to benzocyclobutene products.  相似文献

Poly(vinyl chloride) degrades when exposed to ultraviolet light for long durations; therefore, the photostability of polymeric materials should be enhanced through the application of additives. New organotin complexes containing 4-aminonaphthalene-1-sulfonic acid were synthesized and their role as poly(vinyl chloride) photostabilizers were evaluated. The reaction of 4-amino-3-hydroxynaphthalene-1-sulfonic acid and appropriate di- or trisubstituted tin chloride (triphenyltin chloride, tributyltin chloride, dibutyltin dichloride, and dimethyltin dichloride) in methanol under reflux gave the corresponding tin-naphthalene complexes with yields of 75%–95%. Elemental analyses and spectroscopic techniques including infrared and nuclear magnetic resonance (proton and tin) were used to confirm their structures. The tin complexes were added to poly(vinyl chloride) to produce thin films that irradiated with ultraviolet light. Various parameters were assessed, such as the weight loss, formation of specific functional groups, changes in the surface due to photoirradiation, and rate constant of photodegradation, to test the role played by the organotin complexes to reduce photodegradation in polymeric films. The results proved that organotin complexes acted as photostabilizers in these circumstances. The weight loss, formation of fragments containing specific functional groups, and undesirable changes in the surface of polymeric films were limited in the presence of organotin complexes. Organotin complexes containing three phenyl groups showed the most desirable stabilization effect. These act as efficient primary and secondary photostabilizers, and as decomposers for peroxides. In addition, such an additive inhibits the dehydrochlorination process, which is the main cause of poly(vinyl chloride) photodegradation.  相似文献
