A large number of inhibitors of aldose reductase enzyme were submitted to the CASE (computer automated structure evaluation) program in order to ascertain the topological features relevant to activity. On the basis of the twenty-six biophores (activating fragments) and one biophobe (inactivating fragment), a new proposed interaction model was suggested for an aldose reductase enzyme with the chemical inhibitors. The critical relationship between enzyme inhibition and the structure of inhibitors is believed to depend on the relative positions of subordinate regions within the inhibitor structure.  相似文献

The operation of simulated moving beds (SMBs) at their optimal operating conditions is difficult and not robust. Therefore, it is common practice to operate SMB units far from their economic optimum in order to tolerate uncertainties in the system and minimize the effect of disturbances. Recently, we have proposed an on-line optimization based SMB control scheme that allows to exploit the full economic potential of SMB technology. The goal of this work is two-fold. Firstly, to experimentally evaluate and demonstrate the capability of the controller to optimize and operate the SMB units, thus delivering the products with maximum productivity and minimum solvent consumption. Secondly, to show the suitability of the controller even using a minimum of system information, thus making the detailed isotherm measurements redundant and saving time in the separation design phase. This paper reports and discusses the first experimental implementation of the control concept on a high purity separation of nucleosides (uridine, guanosine) with an eight-column four-section SMB where the species to be separated are retained on the source 30RPC stationary phase according to a linear isotherm.  相似文献

The electrochemical behavior of hemin, an iron complex of porphyrin, on binding to DNA at a glassy carbon electrode (GCE) and in solution, is described. Hemin, which interacts with covalently immobilized calf thymus DNA, was detected by use of a bare GCE, a double-stranded DNA-modified GCE (dsDNA-modified GCE), and a single-stranded DNA-modified GCE (ssDNA-modified GCE), in combination with differential pulse voltammetry (DPV). The structural conformation of DNA was determined from changes in the voltammetric signals acquired on reduction of hemin. As a result of its large steric structure and anionic substitution on its porphyrin plane, hemin intercalates between the base pairs of dsDNA. A scan-rate study for hemin and the dsDNA-hemin complex were also performed to determine the electrochemical behavior of the complex. The partition coefficient was obtained from the peak currents measured when different concentrations of hemin were in the presence of dsDNA. By observing the oxidation signals of guanine, damage to DNA after reaction with hemin at the GCE surface was also detected. The electrochemical detection of hybridization between the covalently immobilized probe and its target sequence was detected by use of hemin. These results demonstrate the use of DNA biosensors in conjunction with hemin for electrochemical detection of hybridization and damage to DNA.  相似文献

A simple method is proposed for the determination and speciation of Mn(II) and Mn(VII) in waters utilizing a macroporous resin, Amberlite XAD-7HP. The batch method was employed and flame atomic absorption spectrometry was used in all determinations. Amberlite XAD-7HP resin was shown to retain Mn(VII) between pH 4 and 12. If the solution contains only one of the species, either Mn(II) or Mn(VII), the resin behaves selectively depending on the pH of the solution. The elution from the sorbent was realized using K2C2O4 in HNO3. The efficiency of the method was checked via spike recovery experiments. The proposed method was successfully applied to industrial wastewater samples and quantitative recoveries (≥96.0%) confirmed the accuracy of the method.  相似文献

Varying amounts of nitric acid catalyst, water and ethyl alcohol were used in the preparation of SiO2 sols by hydrolysis and condensation reactions of tetraethyl orthosilicate in a one step acid catalysis process. Hydrolysis of TEOS was followed by FT-IR analyses. Size of SiO2 particles was seen to vary in 8–41 nm range with respect to changing HNO3 and water amounts in the sols. Gelation occurred in some systems. Surfaces of films were examined by FESEM and AFM, after coating on glass substrates by dip coating. Thicknesses of the films were measured to be in the range of 80–120 nm. 5.6 ± 0.2% point increase in light transmittance was obtained when HNO3/TEOS (mol/mol) ratio of 4.74 × 10? 4 and H2O/TEOS (mol/mol) ratio of 9.08 were utilized. Sols were found to be stable for months and coatings prepared after 45 days still provided 5.2 ± 0.2% point increase in light transmittance.  相似文献

We present a graphene oxide (GO) integrated disposable electrochemical sensor for the enhanced detection of nucleic acids and the sensitive monitoring of the surface-confined interactions between the anticancer drug mitomycin C (MC) and DNA. Interfacial interactions between immobilized calf thymus double-stranded (dsDNA) and anticancer drug MC were investigated using differential pulse voltammetry (DPV) and electrochemical impedance spectroscopy (EIS) techniques. Based on three repetitive voltammetric measurements of 120 μg mL(-1) DNA immobilized on GO-modified electrodes, the RSD % (n = 3) was calculated as 10.47% and the detection limit (DL) for dsDNA was found to be 9.06 μg mL(-1). EIS studies revealed that the binding of the drug MC to dsDNA leads to a gradual decrease of its negative charge. As a consequence of this interaction, the negative redox species were allowed to approach the electrode, and thus increase the charge transfer kinetics. On the other hand, DPV studies exploited the decrease of the guanine signal due to drug binding as the basis for specifically probing the biointeraction process between MC and dsDNA.  相似文献

Mesoporous silica SBA-15 has been synthesized and functionalized by one-step synthesis method to widen their various application possibilities. In this study, phenyltrimethoxysilane (PTMS), 3-mercaptopropyltrimethoxysilane (MPTMS) and trimethoxypropylsilane (TMPS) were used as silane precursors for the functionalization, and after treated with HCl solution, their catalytic activities were evaluated in the lactic acid-methanol esterification. The presence of anchoring of functional groups on SBA-15 was proved by XRD, FT-IR, BET surface area and pore size distributions. Good catalytic activity was observed especially for SBA-15-SO(3)H-MPTMS, and the catalytic activity order was determined as follows: SBA-15-SO(3)H-MPTMS>SBA-15-TMPS>SBA-15-PTMS, which is directly associated with the surface area, pore size and pore volume. As compared with homogeneous catalyst, SBA-15-SO(3)H-MPTMS heterogeneous catalyst shows remarkable performance, such as separation, recovery and reusability.  相似文献

A possible DNA damage after interaction of kainic acid (KA) with calf thymus double stranded DNA and genomic DNA was herein determined in in vitro and in vivo conditions using; electrochemical assay and agarose gel electrophoresis. The changes in guanine signal were detected as an indicator of DNA damage in genomic DNA samples isolated from 1 or 10 mg/kg KA‐treated animals. The decreased levels of guanine signal were found as 29% and 33% by 1 and 10 mg/kg KA treatment when compared to controls, respectively. The results of gel electrophoresis confirmed DNA damage obtained in identical samples by electrochemical method.  相似文献
