The skin integrity is essential due to its pivotal role as a biological barrier against external noxious factors. Pentacyclic triterpenes stand as valuable plant-derived natural compounds in the treatment of skin injuries due to their anti-inflammatory, antioxidant, antimicrobial, and healing properties. Consequently, the primary aim of the current investigation was the development as well as the physicochemical and pharmaco-toxicological characterization of betulin- and lupeol-based oleogels (Bet OG and Lup OG) for topical application in skin injuries. The results revealed suitable pH as well as organoleptic, rheological, and textural properties. The penetration and permeation of Bet and Lup oleogels through porcine ear skin as well as the retention of both oleogels in the skin were demonstrated through ex vivo studies. In vitro, Bet OG and Lup OG showed good biocompatibility on HaCaT human immortalized cells. Moreover, Bet OG exerted a potent wound-healing property by stimulating the migration of the HaCaT cells. The in ovo results demonstrated the non-irritative potential of the developed formulations. Additionally, the undertaken in vivo investigation indicated a positive effect of oleogels treatment on skin parameters by increasing skin hydration and decreasing erythema. In conclusion, oleogel formulations are ideal for the local delivery of betulin and lupeol in skin disorders.  相似文献

Photodynamic therapy (PDT) is a tool for the treatment of certain cancerous and pre-cancerous conditions. The natural precursor of porphyrins 5-aminolevulinic acid (ALA) has been extensively used as a pro-photosensitiser in PDT. ALA's poor permeability has been enhanced by chemical esterification with aliphatic alcohols. Some of the ALA esters proved to be more efficient than ALA for porphyrin synthesis. In the present work we studied the nature of porphyrin synthesis regulation from the ALA esters Hexyl-ALA (He-ALA) and R,S-ALA-2-(hydroxymethyl)tetrahydropyranyl ester (THP-ALA) in an adenocarcinoma cell line. We found that He-ALA is incorporated into the cells at a higher rate, followed by THP-ALA and ALA, whereas ALA and ALA esters efflux at the same rate mediated by passive diffusion. Although ALA entrance to the cell might be regulatory at low concentrations, ALA derivative uptake is not a limiting factor. At high concentrations, the regulation of ALA conversion into porphyrins is driven by the enzyme porphobilinogenase, whereas ALA esters hydrolysis is regulated by esterases. The key conclusion of this contribution is that the use of ALA esters has to be limited to low concentrations where no regulation on porphyrin synthesis takes place.  相似文献

After over a century of extensive research, hemoglobin has become the prototype of allosteric and cooperative proteins. Its molecular structure, known in great detail, has allowed the design of hundreds of site directed mutations, aimed at interfering with its function, and thus at testing our hypotheses on the molecular mechanisms of allostery. The wealth of information thus obtained is difficult to read except for specialists, not only because it makes use of many different technical approaches, but also because of its intrinsically patchy nature. Moreover, several researchers have tried to assign specific roles to segments of the polypeptide chains, rather than to single residues, and have tested their hypotheses by multiple point mutations or by complete replacement with the homologous segment from a different hemoglobin to produce chimeric macromolecules. This approach is in great need of a revision since putative functionally relevant segments partially overlap. This review briefly describes the structure and function of hemoglobin, and analyzes the effect of point mutations, multiple mutations and segment replacement, with special attention to possible biotechnological applications, ranging from pharmacology (Hb solutions as resuscitating fluids and sources of the protein found in hemoglobinopathies for biochemical studies) to bioreactors. Occasional reference is made to site directed mutants of myoglobin, whenever this helps clarifying perplexing results obtained on hemoglobin.  相似文献

