Optically active polymers bearing chiral units at the side chain were prepared via reversible addition‐fragmentation chain transfer (RAFT) polymerization in the presence of 2,2′‐azobisisobutyronitrile (AIBN)/benzyl dithiobenzoate (BDB), using a synthesized 6‐Op‐vinylbenzyl‐1,2:3,4‐Di‐O‐isopropylidene‐D ‐galactopyranose (VBPG) as the monomer. The experimental results suggested that the polymerization of the monomer proceeded in a living fashion, providing chiral group polymers with narrow molecular weight distributions. The optically active nature of the obtained poly (6‐Op‐vinylbenzyl‐1,2:3,4‐Di‐O‐isopropylidene‐D ‐galactopyranose) (PVBPG) was studied by investigating the dependence of specific rotation on the molecular weight of PVBPG and the concentration of PVBPG in tetrahydrofuran (THF). The results showed the specific rotation of PVBPG increased greatly with the decrease of the concentration of the PVBPG homopolymer. In addition, the effect of block copolymers of PVBPG on the optically active nature was also investigated by preparing a series of diblock copolymers of poly(methyl methacrylate) (PMMA)‐b‐PVBPG, polystyrene (PS)‐b‐PVBPG, and poly(methyl acrylate) (PMA)‐b‐PVBPG. It was found that both the homopolymer and the diblock copolymers possessed specific rotations. Finally, the ability of chiral recognition of the PVBPG homopolymer was investigated via an enantiomer‐selective adsorption experiment. © 2007 Wiley Periodicals, Inc. Soluble hyperbranched glycopolymers were prepared by copolymerization of glycan monomers with reversible addition‐fragmentation chain transfer polymerization (RAFT) inimers in a simple one‐pot reaction. Two novel RAFT inimers, 2‐(methacryloyloxy)ethyl 4‐cyano‐4‐(phenylcarbonothioylthio)pentanoate (MAE‐CPP) and 2‐(3‐(benzylthiocarbonothioylthio)propanoyloxy)ethyl acrylate (BCP‐EA) were synthesized and used to prepare hyperbranched glycopolymers. Two types of galactose‐based saccharide monomers, 6‐O‐methacryloyl‐1,2:3,4‐di‐O‐isopropylidene‐D ‐galactopyranose (proGal‐M) and 6‐O‐(2′‐acrylamido‐2′‐methylpropanoate)‐1,2:3,4‐di‐O‐isopropylidene‐D ‐galactopyranose (proGal‐A), containing a methacrylate and an acrylamide group, respectively, were also synthesized and polymerized under the mediation of the MAE‐CPP and BCP‐EA inimers, respectively. In addition, hyperbranched poly(proGal‐M), linear poly(proGal‐A), and hyperbranched poly(proGal‐A) were generated and their polymerization kinetics were studied and compared. An unexpected difference was observed in the kinetics between the two monomers during polymerization: the relationship between polymerization rate and concentration of inimer was totally opposite in the two monomer–inimer systems. Branching analysis was conducted by using degree of branching (DB) as the measurement parameter. As expected, a higher DB occurred with increased inimer content. Furthermore, these polymers were readily deprotected by hydrolysis in trifluoroacetic acid solution resulting in water‐soluble polymers. The resulting branched glycopolymers have potential as biomimetics of polysaccharides. © 2012 Wiley Periodicals, Inc. J Polym Sci Part A: Polym Chem, 2012  相似文献

