Simarinolide 4a and Guanepolide 5a are new quassinoids with a C25 basic skeleton isolated from a member of the French Guyanan Simaroubaceae, Simaba cf orinocensis H.B.K. The structure 4a was established by spectral means and that of 5a by X-ray diffraction analysis. The previously known simarolide 1 was also isolated.  相似文献

Born-Oppenheimer equilibrium structures, r(e)(BO), of the electronic ground state of the borazane (BH3NH3) molecule of C3v point-group symmetry are computed ab initio using the CCSD(T) method with basis sets up to quintuple-zeta quality. Inclusion of the counterpoise correction and extrapolation of the structural parameters to the complete basis set limit yield a best estimate of r(e)(BO) of BH3NH3. The anharmonic force field of BH3NH3, computed at the CCSD(T) level of theory with a basis set of triple-zeta quality, allows the determination of semi-experimental equilibrium rotational constants, which in turn result in a semi-experimental equilibrium structure, r(e)(SE). The r(e)(BO) and r(e)(SE) structures are in excellent agreement, indicating the validity of the methods used for their determination. The empirical mass-dependent structure, r(m)(1), of BH3NH3 is also determined. Although it is inferior in quality to the previous two structures, it is much more accurate than the standard empirical r0 and r(s) structures reported earlier for BH3NH3. The semi-experimental r(e)(SE) as well as the empirical r(m)(1) structures determined are based on experimental ground-state rotational constants available from the literature for nine isotopologues of borazane. The effective barrier to the internal rotation of BH3NH3, a molecule isoelectronic with CH3CH3, has been computed ab initio, employing the focal-point analysis (FPA) approach, to be 699 +/- 11 cm(-1). This compares favorably with an empirical redetermination of the effective barrier based on the above r(e)(SE) structure, V3 = 718(17) cm(-1).  相似文献

The biodistribution of colloidal carriers after their administration in vivo depends on the adsorption of some plasma proteins and apolipoproteins on their surface. Poly(methoxypolyethyleneglycol cyanoacrylate-co-hexadecylcyanoacrylate) (PEG-PHDCA) nanoparticles have demonstrated their capacity to cross the blood-brain barrier (BBB) by a mechanism of endocytosis. In order to clarify this mechanism at the molecular level, proteins and especially apolipoproteins adsorbed at the surface of PEG-PHDCA nanoparticles were analyzed by complementary methods such as CE and Protein Lab-on-chip in comparison with 2-D PAGE as a method of reference. Thus, the ability of those methodologies to identify and quantify human and rat plasma protein adsorption onto PEG-PHDCA nanoparticles and conventional PHDCA nanoparticles was evaluated. The lower adsorption of proteins onto PEG-PHDCA nanoparticles comparatively to PHDCA nanoparticles was evidenced by 2-D PAGE and Protein Lab-on-chip methods. CE allowed the quantification of adsorbed proteins without the requirement of a desorption procedure but failed, in this context, to analyze complex mixtures of proteins. The Protein Lab-on-chip method appeared to be very useful to follow the kinetic of protein adsorption from serum onto nanoparticles; it was complementary to 2-D PAGE which allowed the identification (with a relative quantification) of the adsorbed proteins. The overall results suggest the implication of the apolipoprotein E in the mechanism of passage of PEG-PHDCA nanoparticles through the BBB.  相似文献

Composite material nanofilms of controlled thickness constituted by ceramics and polymers find more and more applications to improve the properties and functionalities of material surfaces. In this paper we present a new way to deposit such composite coatings by alternated contacts of a surface with a polyamine solution (either poly(allylamine), poly(ethyleneimine), poly-l-lysine, or poly(diallydimethylammonium)chloride and silicic acid. The experiments are mainly realized by spraying of solutions onto the surface. The polyamines deposited in the first spraying step catalyze silica formation upon further spraying of a silicic acid-containing solution. The film thickness increases linearly with the number of deposition steps, the thickness increment being of the order of a few nanometers per silicic acid/polyamine layer. Infrared spectroscopy in the total attenuated reflection mode reveals spectra that are close to those of pure silica particles. The film morphology is further investigated by means of atomic force microscopy and environmental scanning electron microscopy. This reactive layer-by-layer deposition constitutes a new way to build, in an easy way, nanocomposite coatings with precise control of their thickness.  相似文献

