The purpose of this paper is to study the glass formation tendency in the ternary system B2O3―Bi2O3―MoO3 and to define the main structural units building the amorphous network. A wide glass formation area was determined which is situated near the Bi2O3―B2O3 side. A liquid phase separation region was observed near the MoO3―B2O3 side for compositions containing below 25 mol% Bi2O3 and their microheterogeneous structure was observed by SEM. The phase formation was characterized by X-ray diffraction (XRD). By DTA was established the glass transition temperature (Tg) in the range of 380-420 °C and crystallization temperature (Tx) vary between 420 and 540 °C. The main building units forming the amorphous network are BO3 (1270 and 1200 cm− 1), BO4 (930-880, 1050-1040 cm− 1), MoO4 (840-760 cm− 1) and BiO6 (470 cm− 1). It was proved that Bi2O3 favors the BO3 → BO4 transformations while MoO3 preserves BO3 units in the amorphous network.  相似文献

The possibilities for universal calibration based on multi-element aqueous standard solutions and graphite laboratory reference materials (graphite standards) for the electrothermal vaporization inductively coupled plasma optical emission spectrometric (ETV ICP OES) determination of Al, B, Ba, Cd, Co, Cr, Cu, Fe, Mn, Ni, P, Pb, S, Sr, Ti, V, and Zn in plant materials were investigated. A commercially available state-of-the-art ETV device was coupled with an Echelle ICP spectrometer equipped with a charge-injection-device (CID) camera for spectral detection. The transition area between transport tube and ETV graphite tube and the gas streams for inner gas, bypass gas, and modifier gas were optimized to achieve best transport efficiencies. The influence of four gaseous modifiers (CCl4, CHCl3, CCl2F2, and C3H8) added to the inner gas was studied. Five reference materials (RM P-Alfalfa, Lucerne; NIES CRM No.9 “Sargasso”; CTA-VTL-2 Virginia Tobacco Leaves; NIST SRM 1515 Apple Leaves; IAEA-V-10 Hay Powder) were used for method validation. If certified reference materials are not available, calibration against graphite standards or dried aqueous standard solutions is possible. Slurry sampling ETAAS was successfully applied to the determination of sodium impurities in single crystals of potassium titanyl phosphate (KTP) and potassium gadolinium tungstate (KGW). Platform atomizers coated with titanium carbide or tungsten carbide, respectively, were used in order to avoid sensitivity drift due to the changes in the composition and the structure of the platform surface. Calibration curves with aqueous standards could be used for the KGW slurry (no matrix effects); analysis of KTP slurry required the standard additions method. The precision of the proposed method was better than 3% R.S.D. The results obtained by the present method showed a good agreement with those obtained by an independent method-flame AAS after sample digestion, which is an evidence for the good accuracy of the proposed method.  相似文献

The Bruch's membrane is located beneath the retina in vertebrate eyes. We have used atomic force microscopy to examine the morphological and adhesion properties of collagen fibers located in different portions of the membrane. The D-periodicity of the fibers was 62.54 +/- 4.25 nm and 63.78 +/- 4.14 nm for regions away from the optic nerve and close to it, respectively. The adhesion properties of the collagen fibers were evaluated using force volume imaging on a number of different eye samples. The adhesion force we recorded in regions away from the optic nerve was different compared to regions close to the optic nerve. The reported results allow us to understand the nanoscopic properties of connective tissues in the eye and are important for the design of new and improved biomaterials.  相似文献

A simple and rapid slurry sampling electrothermal atomic absorption spectrometric (ETAAS) method was developed for the determination of traces of sodium and iron in single crystals of rubidium titanyl phosphate (RTP). The finely ground crystal material was dispersed in 5mL of 2% (v/v) HNO3, containing 0.005% (v/v) Triton-X-100. The fast furnace ETAAS analysis was performed using a pyrolytic platform coated with titanium carbide. No matrix interference was registered for either of the analytes in slurries containing 4 to 10mgmL–1 of RTP, which permitted simple calibration against aqueous solutions. The precision was about 2% RSD. The results for sodium and iron content in RTP were in good agreement with those obtained by flame AAS after sample digestion.  相似文献