Poly(benzyl‐L ‐glutamate) (PBLG) macromonomers were synthesized by N‐carboxyanhydride (NCA) polymerization initiated with 4‐vinyl benzylamine. MALDI‐ToF analysis confirmed the presence of styrenic end‐groups in the PBLG. Free‐radical and RAFT polymerization of the macromonomer in the presence of divinyl benzene produced star polymers of various molecular weights, polydispersity, and yield depending on the reaction conditions applied. The highest molecular weight (Mw) of 10,170,000 g/mol was obtained in a free‐radical multibatch approach. It was shown that the PBLG star polymers can be deprotected to obtain poly(glutamic acid) star polymers, which form water soluble pH responsive nanoparticles. © 2010 Wiley Periodicals, Inc. We prepared well‐defined diblock copolymers of thermoresponsive poly(N‐isopropylacrylamide‐coN,N‐dimethylacrylamide) blocks and biodegradable poly(D ,L ‐lactide) blocks by combination of reversible addition‐fragmentation chain transfer radical (RAFT) polymerization and ring‐opening polymerization. α‐Hydroxyl, ω‐dithiobenzoate thermoresponsive polymers were synthesized by RAFT polymerization using hydroxyl RAFT agents. Biodegradable blocks were prepared by ring‐opening polymerization of D ,L ‐lactide initiated by α‐hydroxyl groups of thermoresponsive polymers, which inhibit the thermal decomposition of ω‐dithioester groups. Terminal dithiobenzoate (DTBz) groups of thermoresponsive blocks were easily reduced to thiol groups and reacted with maleimide (Mal). In aqueous media, diblock copolymer products formed surface‐functionalized thermoresponsive micelles. These polymeric micelles had a low critical micelle concentration of 22 μg/L. In thermoresponsive studies of the micelles, hydrophobic DTBz‐surface micelles demonstrated a significant shift in lower critical solution temperature (LCST) to a lower temperature of 30.7 °C than that for Mal‐surface micelles (40.0 °C). In addition, micellar LCST was controlled by changing bulk mixture ratios of respective heterogeneous end‐functional diblock copolymers. Micellar disruption at acidic condition (pH 5.0) was completed within 5 days due to hydrolytic degradation of PLA cores, regardless of showing a slow disruption rate at physiological condition. Furthermore, we successfully improved water‐solubility of hydrophobic drug, paclitaxel by incorporating into the micellar cores. © Wiley Periodicals, Inc. A reversible addition‐fragmentation chain transfer (RAFT) agent was directly anchored onto Fe3O4 nanoparticles in a simple procedure using a ligand exchange reaction of S‐1‐dodecyl‐S′‐(α,α′‐dimethyl‐α″‐acetic acid)trithiocarbonate with oleic acid initially present on the surface of pristine Fe3O4 nanoparticles. The RAFT agent‐functionalized Fe3O4 nanoparticles were then used for the surface‐initiated RAFT copolymerization of N‐isopropylacrylamide and acrolein to fabricate structurally well‐defined hybrid nanoparticles with reactive and thermoresponsive poly(N‐isopropylacrylamide‐co‐acrolein) shell and magnetic Fe3O4 core. Evidence of a well‐controlled surface‐initiated RAFT copolymerization was gained from a linear increase of number‐average molecular weight with overall monomer conversions and relatively narrow molecular weight distributions of the copolymers grown from the nanoparticles. The resulting novel magnetic, reactive, and thermoresponsive core‐shell nanoparticles exhibited temperature‐trigged magnetic separation behavior and high ability to immobilize model protein BSA. © 2009 Wiley Periodicals, Inc. We report here the synthesis of well‐defined homopolymer bearing amino acid diamide, poly(N‐acryloyl‐L ‐valine N′‐methylamide), via reversible addition fragmentation chain transfer (RAFT) polymerization using alkynyl‐functionalized 2‐dodecylsulfanylthiocarbonylsulfanyl‐2‐methyl‐propionic acid propargyl alcohol ester as chain transfer agent (CTA) and 2,2′‐azobis(isobutyronitrile) as initiator. The effects of a variety of parameters, such as temperature and solvent, on RAFT polymerization were examined to determine the optimal control of the polymerization. The controlled nature of RAFT polymerization was evidenced by the controllable molecular weight and low‐molecular‐weight polydispersity index (Mw/Mn) of resulting homopolymers and further demonstrated to have retained end‐group functionality by the fact of the successful formation of block copolymers from further RAFT polymerization by using the resultant polymer as macro‐CTA, as well as from “click” chemistry. Thermoresponsive property of the prepared polymer was evaluated in terms of the lower critical solution temperature in aqueous solution by measuring the transmittance variation at 500 nm from UV/vis spectroscopy. © 2010 Wiley Periodicals, Inc. Four kinds of bio‐based polyurethanes bearing hydroxy groups in the pendants were synthesized by the polyaddition of D ‐mannitol‐ and D,L ‐erythritol‐derived diols (1,2:5,6‐di‐O‐isopropylidene‐D ‐mannitol and 1,2‐O‐isopropylidene‐D,L ‐erythritol) with hexamethylene diisocyanate and methyl (S)‐2,6‐diisocyanatohexanoate and the subsequent deprotection of the isopropylidene groups. They were hydrolyzed much more quickly than the corresponding protected polyurethanes at 50 °C and pH 7.0, although their hydrolytic degradation rate was lower than that of polyurethanes with saccharic and glucuronic lactone groups, which had been reported in our previous articles. The introduction of D ‐mannitol units to the polyether‐polyurethanes containing poly(oxytetramethylene) glycol units also enhanced their hydrolyzibility. © 2010 Wiley Periodicals, Inc. pH‐Sensitive block glycopolymers of poly(2‐(diethylamino)ethyl methacrylate) (PDEAEMA) and poly(3‐O‐methacryloy‐α,β‐D ‐glucopyranose) (PMAGlc) were synthesized via reversible addition–fragmentation chain transfer (RAFT) radical polymerization based on protected glycomonomer 3‐O‐methacryloyl‐1,2:5,6‐di‐O‐isopropylidene‐D ‐glucofuranose (MAIpGlc). It was found that RAFT homopolymerization of MAIpGlc proceeded in a controlled fashion with 4‐cyanopentanoic acid dithiobenzoate as chain transfer agent. Using the dithioester‐capped PDEAEMA as macro‐RAFT agent, block copolymerization of MAIpGlc was in good control as indicated by the linear pseudo first‐order kinetic plot, the linear increment of number‐average molecular weights as well as narrow and symmetrical gel permeation chromatography peaks, and low polydispersities. Well‐defined diblock copolymers of DEAEMA and MAIpGlc were prepared successfully through the chain extension of PDEAEMA. The deprotection of MAIpGlc units in trifluoroacetic acid/H2O solution afforded PDEAEMA‐b‐PMAGlc block glycopolymer. The self‐assembly behavior of PDEAEMA‐b‐PMAGlc in aqueous solution was investigated by using 1H NMR, UV‐vis spectroscopy, dynamic light scattering, and transmission electron microscopy. The results demonstrated that spherical micelles with PDEAEMA as the hydrophobic cores and PMAGlc as the hydrophilic shells were formed in alkaline aqueous solution. These glucose‐installed micelles had specific recognition with Concanavalin A. The combination of pH‐sensitivity of PDEAEMA and biomolecular recognition of PMAGlc in one micellar system may create a multifunctional platform for targeted delivery, biomimetics, and biodection. © 2010 Wiley Periodicals, Inc. Reversible addition fragmentation chain transfer (RAFT) polymerization and bifunctional sparteine/thiourea organocatalyst‐mediated ring opening polymerization (ROP) were combined to produce poly(L ‐lactide) star polymers and poly(L ‐lactide‐co‐styrene) miktoarm star copolymers architecture following a facile experimental procedure, and without the need for specialist equipment. RAFT was used to copolymerize ethyl acrylate (EA) and hydroxyethyl acrylate (HEA) into poly(EA‐co‐HEA) co‐oligomers of degree of polymerization 10 with 2, 3, and 4 units of HEA, which were in turn used as multifunctional initiators for the