A highly sensitive and selective liquid chromatography-mass spectrometry (LC-MS) method has been developed for the determination of epirubicin in serum and cell specimens using daunorubicin as an internal standard. Using atmospheric pressure chemical ionisation (APCI), the epirubicin metabolites were readily distinguishable by their fragmentation pattern in the mass spectrometer. Selected reaction monitoring (SRM) mode was employed for quantitation of epirubicin and the metabolites. Following extraction, chromatography was performed on a C18 column with a mobile phase consisting of water-acetonitrile-formic acid, pH 3.2, with a flow rate of 200 μl/min. The limit of detection (LOD) and the limit of quantitation (LOQ) of this method in serum were determined to be 1.0 and 2.5 ng/ml, respectively. Linearity of the method was verified over the concentration range of 2.5-2000 ng/ml, with a high correlation coefficient (R2 ≥ 0.998). For the extraction procedure, an aliquot of 500 μl serum, spiked with internal standard, was extracted using a chloroform-2-isopropanol (2:1, v/v) mixture. The misuses of veterinary drugs can result in the accumulation of residues in food of animal origin that can make its way to the final consumer. Herein we describe a simple method for the accurate determination of beta-lactams, quinolones, sulphonamides, and tetracyclines in fish, poultry, and red meat. No extraction cartridges were used; instead, the extraction process consisted of the addition of an organic solvents, shaking, centrifugation, and dilution. An extensive validation process demonstrated an excellent linearity (R2 ≥ 0.99) for 23-drug residues. The recovery of drugs in different matrices at two concentration levels (n = 6) was in the range of 82–119%. The method was proved to be repeatable and reproducible with intra/inter-day measurements (RSDs lower than 20%). The quantification limits of drug residues were in the range of 0.8 to 45.3 ug/kg, which is well below the maximum residue limits set by most regulatory authorities. Summary A major problem in the HPLC analysis of alkaloids is the poor peak shape and consequently low resolution, due to the interactions of the basic alkaloids with the residual acidic silanol groups of most reversed phase materials. The performance of new packing materials specially designed for the separation of basic compounds has been studied using mobile phases without the special additives commonly applied in the analysis of alkaloids. Strongly basic Cinchona alkaloids were used as test compounds. Retention characteristics and selectivities of each material were studied, after mobile phase optimisation for the column. The influence of the major factors (nature and content of the organic modifier, pH value, salt concentration) affecting resolution was studied. The mobile phases were chosen so that they could be used in thermospray LC-MS. The addition of salts to the mobile phase improves separation but in general the modification of the mobile phase gave little change in selectivity. The performance of silicabased C18 material proved superior to the polymer materials tested. 相似文献
A method for the determination of isocyanates as di-n-butyl amine (DBA) derivatives using tandem mass spectrometry (MS/MS) and electrospray ionisation (ESI) is presented. Multiple-reaction monitoring (MRM) of the protonated molecular ions and corresponding deuterium-labelled d9-DBA derivatives resulted in selective quantifications with correlation coefficients >0.998 for the DBA derivatives of isocyanic acid (ICA), methyl isocyanate (MIC), ethyl isocyanate (EIC), propyl isocyanate (PIC), phenyl isocyanate (PhI), 1,6-hexamethylene diisocyanate (HDI), 2,4-, 2,6-toluene diisocyanate (TDI), isophorone diisocyanate (IPDI), 4,4′-methylenediphenyl diisocyanate (MDI), 3-ring MDI, 4-ring MDI, HDI-isocyanurate, HDI-diisocyanurate, HDI-biuret and HDI-dibiuret. The instrumental precision for 10 repeated injections of a solution containing 0.1 μg ml−1 of the studied derivatives was <2%. Performing MRM of the product ion [DBA + H]+ (m/z = 130) from the protonated molecular ion resulted in the lowest detection limits, down to 10 amol (for TDI). Quantification of concentrations below 10−6 of the occupational exposure limit (OEL) for TDI during 10 min of air sampling was made possible. In an effort to control the formation