Eggs of the grape vine moth, Lobesia botrana (Denis and Schiffermuller), ranging in age from 1–24 to 73–96 h, were exposed, at 24 h intervals, to gamma radiation ranging from 25–600 Gy. The effects of gamma radiation on egg hatch, pupation, adult emergence, sex ratio and rate of development were examined. Results showed that the radiosensitivity of the grape vine moth eggs decreased with increasing age and increased with increasing radiation dose. Egg hatch in 1–24 h old eggs was significantly affected at 25 Gy and completely prevented at 100 Gy. At the age of 25–48 h, radiation sensitivity was only a little lower; egg hatch at 100 Gy was <1% and at 125 Gy no egg hatch was observed. Egg sensitivity to gamma irradiation decreased significantly in the 49–72 h age group; egg hatch was 66% at 100 Gy, and 500 Gy did not completely stop egg hatch (<1%). Eggs irradiated a few hours before egg hatch (73–96 h old) were the most resistant; 150 Gy had no significant effect on egg hatch and at 600 Gy over 33% of the eggs hatched. When pupation or adult emergence was used as a criterion for measuring effectiveness, however, the effects of gamma radiation were very severe. In the most resistant age group (73–96 h old), 150 Gy completely prevented pupation and adult emergence and all larvae resulting from eggs irradiated <49 h old died before pupation. In addition, the rate of development of immature stages resulting from irradiated eggs was negatively affected and sex ratio was skewed in favor of males.  相似文献

UV radiation (280–400 nm) is known to affect phytoplankton in negative, neutral and positive ways depending on the species or levels of irradiation energy. However, little has been documented on how photosynthetic physiology and growth of red tide alga respond to UVR in a long-term period. We exposed the cells of the marine red tide diatom Skeletonema costatum for 6 days to simulated solar radiations with UV-A (320–400 nm) or UV-A + UV-B (295–400 nm) and examined their changes in photosynthesis and growth. Presence of UV-B continuously reduced the effective photosynthetic quantum yield of PSII, and resulted in complete growth inhibition and death of cells. When UV-B or UV-B + UV-A was screened off, the growth rate decreased initially but regained thereafter. UV-absorbing compounds and carotenoids increased in response to the exposures with UVR. However, mechanisms for photoprotection associated with the increased carotenoids or UV-absorbing compounds were not adequate under the continuous exposure to a constant level of UV-B (0.09 W m?2, DNA-weighted). In contrast, under solar radiation screened off UV-B, the photoprotection was first accomplished by an initial increase of carotenoids and a later increase in UV-absorbing compounds. The overall response of this red tide alga to prolonged UV exposures indicates that S. costatum is a UV-B-sensitive species and increased UV-B irradiance would influence the formation of its blooms.  相似文献

A laboratory study was conducted to determine the effect of three extraction parameters (soaking time, extraction time and the ratio of solid to liquid) on the yield and chemical composition of Foeniculum vulgare seeds essential oils. The bioactivity of the essential oil extracted for the optimum extraction parameters was assessed against Culex pipiens mosquito. F. vulgare essential oil composition included large amounts of phenylpropanoids. Through an extraction time of 6 h and a ratio solid to liquid of 300 g/L we can get over than 72% of trans-anethol without soaking the seeds. With bioassays, essential oils showed different activities on C. pipiens larvae and pupae. Results show that a concentration at 40 mg/L was sufficient to register 50% mortality for the second instars larvae and this, after 2 h exposition time. Moreover, concentration at 60 mg/L ensured after 4 h exposition time 90% mortality for the fourth instars larvae. However, pupae needed 24 h exposition time to show promising mortalities when using concentration at 200 mg/L. Even if laboratory bioassays are only the first step towards the use of essential oils in practical applications, these substances represent a potential alternative to chemical insecticides in some markets.  相似文献

An ‘easy-on, easy-off’ process for the effective resolution of (±)-1-phenylethylamine was designed using the lipase B of Candida antarctica. This two step lipase-catalyzed process for the resolution of a chiral arylalkylamine involves a high-conversion enantioselective condensation of (R)-(+)-1-phenylethylamine with capric acid (conversion 99%, <24 h), followed by the hydrolysis of the corresponding synthesized (R)-(+)-amide (conversion 98%, 48 h). As a result, this efficient enzymatic process yields both (R)- and (S)-enantiomers of 1-phenylethylamine in high enantiomeric purity.  相似文献