ROP of L ‐lactide, using a bifunctional thiourea organocatalytic system. Furthermore, taking advantage of the living nature of RAFT polymerization, the multifunctional initiators were chain extended with styrene (poly((EA‐co‐HEA)‐b‐styrene) copolymers), and used as initiators for the ROP of L ‐lactide, to yield miktoarm star copolymers. The ROP reactions were allowed to proceed to high conversions (>95%) with good control over molecular weights (ca. 28,000‐230,000 g/mol) and polymer structures being observed, although the molecular weight distributions are generally broader (1.3–1.9) than those normally observed for ROP reactions. The orthogonality of both polymerization techniques, coupled with the ubiquity of HEA, which is used as a monomer for RAFT polymerization and as an initiator for ROP, offer a versatile approach to star‐shaped copolymers. Furthermore, this approach offers a practical approach to the synthesis of polylactide star polymers without a glove box or stringent reaction conditions. The phase separation properties of the miktoarm star copolymers were demonstrated via thermal analyses. © 2009 Wiley Periodicals, Inc. J Polym Sci Part A: Polym Chem 47: 6396–6408, 2009  相似文献

Two types of three‐arm and four‐arm, star‐shaped poly(D,L ‐lactic acid‐alt‐glycolic acid)‐b‐poly(L ‐lactic acid) (D,L ‐PLGA50‐b‐PLLA) were successfully synthesized via the sequential ring‐opening polymerization of D,L ‐3‐methylglycolide (MG) and L ‐lactide (L ‐LA) with a multifunctional initiator, such as trimethylolpropane and pentaerythritol, and stannous octoate (SnOct2) as a catalyst. Star‐shaped, hydroxy‐terminated poly(D,L ‐lactic acid‐alt‐glycolic acid) (D,L ‐PLGA50) obtained from the polymerization of MG was used as a macroinitiator to initiate the block polymerization of L ‐LA with the SnOct2 catalyst in bulk at 130 °C. For the polymerization of L ‐LA with the three‐arm, star‐shaped D,L ‐PLGA50 macroinitiator (number‐average molecular weight = 6800) and the SnOct2 catalyst, the molecular weight of the resulting D,L ‐PLGA50‐b‐PLLA polymer linearly increased from 12,600 to 27,400 with the increasing molar ratio (1:1 to 3:1) of L ‐LA to MG, and the molecular weight distribution was rather narrow (weight‐average molecular weight/number‐average molecular weight = 1.09–1.15). The 1H NMR spectrum of the D,L ‐PLGA50‐b‐PLLA block copolymer showed that the molecular weight and unit composition of the block copolymer were controlled by the molar ratio of L ‐LA to the macroinitiator. The 13C NMR spectrum of the block copolymer clearly showed its diblock structures, that is, D,L ‐PLGA50 as the first block and poly(L ‐lactic acid) as the second block. © 2001 John Wiley & Sons, Inc. Thermoresponsive double hydrophilic diblock copolymers poly(2‐(2′‐methoxyethoxy)ethyl methacrylate‐co‐oligo(ethylene glycol) methacrylate)‐b‐poly(6‐O‐methacryloyl‐D ‐galactopyranose) (P(MEO2MA‐co‐OEGMA)‐b‐PMAGP) with various compositions and molecular weights were obtained by deprotection of amphiphilic diblock copolymers P(MEO2MA‐co‐OEGMA)‐b‐poly(6‐O‐methacryloyl‐1,2:3,4‐di‐O‐isopropylidene‐D ‐galactopyranose) (P(MEO2MA‐co‐OEGMA)‐b‐PMAlpGP), which were prepared via reversible addition‐fragmentation chain transfer (RAFT) polymerization using P(MEO2MA‐co‐OEGMA) as macro‐RAFT agent. Dynamic light scattering and UV–vis studies showed that the micelles self‐assembled from P(MEO2MA‐co‐OEGMA)‐b‐PMAlpGP were thermoresponsive. A hydrophobic dye Nile Red could be encapsulated by block copolymers P(MEO2MA‐co‐OEGMA)‐b‐PMAGP upon micellization and released upon dissociation of the formed micelles under different temperatures. The galactose functional groups in the PMAGP block have specific