An α-l-arabinofuranosidase from Fusarium oxysporum F3 was purified to homogeneity by a two-step ion exchange intercalated by a gel filtration chromatography. The enzyme had a molecular mass of 66 kDa and was optimally active at pH 6.0 and 60°C. It hydrolyzed aryl α-l-arabinofuranosides and cleaved arabinosyl side chains from arabinoxylan and arabinan. There was a marked synergistic effect between the α-l-arabinofuranosidase and an endo-(1 →4)-β-d-xylanase produced by F. oxysporum in the extensive hydrolysis of arabinoxylan.  相似文献

Peracetates of β-glycosyl dibenzyl phosphates are formed efficiently in the reaction of cesium dibenzyl phosphate with peracetyl-α-glycosyl nitrates derived froml-fucopyranose,d-galactopyranose, and 2-azido-2-deoxy-d-galactopyranose or with tri-O-acetyl-α-l-fucopyranosyl bromide. On the contrary, the reaction of the above-mentioned glycosyl nitrates with cesium diphenyl phosphate leads to thermodynamically more stable α-glycosyl diphenyl phosphatevia intermediate formation of the corresponding β-anomers. Drying dissipative structures of aqueous solutions of poly-d-lysine hydrobromide, poly-l-lysine hydrobromide, and their low-molecular-weight analogs were studied on a cover glass, a watch glass, and a Petri glass dish. Size of the broad rings, one of the typical macroscopic patterns, decreased as the solute concentration decreased. Microscopic drying crystal patterns of polylysine hydrobromides and their low-molecular-weight analogs, i.e., d-, l-, and dl-lysine hydrochloride changed as a function of the distance from the film center, which is one of the typical features of dissipative crystallization. Macroscopic and microscopic drying patterns were quite similar to each other irrespective of their stereospecificity. The crystal patterns of the mixtures of poly-d-lysine hydrobromide and poly-l-lysine hydrobromide were also similar to those of the corresponding single-component polymers. The kinetics of oxidation of l-lysine by diperiodatoargentate(III) (DPA) in aqueous alkaline medium at a constant ionic strength of 0.50 mol dm−3 was studied spectrophotometrically. The oxidation products are aldehyde, 5-aminopentanal and Ag(I). The main products were identified by spot test, IR and GC-MS. The stoichiometry is [l-lysine]:[DPA] = 1:1. The reaction is first order with respect to diperiodatoargentate(III) concentrations, whereas the order with respect to l-lysine and alkali concentrations changes from first order to zero order as the l-lysine and alkali concentrations are increased. The effects of added products, periodate, ionic strength, and dielectric constant of the reaction medium were investigated. Based on the experimental results, a mechanism involving complex formation between DPA species and l-lysine is proposed. The reaction constants involved in the mechanism were evaluated. The activation parameters with respect to the slow step of the mechanism were determined and discussed.  相似文献

Corynebacterium crenatum SYPA 5-5 is an aerobic and industrial l-arginine producer. It was proved that the Corynebacterium glutamicum/Escherichia coli shuttle vector pJC1 could be extended in C. crenatum efficiently when using the chloramphenicol acetyltransferase gene (cat) as a reporter under the control of promoter tac. The expression system was applied to over-express the gene vgb coding Vitreoscilla hemoglobin (VHb) to further increase the dissolved oxygen in C. crenatum. As a result, the recombinant C. crenatum containing the pJC-tac-vgb plasmid expressed VHb at a level of 3.4 nmol g−1, and the oxygen uptake rates reached 0.25 mg A562−1 h−1 which enhanced 38.8% compared to the wild-type strain. Thus, the final l-arginine concentration of the batch fermentation reached a high level of 35.9 g L−1, and the biomass was largely increased to 6.45 g L−1, which were 17.3% and 10.5% higher than those obtained by the wild-type strain, respectively. To our knowledge, this is the first report that the efficient expression system was constructed to introduce vgb gene increasing the oxygen and energy supply for l-arginine production in C. crenatum, which supplies a good strategy for the improvement of amino acid products.  相似文献

A new synthesis of l-norvaline is described. Using valeric acid as the raw material, α-brominevaleryl chloride was achieved by acyl chlorination and α-position bromination in one-pot with the yield of 84.7%. The yields of the following ammoniation of α-brominevaleryl chloride and resolution of dl-norvaline were 88.7% and 26.7%, respectively. d-Norvaline was also obtained in a similar method from the waste resolution solution. Other optical active amino acids, valine, α-aminobutyric acid and alanine were also synthesized in similar ways.  相似文献