Whey protein isolate was heat-treated at 85 degrees C for 15 min at pH ranging from 6.0 to 7.0 in the presence of NaCl in order to generate the highest possible amount of soluble aggregates before insolubility occurred. These whey protein soluble aggregates were characterized for composition, hydrodynamic diameter, apparent molecular weight, zeta-potential, surface hydrophobicity index, activated thiol group content, and microstructure. The adsorption kinetics and rheological properties (E', etad) of these soluble aggregates were probed at the air/water interface. In addition, the gas permeability of a single bubble stabilized by the whey protein soluble aggregates was determined. Finally, the foaming and foam-stabilizing properties of these aggregates were measured. The amount of whey protein soluble aggregates after heat treatment was increased from 75% to 95% from pH 6.0 to pH 7.0 by addition of 5 mM to 120 mM NaCl, respectively. These soluble aggregates involved major whey protein fractions and exhibited a maximum of activated thiol group content at pH > 6.6. The hydrodynamic radius and the surface hydrophobicity index of the soluble aggregates increased from pH 6.0 to 7.0, but the molecular weight and zeta-potential decreased. This loss of apparent density was clearly confirmed by microscopy as the soluble aggregates shifted from a spherical/compact structure at pH 6.0 to a more fibrillar/elongated structure at pH 7.0. Surface adsorption was faster for soluble aggregates formed at pH 6.8 and 7.0 in the presence of 100 and 120 mM NaCl, respectively. However, interfacial elasticity and viscosity measured at 0.01 Hz were similar from pH 6.0 to 7.0. Single bubble gas permeability significantly decreased for aggregates generated at pH > 6.6. Furthermore, these aggregates exhibited the highest foamability and foam liquid stability. Air bubble size within the foam was the lowest at pH 7.0. The coarsening exponent, alpha, fell within predicted values of 1/3 and 1/2, except for very dry foams where it was 1/5.  相似文献

A bootstrap method for point-based detection of candidate biomarker peaks has been developed from pattern classifiers. Point-based detection methods are advantageous in comparison to peak-based methods. Peak determination and selection are problematic when spectral peaks are not baseline resolved or on a varying baseline. The benefit of point-based detection is that peaks can be globally determined from the characteristic features of the entire data set (i.e., subsets of candidate points) as opposed to the traditional method of selecting peaks from individual spectra and then combining the peak list into a data set. The point-based method is demonstrated to be more effective and efficient using a synthetic data set when compared to using Mahalanobis distance for feature selection. In addition, probabilities that characterize the uniqueness of the peaks are determined.This method was applied for detecting peaks that characterize age-specific patterns of protein expression of developing and adult mouse cerebella from matrix assisted laser desorption/ionization (MALDI) mass spectrometry (MS) data. The mice comprised three age groups: 42 adults, 19 14-day-old pups, and 16 7-day-old pups. Three sequential spectra were obtained from each tissue section to yield 126, 57 and 48 spectra for adult, 14-day-old pup, and 7-day-old pup spectra, respectively. Each spectrum comprised 71,879 mass measurements in a range of 3.5-50 kDa. A previous study revealed that 846 unique peaks were detected that were consistent for 50% of the mice in each age group (C. Laurent, D.F. Levinson, S.A. Schwartz, P.B. Harrington, S.P. Markey, R.M. Caprioli, P. Levitt, Direct profiling of the cerebellum by MALDI MS: a methodological study in postnatal and adult mouse, J. Neurosci. Res. 81 (2005) 613-621.).A fuzzy rule-building expert system (FuRES) was applied to investigate the correlation of age with features in the MS data. FuRES detected two outlier pup-14 spectra. Prediction was evaluated using 100 bootstrap samples of 2 Latin-partitions (i.e., 50:50 split between training and prediction set) of the mice. The spectra without the outliers yielded classification rates of 99.1 ± 0.1%, 90.1 ± 0.8%, and 97.0 ± 0.6% for adults, 14-day-old pups, and 7-day-old pups, respectively. At a 95% level of significance, 100 bootstrap samples disclosed 35 adult and 21 pup distinguishing peaks for separating adults from pups; and 8 14-day-old and 15 7-day-old predictive peaks for separating 14-day-old pup from 7-day-old pup spectra. A compressed matrix comprising 40,393 points that were outside the 95% confidence intervals of one of the two FuRES discriminants was evaluated and the classification improved significantly for all classes. When peaks that satisfied a quality criterion were integrated, the 55 integrated peak areas furnished significantly improved classification for all classes: the selected peak areas furnished classification rates of 100%, 97.3 ± 0.6%, and 97.4 ± 0.3% for adult, 14-day-old pups, and 7-day-old pups using 100 bootstrap Latin partitions evaluations with the predictions averaged. When the bootstrap size was increased to 1000 samples, the results were not significantly affected. The FuRES predictions were consistent with those obtained by discriminant partial least squares (DPLS) classifications.  相似文献

