We present a novel application of independent component analysis (ICA), an exploratory data analysis technique, to two-dimensional electrophoresis (2-DE) gels, which have been used to analyze differentially expressed proteins across groups. Unlike currently used pixel-wise statistical tests, ICA is a data-driven approach that utilizes the information contained in the entire gel data. We also apply ICA on wavelet-transformed 2-DE gels to address the high dimensionality and noise problems typically found in 2-DE gels. Also, we use an analysis-of-variance (ANOVA) approach as a benchmark for comparison. Using simulated data, we show that ICA detects the group differences accurately in both the spatial and wavelet domains. We also apply these techniques to real 2-DE gels. ICA proves to be much faster than ANOVA, and unlike ANOVA it does not depend on the selection of a threshold. Application of principal component analysis reduces the dimensionality and tends to improve the performance by reducing the noise. 相似文献

Three new isomeric dipyridoannulenes were synthesized and characterized. These molecules possess differing conjugation pathways between the substituent alkoxy donating groups and the pyridyl acceptor groups. Optical absorption and emission properties of the dipyridoannulenes and their corresponding acyclic precursors were measured and correlated to structural differences and used to evaluate conjugation effectiveness and charge-transfer pathways. Optical properties of protonated dipyridoannulenes were also measured and found to be somewhat insensitive to the degree of protonation. Density functional studies of these systems at the B3LYP/6-31G* level provided insight into their stabilities, polarities, and quinoidal character. An analysis of the HOMO and LUMO molecular orbitals provided further information regarding charge-transfer behavior. These systems are good metal binding candidates, as the pyridine moiety can act as both an electron-acceptor and a site for metal coordination. 相似文献

Detailing hydrogen bonding and deprotonation equilibria between anions and urea/thiourea derivatives
Spectrophotometric and 1H NMR titrations of N-methoxyethyl-N'-(4-nitrophenyl)thiourea (3) by Bu(4-)NOAc show that in DMSO deprotonation of the receptor and formation of a hydrogen-bonded complex with anion proceed simultaneously but in MeCN deprotonation requires the participation of the second acetate anion. The formation constants of hydrogen-bonded complexes were determined from titrations in the presence of added acetic acid, which suppressed deprotonation. These constants together with independently measured stability constants of (AcO)(2)H(-) complexes were employed for a rigorous numerical analysis of titration results in the absence of added acid, which allowed us to determine the equilibrium deprotonation constants as well as pKa values for 3 in both solvents. Although 3 appeared to be a weaker acid than AcOH in both solvents, it can be deprotonated by acetate in dilute solutions when the concentration of liberated acetic acid is low enough. With disubstituted N,N-bis(methoxyethyl)-N'-(4-nitrophenyl)thiourea 4 only deprotonation equilibrium is observed. In contrast, both parent urea derivatives 1 and 2 cannot be deprotonated by acetate anions. Independent of the real type of equilibrium, whether it is a deprotonation or a hydrogen bonding, titration plots always can be satisfactorily fitted to a formal 1:1 binding isotherm. A relationship between apparent "binding constants" and real equilibrium constants of hydrogen bonding association and deprotonation processes is discussed. 相似文献

The binding of catechol derivatives (LH 2 = catechol, 4-methyl catechol, 4-t-butyl catechol, and dopamine) to 1- and 4.7-nm TiO2 nanoparticles in aqueous, pH 3.5 suspensions has been characterized by UV-vis spectroscopy. The binding constants derived from Benesi-Hildebrand plots are (2-4) x 10(3) M(-1) for the 1-nm nanoparticles and (0.4-1) x 10(4)M(-1) for the 4.7-nm particles. Ti(IV)L3 complexes were prepared from the same catechols. The L = methyl catechol, and dopamine complexes are reported for the first time. The TiL3 reduction potentials are not very sensitive to the nature of the catechol nor evidently are the binding constants to TiO2 nanoparticles. The intense (epsilon > or = 10(3) M(-1)cm(-1)), about 400-nm, ligand-to-metal charge-transfer (LMCT) absorptions of the nanoparticle complexes are compared with those of the TiL 3 complexes (epsilon approximately 10(4)M(-1) cm(-1)) which lie in the same spectral region. The nanoparticle colors are attributed (as are the colors of the Ti(IV)L3 complexes) to the tails of the about 400-nm LMCT bands. 相似文献

