We report the generation of high-power picosecond infrared pulses by stimulated electronic Raman scattering in an atomic vapour. Single 30 ps pulses at 532 nm were Raman-shifted in caesium vapour to produce 0.4 mJ pulses at 2.38 μm, with 20% quantum efficiency. This technique can be extended to cover a wide range of infrared wavelenghts by using various Raman transitions.  相似文献

A wide class of Hamiltonian systems exhibit a mixture of slow motion with superimposed fast oscillations. Under the assumption of scale separation, these systems can be investigated using the principle of adiabatic invariance. In this paper, we start with a review of some of the main theoretical and numerical findings. We then briefly summarize a few important implications for molecular dynamics (MD) before we provide a more extensive discussion of numerical weather prediction (NWP). In particular, the conservative Hamiltonian particle-mesh (HPM) method is extended to Euler's equation and the fundamental concepts of geostrophic and hydrostatic balance are illustrated on the level of fluid blobs. We also demonstrate numerically that symplectic time-stepping methods are able to maintain hydrostatic balance to high accuracy.  相似文献

Very large nonlinear pulse broadening in 10–20 km lengths of monomode fibre is reported. Broadening of 150 ps pulses to as much as 2.5 ns has been observed. There is good qualitative agreement with a numerical analysis of the nonlinear Schrödinger equation.  相似文献

Feeding strategies of earthworms and their influence on soil processes are often inferred from morphological, behavioral and physiological traits. We used (13)C and (15)N natural abundance in earthworms, soils and plants to explore patterns of resource utilization by different species of earthworms in three tropical ecosystems in Puerto Rico. In a high altitude dwarf forest, native earthworms Trigaster longissimus and Estherella sp. showed less (15)N enrichment ((15)N = 3-6 per thousand) than exotic Pontoscolex corethrurus ((15)N =7-9 per thousand) indicating different food sources or stronger isotopic discrimination by the latter. Conversely, in a lower altitude tabonuco forest, Estherella sp. and P. corethrurus overlapped completely in (15)N enrichment ((15)N = 6-9 per thousand), suggesting the potential for interspecific competition for N resources. A tabonuco forest converted to pasture contained only P. corethrurus which were less enriched in (15)N than those in the forest sites, but more highly enriched in (13)C suggesting assimilation of C from the predominant C(4) grass. These results support the utility of stable isotopes to delineate resource partitioning and potential competitive interactions among earthworm species. Copyright 1999 John Wiley & Sons, Ltd.  相似文献

Lacticin 3147 is a lantibiotic with seven lanthionine bridges across its two component peptides, Ltnα and Ltnβ. Although it has been proposed that the eponymous lanthionine and (β-methyl)lanthionine (Lan and meLan) bridges present in lantibiotics make an important contribution to protecting the peptides from thermal or proteolytic degradation, few studies have investigated this link. We have generated a bank of bioengineered derivatives of lacticin 3147, in which selected bridges were removed or converted between Lan and meLan, which were exposed to high temperature or proteolytic enzymes. Although switching Lan and meLan bridges has variable consequences, it was consistently observed that an intact N-terminal lanthionine bridge (Ring A) confers Ltnα with enhanced resistance to thermal and proteolytic degradation.  相似文献

The continuing problem of the emergence of multidrug resistance in pathogens has resulted in renewed efforts to identify novel antimicrobials that could be used in clinical settings. Lantibiotics are bacterially produced gene encoded antimicrobial peptides which have been the focus of extensive investigation in recent years because of their broad spectrum of activity. Lantibiotics (lanthionine-containing antibiotics), which have traditionally been regarded as antimicrobials for use in food or veterinary medicine, may provide at least part of the solution to these problems. Lacticin 3147 is a two peptide lantibiotic (consisting of the peptides Ltnα and Ltnβ) which is active at low concentrations against many pathogens. It has been the subject of extensive research, which has generated significant insights into the mechanisms of lacticin 3147 biosynthesis, immunity, structure function relationships and the consequences of molecular bioengineering. The merits of employing lacticin 3147 to control spoilage microbes as well as its potential in the elimination of food, human and veterinary pathogens have also been highlighted. Here we review the knowledge which has been gained with respect to lacticin 3147 since its discovery in 1995.  相似文献

The core histones, H2A, H2B, H3 and H4, undergo post‐translational modifications (PTMs) including lysine acetylation, methylation and ubiquitylation, arginine methylation and serine phosphorylation. Lysine residues may be mono‐, di‐ and trimethylated, the latter resulting in an addition of mass to the protein that differs from acetylation by only 0.03639 Da, but that can be distinguished either on high‐performance mass spectrometers with sufficient mass accuracy and mass resolution or via retention times. Here we describe the use of chemical derivatization to quantify methylated and acetylated histone isoforms by forming deuteroacetylated histone derivatives prior to tryptic digestion and bottom‐up liquid chromatography‐mass spectrometric analysis. The deuteroacetylation of unmodified or mono‐methylated lysine residues produces a chemically identical set of tryptic peptides when comparing the unmodified and modified versions of a protein, making it possible to directly quantify lysine acetylation. In this work, the deuteroacetylation technique is used to examine a single histone H3 peptide with methyl and acetyl modifications at different lysine residues and to quantify the relative abundance of each modification in different deacetylase and methylase knockout yeast strains. This application demonstrates the use of the deuteroacetylation technique to characterize modification ‘cross‐talk’ by correlating different PTMs on the same histone tail.