interaction with HepG2 cells, and P(MEO2MA‐co‐OEGMA)‐b‐PMAGP has potential applications in hepatoma‐targeting drug delivery and biodetection. © 2011 Wiley Periodicals, Inc. Amphiphilic double‐brush copolymers (DBCs) with each graft site quantitatively carrying both a hydrophilic poly(ethylene oxide) (PEO) graft and a hydrophobic polystyrene (PSt) graft are synthesized by sequential reversible addition‐fragmentation chain transfer (RAFT) polymerization and ring‐opening metathesis polymerization (ROMP). These DBCs are used as both surfactants and polyfunctional RAFT agents in the radical polymerization of St in aqueous dispersed media. Miniemulsions with narrowly dispersed St‐based nanodroplets are readily obtained after ultrasonication of the reaction mixtures. Without the presence of crosslinker, chain‐extension polymerization of St from the DBCs yields well‐defined polymeric latexes with narrow size distributions. However, with the presence of divinylbenzene (DVB) as the crosslinker, vesicular polymeric nanoparticles are formed as the major product. Such crosslinking‐induced change in morphology of the resulting latex nanomaterials may be ascribed to the increase of interfacial curvature in the heterophase systems during crosslinking polymerization. © 2014 Wiley Periodicals, Inc. The amino acid tryptophan has been converted into acrylamide monomers using L /D ‐tryptophan methyl ester forming the enantiopure chiral monomers. Attempts were made to polymerize these monomers via reversible addition fragmentation chain transfer (RAFT) polymerization to form poly(tryptophan). Unfortunately, this proved difficult, and instead, a postpolymerization modification route was used by first synthesizing poly(pentafluorophenyl acrylate) via RAFT, which was then substituted with L ‐tryptophan methyl ester to give poly(L ‐tryptophan). The interactions of the newly synthesized tryptophan monomers, as well as previously reported phenylalanine monomers, were studied in the presence of rac‐BINOL. It has been shown that the enantiomers of tryptophan have a stronger interaction with BINOL than phenylalanine and this has been attributed to the larger π system on the side chain. By monitoring the shifts and splitting of the phenolic protons of BINOL, it has been observed that S‐BINOL interacts more favorably with L ‐monomer enantiomers and R‐BINOL with D ‐monomer enantiomers. Similar interactions have also been seen with poly(phenylalanine) and the newly synthesized poly(tryptophan) materials. © 2012 Wiley Periodicals, Inc. J Polym Sci Part A: Polym Chem, 2012  相似文献

Amphiphilic, biocompatible poly(N‐vinylpyrrolidone)‐b‐poly(l ‐lactide) (PVP‐b‐PLLA) block polymers were synthesized at 60 °C using a hydroxyl‐functionalized N,N‐diphenyldithiocarbamate reversible addition–fragmentation chain transfer (RAFT) agent, 2‐hydroxyethyl 2‐(N,N‐diphenylcarbamothioylthio)propanoate (HDPCP), as a dual initiator for RAFT polymerization and ring‐opening polymerization (ROP) in a one‐step procedure. 4‐Dimethylamino pyridine was used as the ROP catalyst for l ‐lactide. The two polymerization reactions proceeded in a controlled manner, but their polymerization rates were affected by the other polymerization process. This one‐step procedure is believed to be the most convenient method for synthesizing PVP‐b‐PLLA block copolymers. HDPCP can also be used for the one‐step synthesis of poly(N‐vinylcarbazole)‐b‐PLLA block copolymers. © 2014 Wiley Periodicals, Inc. J. Polym. Sci., Part A: Polym. Chem. 2014 , 52, 1607–1613  相似文献