Due to physical decay properties commonly associated with therapeutic radionuclides, 188Re (t 1/2 = 16.98 h, E max = 2.12 MeV) is of high interest for endovascular brachytherapy and endoradiotherapy in general. Rhenium precursors in the low oxidation state +I, such as the organometallic fac-[Re(H2O)3(CO)3]+ are promising lead compounds compared to those with oxidation states +III and +V since they can be prepared under mild conditions and do not tend to reoxidize to oxidation state +VII while multidentate ligands can be attached under substitution of coordinated water molecules. This study comprises the application of the Free-Ion Selective Radiotracer Extraction (FISRE) technique in order to determine dissociation rate constants of complexes bearing the [188Re(CO)3]+-core at tracer levels in vitro with regards to their time-dependent kinetic speciation. As ligands, the tridentate l-histidine as well as the dipeptides l-carnosine (β-alanyl-l-histidine) and glycyl-l-histidine were chosen in order to study the effects of different moieties attached to the primary amine of l-histidine.  相似文献

Summary. Secreted peptides from diverse sources have been found to contain a d-amino acid. From the sequence of cloned mRNAs coding for the precursors of such peptides it could be deduced that in all cases tested so far the d-amino acid in the final product is derived from the corresponding l-amino acid present in the primary product of translation. Enzymes catalyzing such an l- to d-isomerization in peptide linkage have been isolated from the venom of a spider and the skin secretions of frogs. Even though these are completely different proteins, the reaction mechanism is the same, namely a de-protonation/re-protonation of the α-carbon of an amino acid with concomitant inversion of the chirality. Sequences potentially coding for homologues of the frog enzyme are present in the genome of different vertebrate species.  相似文献

The enzyme aspartate racemase from Pyrococcus horikoshii OT3 catalyzes the interconversion between l- and d-Asp. In this work, we employed the hybrid QM/MM approach with the self-consistent charge-density functional tight binding (SCC-DFTB) model to study the catalytic mechanism for the conversion of l-Asp into d-Asp. The molecular dynamics simulation showed that the substrate l-Asp forms an extensive network of interactions with the active-site residues of the aspartate racemase through its side chain carboxylate, ammonium group, and α-carboxylate. The potential of mean force calculations confirmed that the racemization reaction involves two proton transfers (from the α-carbon to Cys194 and from Cys82 to the α-carbon), which occurs in a concerted way, although highly asynchronous. The calculated free energy of activation is 17.5 kcal/mol, which is consistent with the reaction rate measured from experiment. An electrostatic interaction analysis was performed to estimate the key role played by individual residues in stabilizing the transition state. The docking study on the binding of l-Asp and d-Asp to aspartate racemase indicates that this enzyme employs a “two-base” mechanism not a “one-base” mechanism.  相似文献

The aim of this work was to have cellulase activity and hemicellulase activity screenings of endophyte Acremonium species (Acremonium zeae EA0802 and Acremonium sp. EA0810). Both fungi were cultivated in submerged culture (SC) containing l-arabinose, d-xylose, oat spelt xylan, sugarcane bagasse, or corn straw as carbon source. In solid-state fermentation, it was tested as carbon source sugarcane bagasse or corn straw. The highest FPase, endoglucanase, and xylanase activities were produced by Acremonium sp. EA0810 cultivated in SC containing sugarcane bagasse as a carbon source. The highest β-glucosidase activity was produced by Acremonium sp. EA0810 cultivated in SC using d-xylose as carbon source. A. zeae EA0802 has highest α-arabinofuranosidase and α-galactosidase activities in SC using xylan as a carbon source. FPase, endoglucanase, β-glucosidase, and xylanase from Acremonium sp. EA0810 has optimum pH and temperatures of 6.0, 55 °C; 5.0, 70 °C; 4.5, 60 °C; and 6.5, 50 °C, respectively. α-Arabinofuranosidase and α-galactosidase from A. zeae EA0802 has optimum pH and temperatures of 5.0, 60 °C and 4.5, 45 °C, respectively. It was analyzed the application of Acremonium sp. EA0810 to hydrolyze sugarcane bagasse, and it was achieved 63% of conversion into reducing sugar and 42% of conversion into glucose.  相似文献

Summary. 2-(Arylamino)pyrimidin-4-ones were synthesized, silylated, and condensed with l,2,3,5-tetra-O-acetyl-β- d-ribofuranoside to afford the corresponding N 2-aryl protected isocytidines. Deprotection of the acetylated isocytidines using saturated NH3 in MeOH solution gave 1-(β-d-ribofuranosyl)-2-(arylamino)-4-pyrimidinones. Methyl 2-deoxy-3,5-di-O-toluyl-α/β-d-ribofuranoside was prepared and condensed with the previously silylated bases to afford the anomeric mixture of protected nucleosides. The pure β-anomers were synthesized with better yield by treating the sodium salts of N 2-arylisocytosine derivatives with 2-deoxy-3,5-di-O-toluyl-α-d-ribofuranosyl chloride. Deprotection of the latter anomers afforded the corresponding free hydroxyl derivatives. The synthesized free nucleosides are under antiviral and oligonucleotide investigations.  相似文献