Weakly-agglomerated nanocrystalline (ZrO2)1−x(Yb2O3)x (x=0.02–0.2) powders with high surface area (109–151 m2 g−1) were synthesized by a two-step hydrothermal process in the presence of urea: a stock solution of metal nitrates and urea was heated at 80 °C for 24 h and then at 180 °C for 48 h. For x=0.04–0.2, the as-derived powders were an assembly of uniform nanoparticles with well-defined edges in the size between 6.1–8.4 nm. Before and after calcination at 800 °C, the lattice parameters, microstrain and surface area of the (ZrO2)1−x(Yb2O3)x samples tended to increase with Yb3+ concentration; while, the average crystallite size decreased correspondingly. In the Arrhenius plots over the measurement temperature range of 400–800 °C, the bulk ionic conductivity of the compacts sintered at 1400 °C for 24 h showed a maximum value at the composition of x=0.08 in cubic structure, with an activation energy of 0.89 eV. The UVA-irradiation of 10 J/cm2 on HaCaT keratinocytes increased 59.1% of the intracellular reactive oxygen species (ROS) by NBT assay and the cell viability decreased to 31.5% by WST-1 assay, comparing to the non-irradiated control. In the presence of fullerene-C60 (C60) incorporated in phospholipid membrane vehicle (LiposomeFullerene: Lpsm-Flln) of 250–500 ppm, they were restored to ?9.1% to  + 2.3% of the ROS and 83.0–84.8% of the cell viability, but scarcely restored by the liposome without C60 (Lpsm). In HaCaT cells administered with Lpsm-Flln (150 ppm), C60 was ingested at the intracellular concentrations of 1.4–21.9 ppm for 4–24 h, and, intracellular C60 was excreted by 80% at 4 h after rinsing-out, and decreased to 2–10% after 24–48 h. C60 was predominantly distributed around the outside of nuclear membrane without deterioration of intact cell morphology according to fluorescent immunostain. Thus Lpsm-Flln is found to be an effective antioxidant that could preserve HaCaT keratinocytes against UVA-induced cellular injury. Lpsm-Flln has a potential to serve as a cosmetic material for skin protection against UVA.  相似文献

Chiral Brønsted acids (R)- and (S)-BINOL were employed as additives in the classic l-proline catalyzed direct aldol reaction. Eighteen substrates were tested with 0.5 mol % (R)-BINOL loading and 1 mol % of (S)-BINOL loading, and the enantioselectivity was improved from 72% ee without additive to 98% ee. In the proposed transition state, the chiral Brønsted acid promoted the reaction through hydrogen bonding, which not only activated the carbonyl group but also stabilized the transition state.  相似文献

The purpose of this study was to evaluate microbial populations, Hunter's color values (L?, a?, b?) and the sensory quality of freeze-dried miyeokguk, Korean seaweed soup, in order to use it as space food. Microorganisms were not detected in non-irradiated freeze-dried miyeokguk within the detection limit of 1.00 log CFU/g. However, the microbial population in rehydrated miyeokguk was 7.01 log CFU/g after incubation at 35 °C for 48 h, indicating that freeze-dried miyeokguk was not sterilized by heat treatment during the preparation process. Bacteria in the freeze-dried miyeokguk were tentatively identified as Bacillus cereus, B. subtilis, Enterobacter hormaechei, and Ancinetobacter genomosp. using the 16S rDNA sequencing. In samples that were gamma-irradiated above 10 kGy, it was confirmed that all microorganisms were inactivated. Hunter's color values of the samples irradiated at doses less than 10 kGy were not significantly altered from their baseline appearance (p>0.05). Sensory evaluation showed that preference scores in all sensory properties decreased when freeze-dried miyeokguk was irradiated at doses greater than 10 kGy. Therefore, the results of this study suggest that gamma irradiation at 10 kGy is sufficient to sterilize freeze-dried miyeokguk without significant deterioration in the sensory quality, and thus, the freeze-dried and irradiated miyeokguk at 10 kGy fulfills the microbiological requirements as space food.  相似文献

A series of novel 1,3,4-oxadiazole derivatives (OSD, OCOD, ONOD, OPD, COD, PMOD, and PCOD) were synthesized and characterized. Their structures were confirmed on the basis of IR, NMR and mass spectroscopy and molecular weights were found in the range 300–325 g/mol. Cancerous cell lines (MCF-7, HepG2) and non-cancerous cell lines (Chang liver cells) were treated with these compounds for 48 h, which caused dose dependent decrease in the cell viability. From the seven derivatives, OSD was found to be most potent with IC50 value close to 50 μM on all tested cell lines. Hence, this compound was selected for mechanistic study on HepG2 cell lines. Fluorescent cell staining and DNA fragmentation study of 50 μM OSD on HepG2 cells, showed events marked by apoptosis such as nuclear fragmentation, cytoplasm shrinkage and DNA damage. Further, the cells with same treatment were quantified for apoptosis using annexin V-PI flow cytometric technique. The percentage of apoptotic cells was significantly higher (p < 0.05) after OSD treatment compared to control cells. OSD induced a significant increase (p < 0.05) in the expression of the tumor suppressor p53 in HepG2 cells. The constitutive expression of anti-apoptotic protein Bcl-2 significantly decreased (p < 0.05) after treatment, while the expression of proapoptotic protein Bax significantly increased (p < 0.05). The change in Bax to Bcl-2 ratio suggested involvement of Bcl-2 family in induction of apoptosis. Furthermore, the levels of caspase-9 and caspase-3 were significantly (p < 0.05) up regulated in HepG2 cells after OSD treatment. The data suggest that 1,3,4-oxadiazole derivatives induce apoptosis mediated by intrinsic pathway of apoptosis. The findings strengthen the potential of the 1,3,4-oxadiazole scaffold OSD, as an agent with chemotherapeutic and cytostatic activity in human hepatocellular carcinoma in vitro.  相似文献