Aqueous RAFT polymerization of N‐isopropylacrylamide (NIPAM) mediated with hydrophilic macro‐RAFT agent is generally used to prepare poly(N‐isopropylacrylamide) (PNIPAM)‐based block copolymer. Because of the phase transition temperature of the block copolymer in water being dependent on the chain length of the PNIPAM block, the aqueous RAFT polymerization is much more complex than expected. Herein, the aqueous RAFT polymerization of NIPAM in the presence of the hydrophilic macro‐RAFT agent of poly(dimethylacrylamide) trithiocarbonate is studied and compared with the homogeneous solution RAFT polymerization. This aqueous RAFT polymerization leads to the well‐defined poly(dimethylacrylamide)‐b‐poly(N‐isopropylacrylamide)‐b‐poly(dimethylacrylamide) (PDMA‐b‐PNIPAM‐b‐PDMA) triblock copolymer. It is found, when the triblock copolymer contains a short PNIPAM block, the aqueous RAFT polymerization undergoes just like the homogeneous one; whereas when the triblock copolymer contains a long PNIPAM block, both the initial homogeneous polymerization and the subsequent dispersion polymerization are involved and the two‐stage ln([M]o/[M])‐time plots are indicated. The reason that the PNIPAM chain length greatly affects the aqueous RAFT polymerization is discussed. The present study is anticipated to be helpful to understand the chain extension of thermoresponsive block copolymer during aqueous RAFT polymerization. © 2013 Wiley Periodicals, Inc. Core‐shell structured nanoparticles of poly(ethylene glycol) (PEG)/polypeptide/poly(D ,L ‐lactide) (PLA) copolymers were prepared and their properties were investigated. The copolymers had a poly(L ‐serine) or poly(L ‐phenylalanine) block as a linker between a hydrophilic PEG and a hydrophobic PLA unit. They formed core‐shell structured nanoparticles, where the polypeptide block resided at the interface between a hydrophilic PEG shell and a hydrophobic PLA core. In the synthesis, poly(ethylene glycol)‐b‐poly(L ‐serine) (PEG‐PSER) was prepared by ring opening polymerization of N‐carboxyanhydride of O‐(tert‐butyl)‐L ‐serine and subsequent removal of tert‐butyl groups. Poly(ethylene glycol)‐b‐poly(L ‐phenylalanine) (PEG‐PPA) was obtained by ring opening polymerization of N‐carboxyanhydride of L ‐phenylalanine. Methoxy‐poly(ethylene glycol)‐amine with a MW of 5000 was used as an initiator for both polymerizations. The polymerization of D ,L ‐lactide by initiation with PEG‐PSER and PEG‐PPA produced a comb‐like copolymer, poly(ethylene glycol)‐b‐[poly(L ‐serine)‐g‐poly(D ,L ‐lactide)] (PEG‐PSER‐PLA) and a linear copolymer, poly(ethylene glycol)‐b‐poly(L ‐phenylalanine)‐b‐poly(D ,L ‐lactide) (PEG‐PPA‐PLA), respectively. The nanoparticles obtained from PEG‐PPA‐PLA showed a negative zeta potential value of ?16.6 mV, while those of PEG‐PSER‐PLA exhibited a positive value of about 19.3 mV. In pH 7.0 phosphate buffer solution at 36 °C, the nanoparticles of PEG/polypeptide/PLA copolymers showed much better stability than those of a linear PEG‐PLA copolymer having a comparable molecular weight. © 2011 Wiley Periodicals, Inc. A reversible addition‐fragmentation chain transfer (RAFT) agent was directly anchored onto superparamagnetic Fe3O4 nanoparticles (SPNPs) in a simple procedure using a ligand exchange reaction of 2‐[(dodecylsulfanylcarbonylthiolsulfanyl) propionic acid] (DCPA) with oleic acid initially present on the surface of Fe3O4 nanoparticles. The DCPA‐modified SPNPs were then used for the surface‐mediated RAFT polymerization of di(ethylene glycol) ethyl ether acrylate and (oligoethylene glycol) methyl ether acrylate to fabricate structurally well‐defined hybrid SPNPs with temperature‐responsive poly[di(ethylene glycol) ethyl ether acrylate‐co‐(oligoethylene glycol) methyl ether acrylate] shell and magnetic Fe3O4 core. Evidence of a well‐controlled surface‐mediated RAFT polymerization was