The most active and robust current catalysts for the copolymerization of carbon dioxide and epoxides or oxetanes, (salen)CrX in conjunction with PPNX (PPN(+) = (Ph3P)2N(+)) or n-Bu4NX (X = Cl, N3, CN, NCO), are characterized both in solution by infrared spectroscopy and in the solid-state by X-ray crystallography. All anions (X) afford six-coordinate chromium(III) PPN(+) or n-Bu4N(+) salts composed of trans-(salen)CrX2(-) species. Of the X groups investigated in (salen)CrX, chloride is easily displaced by the others, that is, the reaction of (salen)CrCl with 2 equiv of N3(-), CN(-), or NCO(-) quantitatively provide (salen)Cr(N3)2(-), (salen)Cr(CN)2(-), and (salen)Cr(NCO)2(-), respectively. On the other hand, addition of less than 2 equiv of azide to (salen)CrCl leads to a Schlenk (ligand redistribution) equilibrium of the three possible anions both in solution and in the solid-state as shown by X-ray crystallography and electrospray ionization mass spectrometry. It was further demonstrated that all trans-(salen)CrX2(-) anions react with the epoxide or oxetane monomers in TCE (tetrachloroethane) solution to afford an equilibrium mixture containing (salen)CrX x monomer, with the oxetane adduct being thermodynamically more favored. The ring-opening steps of the bound cyclic ether monomers by X(-) were examined, revealing the rate of ring-opening of the epoxides (cyclohexene oxide and propylene oxide) to be much faster than of oxetane, with propylene oxide faster than cyclohexene oxide. The electrochemical oxidation of promethazine hydrochloride was made on highly boron‐doped diamond electrodes. Cyclic voltammetry experiments showed that the oxidation mechanisms involved the formation of an adsorbed product that is more readily oxidized, producing a new peak with lower potential values whose intensity can be increased by applying the accumulation potential for given times. The parameters were optimized and the highest current intensities were obtained by applying +0.78 V for 30 seconds. The square‐wave adsorptive voltammetry results obtained in BR buffer showed two well‐defined peaks, dependent on the pH and on the voltammetric parameters. The best responses were obtained at pH 4.0, frequency of 50 s?1, step of 2 mV, and amplitude of 50 mV. Under these conditions, linear responses were obtained for concentrations from 5.96×10?7 to 4.76×10?6 mol L?1, and calculated detection limits of 2.66×10?8 mol L?1 (8.51 μg L?1) for peak 1 and of 4.61×10?8 mol L?1 (14.77 μg L?1) for peak 2. The precision and accuracy were evaluated by repeatability and reproducibility experiments, which yielded values of less than 5.00% for both voltammetric peaks. The applicability of this procedure was tested on commercial formulations of promethazine hydrochloride by observing the stability, specificity, recovery and precision of the procedure in complex samples. All results obtained were compared to recommended procedure by British Pharmacopeia. The voltammetric results indicate that the proposed procedure is stable and sensitive, with good reproducibility even when the accumulation steps involve short times. The accuracy, repeatability, and reproducibility characteristics of a method using multitoxin immunoaffinity column cleanup with liquid chromatography (LC) for determination of aflatoxins (AF; sum of aflatoxins B1, B2, G1, and G2) and ochratoxin A (OTA) in powdered ginseng and ginger have been established in a collaborative study involving 13 laboratories from 7 countries. Blind duplicate samples of blank, spiked (AF and OTA added) at levels ranging from 0.25 to 16.0 microg/kg for AF and 0.25 to 8.0 microg/kg for OTA were analyzed. A naturally contaminated powdered ginger sample was also included. Test samples were extracted with methanol and 0.5% aqueous sodium hydrogen carbonate solution (700 + 300, v/v). The extract was centrifuged, diluted with phosphate buffer (PB), filtered, and applied to an immunoaffinity column containing antibodies specific for AF and OTA. After washing the column with water, the toxins were eluted from the column with methanol, and quantified by high-performance LC with fluorescence detection. Average recoveries of AF from ginseng and ginger ranged from 70 to 87% (at spiking levels ranging from 2 to 16 microg/kg), and of OTA, from 86 to 113% (at spiking levels ranging from 1 to 8 microg/kg). Relative standard deviations for within-laboratory repeatability (RSDr) ranged from 2.6 to 8.3% for AF, and from 2.5 to 10.7% for OTA. Relative standard deviations for between-laboratory reproducibility (RSDR) ranged from 5.7 to 28.6% for AF, and from 5.5 to 10.7% for OTA. The present work is focused on studying chlorine and fluorine identity SN2 substitutions on a methyl center, within the framework of the newly introduced reaction electronic flux J(xi), that allows one to identify charge transfer and polarization mechanisms that take place along the reaction coordinate. The main results concern the discovery of different charge transfer mechanism, despite both reactions have the same energetic pattern with simultaneous bond breaking and formation. It turns out that the chlorine substitution is mainly driven by polarization effects and characterized by through bond interactions while intermolecular charge transfer dominates the fluorine exchange reaction, that is characterized by through space interactions. 相似文献
