This paper describes a novel clustering methodology for classifying over 700 conformations of a flexible analogue of GBR 12909, a dopamine reuptake inhibitor that has completed phase I clinical trials as a treatment for cocaine abuse. The major aspect of the clustering methodology includes an efficient data-conditioning scheme where a systematic feature extraction procedure based on the structural properties of the molecule was used to reduce the associated feature space. This allowed region-specific clustering that focused on individual pharmacophore elements of the molecule. For clustering of the reduced feature set, the fuzzy clustering partitional method was utilized. Due to the relational nature of the feature data, fuzzy relational clustering was employed, and it successfully detected natural groups defined by rotational minima around N(sp(3))-C(sp(3)), O(sp(3))-C(sp(3)), and C(sp(3))-C(sp(2)) bonds. The proposed clustering methodology also employed several cluster validity measures, which corroborated the partitions produced by the clustering technique and agreed with the results of hierarchical clustering using the XCluster program. Representative structures which exhibited a reasonable spread of energies and showed good spatial coverage of the conformational space were identified for use as putative bioactive conformations in a future Comparative Molecular Field Analysis of GBR 12909 analogues. The clustering methodology developed here is capable of handling other computational chemistry problems, and the feature extraction technique can be easily generalized to other molecules.  相似文献

The reactivity of free radical cations, produced by anodic oxidation of 9, 10-diphenylanthracene (and perylene), with polymerizable organic monomers has been investigated by amperometry.It results that the radical cations, which are relatively stable in acetonitrile and nitrobenzene, decay with the monomer substrate by either an addition reaction or an electron transfer process. The cationic species of the monomer thus produced by catalyze the cationic polymerization of the substrate.The cationic polymerization initiated in this way has been investigated in the case of styrene: this reaction shows some similarity with the process initiated by perchloric acid, but carbonium ion carriers seem to give the main contribution to the propagation stage.  相似文献

Reaction of [(Tp(Me)2)(2)UI] with KNR(2) (R = C(6)H(5), SiMe(3)) in tetrahydrofuran (THF) afforded the monomeric trivalent actinide amide complexes [(Tp(Me)2)(2)U[N(C(6)H(5))(2)]], 1, and [(Tp(Me)2)(2)U[N(SiMe(3))(2)]], 2. The complexes have been fully characterized by spectroscopic methods and their structures were confirmed by X-ray crystallographic studies. In the solid state 1 and 2 exhibit distorted pentagonal bipyramidal geometries. The U-NR(2) bond lengths in both complexes are the same but in complex 2 the greater steric demands of the N(SiMe(3))(2) ligand led to elongated U-N(pz) bonds, especially those opposite the amido ligand.  相似文献

The concept of using a short ionisation event, in this case a pulsed corona discharge, in conjunction with programmed gate delay is described. This technique is proposed for the selective study of different ionisation processes within the reaction region of an ion mobility spectrometer. The utility of such an approach was tested in a study of the ionisation of dipropylene-glycol-monomethyl-ether (DPM); a compound commonly used to test the operation of ion mobility spectrometers. Dipropylene-glycol-monomethyl-ether at a concentration of 113 microg m(-3) in air, with a water level of 75 mg m(-3) in air, was analysed using a switchable, high resolution ion mobility spectrometer, operating in the positive mode at 40 degrees C at ambient pressure. The ion mobility spectrometer was fitted with a pulsed corona discharge ionisation source, doped with ammonia at a concentration of 1.3 mg m(-3) in the reaction region, and interfaced to a mass spectrometer. Synchronisation of the ionisation event to the operation of the shutter grids for the drift region enabled different parts of the product ion population to be injected into the drift tube, and programming the gate delays produced a map of the gate delay verses drift time response surface. Ammonium bound dipropylene-glycol-monomethyl-ether was observed, [(DPM)NH4]+ (m/z 166) as well as the ammonium bound dimer [(DPM)2NH4]+ (m/z 314), the same as those observed with a 63Ni source. Two other species were also observed, but their molecular identity was not elucidated. One of them m/z 146, also observed with 63Ni, formed ammonium bound ions [(m/z 146)NH4]+ (K0= 1.49 cm2 V(-1) s(-1)), ammonium bound dimer ions [(m/z 146)2NH4]+(K0= 1.18 cm2 V(-1) s(-1)) and a mixed cluster ion with DPM [(m/z 146)(DPM)NH4]+(K0= 1.18 cm2 V(-1) s(-1)); while the other, m/z 88 a decomposition product, formed ammonium bound monomer [(m/z 88)NH4]+(K0= 1.68 cm2 V(-1) s(-1)), dimer ions [(m/z 88)2NH4]+(K0= 1.40 cm2 V(-1) s(-1)) and a mixed cluster ion containing DPM and ammonium, [(DPM)(m/z 88)2NH4]+(K0= 1.40 cm2 V(-1) s(-1)). The assignment of responses to these ions required the additional dimensionality in the data provided from the gate delay studies. The relationships evident in the programmable gate delay data enabled these ions to be differentiated from alternative assignments of possible nitrogen clusters, formed at the interface of the mass spectrometer.  相似文献