We prepared poly(d,l-lactide) (PLA) microspheres by anionic dispersion polymerization of d,l-lactide. The polymerization was carried out in xylene/heptane (1:2 in v/v) mixture solution at 368 K for 9 h, with poly(dodecyl methacrylate)-co-poly[α-methacryloxyethoxy-poly(l-lactide)] (PDMA-co-P(MA-PLLA)) synthesized in this study, as a dispersion stabilizer. The number-averaged diameter and diameter distribution (coefficient of variation) of obtained PLA microspheres ranged from 180 to 800 nm and 14–40%, respectively, depending on the preparation condition. Furthermore, the time courses of monomer conversion, particle diameter, and particle number were investigated to clarify the formation mechanism of microspheres with PDMA-co-P(MA-PLLA) as a dispersion stabilizer. From this experiment, we found that the aggregation of primary particles occurred in anionic dispersion polymerization, and the particle diameter of obtained PLA microspheres decreased with increasing PDMA-co-P(MA-PLLA) concentration. In conclusion, we clarified that PDMA-co-P(MA-PLLA) effectively contributed to the stability of primary particles.  相似文献

The stoichiometric protonation constants of some α-amino acid esters (glycine methyl ester, glycine t-butyl ester, l-valine methyl ester, l-valine ethyl ester, l-valine t-butyl ester, l-serine methyl ester, l-serine ethyl ester, l-leucine methyl ester, l-leucine ethyl ester, l-leucine t-butyl ester, l-alanine methyl ester, l-alanine benzyl ester, l-phenylalanine methyl ester, l-phenylalanine ethyl ester, and l-phenylalanine t-butyl ester) in water and 20%, 40%, and 60% (v/v) 1,4-dioxane–water mixtures have been determined at an ionic strength of 0.10 mol⋅L−1 NaCl and at 25.0±0.1 °C under a nitrogen atmosphere. A potentiometric method was used and the calculation of the protonation constants has been carried out using the BEST computer program. The results were discussed in terms of macroscopic properties of the mixed solvent. The stoichiometric protonation constants were influenced by changes in solvent composition and their variations were discussed in terms of preferential solvation. Also, knowledge the protonation constant of α-amino acid esters will be helpful when determining the microscopic equilibrium constants of their corresponding amino acids.  相似文献

Two new polar steroidal glycosides identified as sodium (20R,22E,24R,25S)-3-O-(β-d-xylopyranosyl)-24-methyl-5α-cholest-22-ene-3β,6β,8,15α,26-pentol 26-sulfate (evasterioside A) and sodium (20R,22E)-24-O-(β-d-xylopyranosyl)-5α-cholest-22-ene-3β,6β,8,15α,24-pentol 3-sulfate (evasterioside B) were isolated from the Pacific starfish Evasterias retifera collected in the Sea of Japan. Five known compounds, viz., coscinasterioside B, aphelasterioside A, marthasterone 3-sulfate and (20R)-cholest-7-en-3β-ol and cholesterol sulfates, were identified. The structures of the new natural compounds were established using their 2D NMR and mass spectra and some chemical transformations.  相似文献

Compound 1 dissolved both in distilled water and chloroform but compound 2 dissolved in distilled water not in chloroform, though compounds 1 and 2 consist of 4 tri-O-methyl-glucopyranosyl and 2 unmodified anhydro glucopyranosyl units.  相似文献

Catalytically efficient β-d-xylosidase from Selenomonas ruminantium (SXA) exhibits pK as 5 and 7 (assigned to catalytic base, D14, and catalytic acid, E186) for k cat/K m with substrates 1,4-β-d-xylobiose (X2) and 1,4-β-d-xylotriose (X3). Catalytically inactive, dianionic SXA (D14E186) has threefold lower affinity than catalytically active, monoanionic SXA (D14E186H) for X2 and X3, whereas D14E186 has twofold higher affinity than D14E186H for 4-nitrophenyl-β-d-xylopyranoside (4NPX), and D14E186 has no affinity for 4-nitrophenyl-α-l-arabinofuranoside. Anomeric isomers, α-d-xylose and β-d-xylose, have similar affinity for SXA. 4-Nitrophenol competitively inhibits SXA-catalyzed hydrolysis of 4NPX. SXA steady-state kinetic parameters account for complete progress curves of SXA-catalyzed hydrolysis reactions.  The apparent molar volume and the apparent molar adiabatic compressibility of the sugar-based surfactants methyl 6-O-octanoyl-α-d-glucopyranoside (α-MOnG), methyl 6-O-octanoyl-β-d-glucopyranoside (β-MOnG), and octyl β-d-glucoside were measured over a wide concentration range. Also, the aggregation number of their micelles was determined from the Debye plot using static light scattering data. It was found that the micellar aggregation number for α-MOnG is 179 at 35 °C, which is 1.5 times larger than that for β-MOnG, suggesting that the anomerism of the head group influences the packing of the monomers during micelle formation.