A new simple isocratic chiral liquid chromatographic method was developed for the enantiomeric purity of Ramelteon[(S)-N-[2-(1,6,7,8-tetrahydro-2H-indeno[5,4-b]furan-8-yl) ethyl]-propionamide], a melatonin agonist in bulk drugs. The chromatographic separation was achieved on Chiralpak AD-H, 250 mm × 4.6 mm, 5 μm column using a mobile phase system consisting of n-hexane, ethanol and methanesulfonic acid in the ratio of 900:100:0.1 (v/v/v). The mobile phase was pumped on the column at the flow rate of 1 mL min?1. Addition of methane sulfonic acid in the mobile phase enhanced chromatographic efficiency and resolution between the enantiomers. The resolution between the enantiomers was found to be more than four. The developed method was subsequently validated and proved to be accurate and precise. The experimentally established limit of detection and quantification of (R)-enantiomer were found to be 25.5 and 77.2 ng ml?1, respectively, for 20 μl injection volumes. The percentage recovery of (R)-enantiomer was ranged from 98.5 to 101.9 in bulk drug samples of Ramelteon. The stability of Ramelteon sample in analytical solution was checked for about 48 h at room temperature and was found to be stable for about 48 h. The proposed method was found to be suitable and accurate for the quantitative determination of (R)-enantiomer in drug substance.  相似文献

The study is intended to monitor the comparative degradation rates of acetaminophen in binary and ternary combinations by UV–vis spectroscopy. The drugs were exposed to UV-rays in blister packing. The exposition time was 24, 48 and 72 h for both shorter and longer wavelengths. The problem of overlapping UV bands of aspirin and caffeine with acetaminophen was solved by extracting them in diethylether, therefore, we developed a straightforward, rapid and accurate assay method for measuring acetaminophen concentration in binary and ternary mixtures and to monitor its degradation.  相似文献

The nature of the ultrafiltration resistance of Escherichia coli broth harvested at different culture times was investigated. E. coli broth was harvested at the point of glucose depletion (0–h aged broth) and at 12 h after that (12 h aged broth). The filtration resistance of whole broth nearly doubled during the 12 h aging. In the case of the 0 h aged broth, the dominant resistance was from the deposited cells. However, for the 12 h aged broth, an interaction between cells and soluble broth components determined the filtration resistance. If either were missing, the resistance increase was markedly less. The cause of the interaction appeared to be a change in the cell (surface) during the aging, leading to binding with some broth component(s) that increased the resistance. The increased resistance occurred both when broth components were present during cake formation and when exposure came after cake deposition from washed cells. The interacting soluble component permeated a 0.16 μm membrane and was retained by a 300 kDa MWCO membrane. Since DNA would show such permeation and DNA added to aged, washed cells increased resistance in a similar fashion as occurred with aged broth, it is likely that DNA released from cells contributes to the interaction with aged cells.  相似文献

Vapour pressures of water over saturated solutions of magnesium, calcium, nickel and zinc acetates were determined as a function of temperature. The vapour pressures served to evaluate the water activities, osmotic coefficients and molar enthalpies of vaporization. Apparent molar volumes Vϕ were determined for aqueous adonitol, dulcitol, glycerol, meso-erythritol, myo-inositol, d-sorbitol, and xylitol at temperatures from (278.15 to 368.15) K and at the pressure 0.35 MPa, and apparent molar heat capacities Cp,ϕ of the same solutions were determined at temperatures from (278.15 to 363.15) K at the same pressure. Molalities m/(mol · kg−1) of the solutions were in the range (0.02  m  3.2) for adonitol, (0.02  m  0.15) for dulcitol, (0.02  m  5.0) for glycerol, (0.02  m  3.0) for meso-erythritol, (0.02  m  0.5) for myo-inositol, (0.02  m  2.0) for d-sorbitol, and (0.02  m  2.7) for xylitol. A vibrating tube densimeter was used to obtain solution densities and a fixed-cell temperature scanning calorimeter was used to obtain heat capacities. Values of Vϕ and Cp,ϕ for these sugar alcohols are discussed relative to one another and compared to values from the literature, where available.  相似文献