gained from a linear increase of number‐average molecular weight with overall monomer conversions and relatively narrow polydispersity indices of the copolymers grown from the SPNPs. The resultant hybrid nanoparticles exhibited superparamagnetic property with a saturation magnetization of 55.1–19.4 emu/g and showed a temperature‐responsive phenomenon as the temperature changed between 25 and 40 °C. © 2013 Wiley Periodicals, Inc. The micellar macro‐RAFT agent‐mediated dispersion polymerization of styrene in the methanol/water mixture is performed and synthesis of temperature‐sensitive ABC triblock copolymer nanoparticles is investigated. The thermoresponsive diblock copolymer of poly(N,N‐dimethylacrylamide)‐block‐poly[N‐(4‐vinylbenzyl)‐N,N‐diethylamine] trithiocarbonate forms micelles in the polymerization solvent at the polymerization temperature and, therefore, the dispersion RAFT polymerization undergoes as similarly as seeded dispersion polymerization with accelerated polymerization rate. With the progress of the RAFT polymerization, the molecular weight of the synthesized triblock copolymer of poly(N,N‐dimethylacrylamide)‐block‐poly[N‐(4‐vinylbenzyl)‐N,N‐diethylamine]‐b‐polystyrene linearly increases with the monomer conversion, and the PDI values of the triblock copolymers are below 1.2. The dispersion RAFT polymerization affords the in situ synthesis of the triblock copolymer nanoparticles, and the mean diameter of the triblock copolymer nanoparticles increases with the polymerization degree of the polystyrene block. The triblock copolymer nanoparticles contain a central thermoresponsive poly [N‐(4‐vinylbenzyl)‐N,N‐diethylamine] block, and the soluble‐to‐insoluble ‐‐transition temperature is dependent on the methanol content in the methanol/water mixture. © 2014 Wiley Periodicals, Inc. Well‐defined macromolecular brushes with poly(N‐isopropyl acrylamide) (PNIPAM) side chains on random copolymer backbones were synthesized by “grafting from” approach based on click chemistry and reversible addition‐fragmentation chain transfer (RAFT) polymerization. To prepare macromolecular brushes, two linear random copolymers of 2‐(trimethylsilyloxy)ethyl methacrylate (HEMA‐TMS) and methyl methacrylate (MMA) (poly(MMA‐co‐HEMA‐TMS)) were synthesized by atom transfer radical polymerization and were subsequently derivated to azide‐containing polymers. Novel alkyne‐terminated RAFT chain transfer agent (CTA) was grafted to polymer backbones by copper‐catalyzed 1,3‐dipolar cycloaddition (azide‐alkyne click chemistry), and macro‐RAFT CTAs were obtained. PNIPAM side chains were prepared by RAFT polymerization. The macromolecular brushes have well‐defined structures, controlled molecular weights, and molecular weight distributions (Mw/Mn ≦ 1.23). The RAFT polymerization of NIPAM exhibited pseudo‐first‐order kinetics and a linear molecular weight dependence on monomer conversion, and no detectable termination was observed in the polymerization. A poly(D,L ‐lactide)–bromine macroinitiator was synthesized for use in the preparation of a novel biocompatible polymer. This amphiphilic diblock copolymer consisted of biodegradable poly(D,L ‐lactide) and 2‐methacryloyloxyethyl phosphorylcholine and was formed by atom transfer radical polymerization. Polymeric nanoparticles were prepared by a dialysis process in a select solvent. The shape and structure of the polymeric nanoparticles were determined by 1H NMR, atomic force microscopy, and ζ‐potential measurements. The results of cytotoxicity tests showed the good cytocompatibility of the lipid‐like diblock copolymer poly(2‐methacryloyloxyethyl phosphorylcholine)‐block‐poly(D,L ‐lactide). © 2006 Wiley Periodicals, Inc.