The free-radical copolymerisation of various acrylates and methacrylates resulting in complex copolymers for cosmetic applications were investigated using different chromatographic techniques including HPLC and on-line coupled two-dimensional (2D) liquid chromatography. The complete separation of all polymerisation products was achieved by gradient HPLC. A computated optimisation procedure, using the Polymer Chromatographic Model allowed us to design a step mobile phase gradient to improve resolution of homopolymer chromatographic separation. By combining gradient HPLC and SEC (Size Exclusion Chromatography) in a fully automated two-dimensional chromatography setup, the complex distributions of chemical composition and molar mass could be simultaneously described and fingerprinted.  相似文献

Due to their particular tensile properties and composition, flax cellulosic fibres are widely used in various applications and are important in the stem architecture, as a source of mechanical support. Here, we have examined how various chemical extractions of cell-wall polysaccharides might improve our understanding of the structure and composition of these fibres. A set of chemical extractions, which had previously been independently studied on fibres of different origins and by different laboratories, was run on the same batch of technical fibres, variety Eden, and the released non-cellulosic polysaccharides were analyzed in terms of sugar composition and size exclusion chromatography. No single-step protocol was able to extract all the non-cellulosic polysaccharides estimated to be in the range of 130–140 mg per g of dry fibres. The major polymers consisted of rhamnogalacturonan and glucomannan, while the minor ones were xyloglucan and xylan. Several types of stepwise extractions helped to elucidate the subfractions of each polymer that are differently retained within cell wall. From our data, we have pointed out a new model which might help to approach the supramolecular design of the flax cellulosic fibres and, afterwards, the relationships between their chemical composition and their mechanical properties.  相似文献

Local strain and temperature of an AA5754-O aluminum alloy sheet have been full-field measured during monotonous tensile tests carried out at room temperature. Sharp strain increases and temperature bursts which are locally generated by the Portevin-Le Chatelier phenomenon have been measured at the same point for two strain rates: V2?=?1.9?×?10?3?s?1 and V10?=?9.7?×?10?3?s?1. A relationship, which is based on the underlying physical mechanisms, has been established between the strain and the temperature and experimentally verified for the highest strain rate V10. The discrepancy between the theoretical and experimental results for the lowest strain rate V2 suggests that the localized plastic deformations do not follow an adiabatic transformation. Such a set-up seems to offer a direct and experimental method to check the adiabatic character of localized plastic deformations.  相似文献