The interstrand N2,N2-dG DNA cross-linking chemistry of the acrolein-derived gamma-OH-1,N2-propanodeoxyguanosine (gamma-OH-PdG) adduct in the 5'-CpG-3' sequence was monitored within a dodecamer duplex by NMR spectroscopy, in situ, using a series of site-specific 13C- and 15N-edited experiments. At equilibrium 40% of the DNA was cross-linked, with the carbinolamine form of the cross-link predominating. The cross-link existed in equilibrium with the non-crosslinked N2-(3-oxo-propyl)-dG aldehyde and its geminal diol hydrate. The ratio of aldehyde/diol increased at higher temperatures. The 1,N2-dG cyclic adduct was not detected. Molecular modeling suggested that the carbinolamine linkage should be capable of maintaining Watson-Crick hydrogen bonding at both of the tandem C x G base pairs. In contrast, dehydration of the carbinolamine cross-link to an imine (Schiff base) cross-link, or cyclization of the latter to form a pyrimidopurinone cross-link, was predicted to require disruption of Watson-Crick hydrogen bonding at one or both of the tandem cross-linked C x G base pairs. When the gamma-OH-PdG adduct contained within the 5'-CpG-3' sequence was instead annealed into duplex DNA opposite T, a mixture of the 1,N2-dG cyclic adduct, the aldehyde, and the diol, but no cross-link, was observed. With this mismatched duplex, reaction with the tetrapeptide KWKK formed DNA-peptide cross-links efficiently. When annealed opposite dA, gamma-OH-PdG remained as the 1,N2-dG cyclic adduct although transient epimerization was detected by trapping with the peptide KWKK. The results provide a rationale for the stability of interstrand cross-links formed by acrolein and perhaps other alpha,beta-unsaturated aldehydes. These sequence-specific carbinolamine cross-links are anticipated to interfere with DNA replication and contribute to acrolein-mediated genotoxicity.  相似文献

A TAT peptide was used to functionalize a gold surface by three different methods: adsorption from solution, microcontact printing, and dip-pen nanolithography (DPN). The composition and structure of the modified gold was characterized by atomic force microscopy (AFM), X-ray photoelectron spectroscopy (XPS), and Fourier transform -infrared reflection absorption spectroscopy (FT-IRRAS). We used two well-studied alkylthiols, mercaptohexadecanoic acid and 1-octadecanethiol, as a comparison in order to understand the structure of the TAT peptide monolayers prepared by the three methods. AFM studies allowed us to assess the homogeneity after each modification protocol. XPS was used to characterize the chemical composition of the gold surface after each functionalization procedure. The XPS results showed that surfaces modified with the TAT peptide by the three methods exhibit similar surface chemistry. Finally, FT-IRRAS experiments allowed us to conclude that the structure of the alkyl chains of the TAT peptides is fairly disordered and different after each procedure. Regardless of the type of surface functionalization method used, the monolayer of TAT peptide formed on the surface was of "liquidlike" nature.  相似文献

Integral membrane proteins in bacteria are co‐translationally targeted to the SecYEG translocon for membrane insertion via the signal recognition particle (SRP) pathway. The SRP receptor FtsY and its N‐terminal A domain, which is lacking in any structural model of FtsY, were studied using NMR and fluorescence spectroscopy. The A domain is mainly disordered and highly flexible; it binds to lipids via its N terminus and the C‐terminal membrane targeting sequence. The central A domain binds to the translocon non‐specifically and maintains disorder. Translocon targeting and binding of the A domain is driven by electrostatic interactions. The intrinsically disordered A domain tethers FtsY to the translocon, and because of its flexibility, allows the FtsY NG domain to scan a large area for binding to the NG domain of ribosome‐bound SRP, thereby promoting the formation of the quaternary transfer complex at the membrane.  相似文献

Juniperus excelsa M. Bieb and J. sabina L. contain essential oil (EO), while J. sabina also contains podophyllotoxin, which is used as a precursor for anti-cancer drugs. Two studies were conducted. The first assessed the variability in the EO profile and podophyllotoxin concentration of the two junipers, depending on the location and tree gender. The main EO constituents of J. excelsa were α-cedrol, α-limonene and α-pinene, while the constituents in J. sabina were sabinene, terpinen-4-ol, myrtenyl acetate and α-cadinol. The podophyllotoxin yield of 18 J. sabina accessions was 0.07–0.32% (w/w), but this was not found in any of the J. excelsa accessions. The second study assessed the effect of hydrodistillation (Clevenger apparatus) and steam distillation (in a semi-commercial apparatus) on the EO profile and bioactivity. The extraction type did not significantly alter the EO composition. The EO profiles of the two junipers and their accessions were different and may be of interest to the industry utilizing juniper leaf EO. Breeding and selection programs could be developed with the two junipers (protected species) in order to identify chemotypes with (1) a high EO content and desirable composition, and (2) a high concentration of podophyllotoxin in J. sabina. Such chemotypes could be established as agricultural crops for the commercial production of podophyllotoxin and EO.  相似文献

A capillary 2-D LC method coupled with IT MS has been used for separation and identification of peptides in rat hypothalamus. Animals of two different age groups (8 and 50 wk) were exposed to two different rates of CO(2 )in inhaled air to investigate the influence of different hypoxia/hypercapnia levels and their stress-related factor on the peptide excretion. Peptide compounds were fractionated (strong cation exchange chromatography), trapped, and separated (RP chromatography), and MS/MS mass spectra were used for identification. About 107 peptide compounds were identified and 88 of them were semiquantified. Among the characterized peptides, there were fragments from proteins such as proenkephalin A, proSAAS, prosomatostatin, prooxytocin, vasopressin, etc. Explorative principal component analysis (PCA) combined with hypothesis testing was applied to the obtained data to investigate the impact of age and hypoxic stress factors on the peptide pattern. Twenty-six peptides revealed significant differences in concentrations between the animal groups influenced by age and influx rate.  相似文献