of alkali adducts, addition of lithium acetate to the mobile phase and monitoring of lithium adducts was evaluated. Having lithium present in the mobile phase resulted in complete domination of [M + Li]+ adducts, but detection limits for the studied compounds were not improved. Different deuterium-labelled derivatives as internal standards were evaluated. (1) DBA derivatives of deuterium-labelled isocyanates (d4-HDI, d3-2,4-TDI, d3-2,6-TDI and d2-MDI), (2) d9-DBA derivatives of the corresponding isocyanates and (3) d18-DBA derivatives of the corresponding isocyanates. We have developed a new method for the analysis of the fluoroquinolones enrofloxacin and ciprofloxacin in eggs using a diphasic dialysis procedure as extraction and purification method. High pressure liquid chromatography-mass spectrometry (HPLC-MS) was used for the confirmatory determination of these compounds. The method was found to be linear between 10 and 800 ng g−1 for enrofloxacin, and between 20 and 1600 ng g−1 for ciprofloxacin. The recovery percentages were in the 70-104% range for enrofloxacin, and 55-97% for ciprofloxacin. The assay described was repeatable and reproducible with a limit of quantitation of 2 and 4 ng g−1 for enrofloxacin and ciprofloxacin from egg, respectively. 相似文献
Electrospray ionization mass spectrometry was applied to the study of the amines IQ, Trp-P-1, Trp-P-2, PhIP and AC and the co-mutagens harman and norharman. The results obtained on a triple quadrupole mass spectrometer equipped with a pneumatically assisted electrospray source are reported. The chromatographic conditions were optimized with a reversed-phase column (1 mm I.D.) using acetonitrile-5 mM ammonium acetate (pH 6.7) (50:50) as the mobile phase at a flow-rate of 50 μl min−1. Different parameters influencing the mass spectra were investigated. For these compounds [M + H]+ in the positive-ion mode and also some fragments produced through collisionally activated decomposition in the interface were observed. Detection limits of 5.4–44 pg were obtained for standard solutions of these amines. Analysis of a meat extract was performed by HPLC-MS using single-ion monitoring after a solid-phase extraction clean-up. 相似文献
A new and simple LC-MS method for analysis of flavonoids from Sambucus ebulus berry extracts was developed and validated. Successfully were quantitated seven polyphenols: epicatechin, epigallocatechin gallate, rutin, resveratrol, myricetin, quercetin, and kaempferol.
Two detectors, working in parallel, were used: photodiode-array and single quadrupole mass-detector. The mass detection was used for identification and quantification of the analytes, while the diode-array detector was as confirmation tool. The following m/z were tracked: 457.15 (epigallocatechin gallate); 289.06 (epicatechin); 609.13 (rutin); 227.05 (resveratrol); 317.0 (myricetin); 301.02 (quercetin); 285.02 (kaempferol). For optimization the chromatographic separation three wavelengths 205?nm, 305?nm, 272?nm were monitored. The method was capable to detect in one run compounds with no UV or fluorescence chromophore and with very similar structures, such as plant polyphenols. The linearity was from 0.05?mg/L to 50?mg/L (R2 0.9962–0.9987). The recoveries for all tested analytes were between 81.6% and 104.7%.
The method was applied for analysis of crude extract of Sambucus ebulus ripe fruits. Three major polyphenols – epicatechin (0.84?mg/100gFW), quercetin (0.15?mg/100gFW) and kaempferol (0.05?mg/100gFW) were identified and quantified.
The proposed method could be successfully used for routine analysis of epigallocatechin gallate, epicatechin, rutin, resveratrol, myricetin, quercetin, and kaempferol in Sambucus ebulus extracts. 相似文献
Abstract The coupling between liquid chromatography and mass spectrometry with an APCI or ESI interface (in positive or negative mode) is used here for multi-residue analyses in natural waters, covering basic and neutral pesticides as well as acid pesticides. The methods developed are applied to drinking and, river waters after the samples are concentrated by liquid-liquid extraction or solid phase extraction on C18 cartridges. Comparisons are made between UV detection and mass spectrometry and between two chromatographic methods for acid substances. The quantitation limits range from 0.01 to 0.1 μg/l according to the substance. 相似文献