Regioselectivity control was studied in palladium catalysed hydroxycarbonylation of styrene in neat water with water-soluble phosphines, mostly trisulfonated triphenylphosphine, TPPTS, but also N-bis(N',N'-diethyl-2-aminoethyl)-4-aminomethylphenyl-diphenylphosphine, N3P. The factor giving the highest changes in regioselectivity in the TPPTS system, under similar reaction conditions, is the temperature. In the N3P case, only a minor variation in the n/i ratio as a function of temperature is observed. Insitu normal- and high-pressure NMR experiments were performed to obtain further information about the catalytic cycle and the reaction intermediates. Two palladium hydride intermediates, a palladium eta3-benzylic complex and both the branched and linear palladium acyl complexes were identified in the HP NMR experiments. The hydroxycarbonylation in water using styrene as a substrate operates using a hydride mechanism for pathways leading to both linear and branched product. Insertion of styrene in the palladium-hydride bond gives an eta3-benzyl compound. A high CO pressure gives a kinetic preference for the iso-acyl in the next step. In the TPPTS system, at moderate temperatures, the hydrolysis of the iso-acyl is faster than its conversion to the thermodynamically more stable n-acyl. A low n/i therefore requires high pressures and reasonably low temperatures. The N3P ligand always favours the linear product since isomerisation of the iso-acyl to the n-acyl in this system is fast under all conditions investigated.  相似文献

We define and study numerical ranges for pairs of nonlinear operators F and J which act between some Banach space X and its dual X*, with respect to some increasing gauge function φ. Connections with spectra for certain classes of nonlinear operators introduced recently in the literature are also established. As a sample example, we consider the case when F is the duality map of the Lebesgue space L p (Ω), J is the duality map of the corresponding Sobolev space W 0 1,p (Ω), and φ(t)=t p?1 (1<p<∞). This leads to existence, uniqueness, and perturbation results for a homogeneous eigenvalue problem involving the p-Laplace operator.  相似文献

We consider in this paper the equations of motion of third grade fluids on a bounded domain of or with Navier boundary conditions. Under the assumption that the initial data belong to the Sobolev space H 2, we prove the existence of a global weak solution. In dimension two, the uniqueness of such solutions is proven. Additional regularity of bidimensional initial data is shown to imply the same additional regularity for the solution. No smallness condition on the data is assumed.  相似文献

This paper describes the morphology of the sexual phase and spores of Platycerium andinum and Platycerium wandae. Spores were sown in Thompson's media and the cultures were kept at 24–25 °C, with 12 h light/darkness photoperiod. Developmental phases were fixed in FAA and processed for observation with the scanning electron microscope. Spores of both species are monolete; Vittaria-type germination and Aspidium-type prothallial development were observed. In the phase of development, the gametophytes develop unicellular secretory and as they mature, develop bifurcated or branched pluricellular trichomes, both in the cushion and near the meristematic zone. Adult gametophytes in culture are cordiform-spatulate to cordiform-reniform, most are unisexual and a few are bisexual. Gametangia belong to the leptosporangiate fern type. Archegonial morphology is uniform, with an elongate, thin neck curved toward the base of the gametophyte. Antheridia have a basal cell, an annular cell and an undivided opercular cell. Three hundred days after the spores were sown, sporophytes still had not developed. In both species, some spores germinate inside the sporangial capsule (intra-sporangial germination). We provide new information on morphogenesis in the genus Platycerium.  相似文献

In this article, we describe the synthesis of a small library of short peptoids composed of four glycine residues and acylated with a fatty acid that showed a remarkable in vitro activity against two fungal plant pathogens. Their straightforward synthesis implied two consecutive Ugi reactions and can be efficiently extended to the construction of highly diverse libraries.  相似文献

We deal with nonlinear T-periodic differential systems depending on a small parameter. The unperturbed system has an invariant manifold of periodic solutions. We provide the expressions of the bifurcation functions up to second order in the small parameter in order that their simple zeros are initial values of the periodic solutions that persist after the perturbation. In the end two applications are done. The key tool for proving the main result is the Lyapunov-Schmidt reduction method applied to the T-Poincaré-Andronov mapping.  相似文献