Reactions of trimethylaluminum, triethylaluminum, and diethylaluminum chloride and ethylaluminum dichloride with silica gel have been studied experimentally by infrared spectroscopy and elemental analysis. The silica gel was subjected to different pretreatments to alter surface functionalities prior to reaction. In all cases the extent of surface modification reaction follows the trend unmodified > 600 degrees C pretreated > hexamethyldisilazane (HMDZ) pretreated > 600 degrees C/HMDZ pretreated. All of the aluminum compounds studied completely react non-hydrogen-bonded silanols, while also reacting with hydrogen-bonded silanols and siloxanes. Primarily monomeric surface species result from the surface modification reaction. Ethylaluminum chlorides preferentially react with silanols through cleavage of the Al-C bond rather than the Al-Cl bond. Singly bonded Si(s)-O-AlCl(2) surface species are readily synthesized by reaction of ethylaluminum dichloride with HMDZ-pretreated silica gel. Bridged bonded (Si(s)-O)(2)-AlCl surface species are readily synthesized by reaction of diethylaluminum chloride with HMDZ-pretreated silica gel. Computational ab initio studies of the cluster Si(4)O(6)(OH)(4) as a model to study the reaction of monomeric and dimeric methylaluminum dichloride with a silica silanol are also described. Comparison of the potential energy surface (PES) of monomer and dimer indicates that the energetics favor monomer reaction, consistent with experimental results. The energy cost in the dimer reaction is primarily from cleavage of a bridged Al-Cl bond upon adsorption. This does not occur when the monomer adsorbs. A comparison of the PES for the two reaction pathways resulting from cleavage of either an Al-Cl or Al-C bond indicates that while the former reaction is slightly kinetically favored (E(a) = 23.1 kJ/mol for Al-Cl bond cleavage versus E(a) = 31.1 kJ/mol for Al-C bond cleavage), the latter is strongly thermodynamically favored with an overall free energy difference between the two reaction pathways of 135 kJ/mol favorable to Al-C bond cleavage. A method for preconcentration and determination of trace amounts of cadmium in high saline samples is described. It is based on the adsorption of the metal in the activated carbon as complex cadmium(II)-4-(2-pyridylazo-resorcinol) (PAR). The final determination was carried by flame atomic absorption spectrometry (FAAS). The optimization of extraction parameters such as the pH effect, PAR mass, activated carbon mass and shaking time was carried out using a two-level full factorial design (24) and two Doehlert matrix designs. The results of the factorial design, considering the analysis of variance (ANOVA), demonstrate that all these factors are statistically significant, as well as the interactions (pH×PAR mass), (pH×activated carbon mass) and (activated carbon mass×shaking time). The final optimization was carried out using Doehlert matrix designs considering the results of the factorial design. The recoveries were quantitative (96.0-106.7%) for seawater samples spiked with Cd at concentrations of 0.125 and 0.625 μg l−1. A preconcentration factor of 149 was obtained. The effect of diverse metallic ions on the proposed procedure was investigated too. The procedure was used for cadmium determination in surface seawater samples collected in Salvador City, Brazil. The cadmium content in the analysed samples varies from 0.035 to 0.17 μg l−1. These results are agreement with other data reported in the literature.  相似文献

Nanocomposites from polyamide 11 and dried cellulose nanofibers (CNs), 16–30 nm in thickness and 50–400 nm in length, were prepared via direct melt mixing and their micro- and nano-mechanical properties were studied. (PF) QNM (Quantitative Nanomechanical Mapping) method was used to map nanomechanical properties at the surface of polyamide 11 and nanocomposites. This new AFM method emphasized both the increased modulus in nanocomposites as compared to the matrix and the microstructure on different levels in polyamide 11 and its nanocomposites. PF QNM showed that their crystalline structure consists of bundles of lamellar stacks, 200–350 nm in width and 20–40 nm wide lamellar stacks. Moreover, PF QNM study emphasized higher structural order in nanocomposites with 3 and 5 wt.% CNs and lower in the nanocomposite with 8 wt.% CNs as compared to the reference. These observations were verified and are consistent with both crystallinity values determined by DSC and micro-mechanical test results. The oriented bundles of lamellar stacks, observed by PF QNM, could be considered as the main blocks determining high mechanical properties for the studied nanomaterials.  相似文献

The temporal evolution of concentrations of dimethylsulphide (DMS), its precursor dimethylsulphoniopropionate (DMSP) and chlorophyll a is surveyed weekly in the water column and in a landfast ice core at a coastal station of Gerlache Inlet (Terra Nova Bay, Antarctica) from 27 November 2000 to 14 February 2001. The DMS and DMSP profile concentrations in the water column are similar and show a clear temporal trend, with minimum values (<0.7?nM) at all depths occurring on 27 November 2000 and maximum values (4.8 × 102?nM for DMS and 1.8 × 102?nM for DMSP) in surface water on 27 December 2000 for DMS and on 19 December 2000 for DMSP. When the sea-ice cover is present, the temporal evolution of DMSP closely follows that of chlorophyll a in the water column, supporting the idea that DMSP, and therefore DMS, has a phytoplanktonic origin. However, when the ice cover breaks up during the late austral summer, a second phytoplankton bloom occurs, while the DMSP concentration in the sea-water column remains very low. In the ice core, the results show higher concentrations of DMSP than those of the underlying sea water, highlighting the important role of sea ice in the sulphur cycle of the Antarctic ecosystem.  相似文献