A perovskite-type oxide of Ba0.5Sr0.5Co0.8Fe0.2O3−δ (BSCFO) with mixed electronic and oxygen ionic conductivity at high temperatures was used as an oxygen-permeable membrane. A tubular membrane of BSCFO made by extrusion method has been used in the membrane reactor to exclusively transport oxygen for the partial oxidation of ethane (POE) to syngas with catalyst of LiLaNiO/γ-Al2O3 at temperatures of 800–900 °C. After only 30 min POE reaction in the membrane reactor, the oxygen permeation flux reached at 8.2 ml cm−2 min−1. After that, the oxygen permeation flux increased slowly and it took 12 h to reach at 11.0 ml cm−2 min−1. SEM and EDS analysis showed that Sr and Ba segregations occurred on the used membrane surface exposed to air while Co slightly enriched on the membrane surface exposed to ethane. The oxygen permeation flux increased with increasing of concentration of C2H6, which was attributed to increasing of the driving force resulting from the more reducing conditions produced with an increase of concentration of C2H6 in the feed gas. The tubular membrane reactor was successfully operated for POE reaction at 875 °C for more than 100 h without failure, with ethane conversion of ∼100%, CO selectivity of >91% and oxygen permeation fluxes of 10–11 ml cm−2 min−1.  相似文献

The synthesis, spectral characterization, crystal structure and antimicrobial activity of the novel synthetic molecule 7a-Aza-B-homostigmast-5-eno [7a,7-d] tetrazole, C29H48N4 has been reported. The structure has also been determined by X-ray diffraction technique using direct method and was refined on F2 by the full-matrix least-squares. Crystals are orthorhombic and their space group is P212121, with a = 7.230(3), b = 31.451(13), c = 11.974(5) (Å), α = β = γ = 90°. It can be conveniently obtained by the reaction of 7-Oxostigmast-5-ene with hydrazoic acid. The molecule has also been screened for its possible in vitro antimicrobial activity against Staphylococcus aureus, Streptococcus mutans, Streptococcus pyogenes, Staphylococcus epidermidis, Bacillus cereus, Corynebacterium xerosis, Escherichia coli, Klebsiella pneumoniae, Proteus vulgaris and Pseudomonas aeruginosa (MTCC 424). Minimum inhibitory concentration (MIC) of the synthesized compound has also been evaluated. The highest activity is observed against C. xerosi and P. vulgaris. Moreover, the compound has also been screened for its in vitro cytotoxicity against human colon carcinoma cell line, HCT116 and human liver hepatocellular carcinoma cell line, HepG2, using doxorubicin as standard. On the basis of its IC50 values, 7a-Aza-B-homostigmast-5-eno [7a,7-d] tetrazole was found to inhibit the cancer cells effectively.  相似文献

The solubility measurements of sodium dicarboxylate salts; sodium oxalate, malonate, succinate, glutarate, and adipate in water at temperatures from (278.15 to 358.15 K) were determined. The molar enthalpies of solution at T = 298.15 K were derived: ΔsolHm (m = 2.11 mol · kg?1) = 13.86 kJ · mol?1 for sodium oxalate; ΔsolHm (m = 3.99 mol · kg?1) = 14.83 kJ · mol?1 for sodium malonate; ΔsolHm (m = 2.45 mol · kg?1) = 14.83 kJ · mol?1 for sodium succinate; ΔsolHm (m = 4.53 mol · kg?1) = 16.55 kJ · mol?1 for sodium glutarate, and ΔsolHm (m = 3.52 mol · kg?1) = 15.70 kJ · mol?1 for sodium adipate. The solubility value exhibits a prominent odd–even effect with respect to terms with odd number of sodium dicarboxylate carbon numbers showing much higher solubility. This odd–even effect may have implications for the relative abundance of these compounds in industrial applications and also in the atmospheric aerosols.  相似文献

Systems based on naphthalene and alkali metals (Li, Na, K) in THF are able to induce the alkylation of naphthalene with ethene at room temperature and atmospheric pressure. The highest activity in this reaction is exhibited by the naphthalene–potassium system which converts naphthalene into 1-ethylnaphthalene (1) and small amounts of two isomeric dihydro derivatives of 1 in a yield of 85% (24 h, K:C10H8 = 2:1). The same alkylation products are formed when metallic sodium is used instead of potassium. The interaction of ethene with the naphthalene–lithium system (24 h, Li:C10H8 = 2:1) affords 1 together with 1-n-butylnaphthalene (4), 1-n-hexylnaphthalene (5), 1-n-oktylnaphthalene (6) and dihydro derivatives of 5 and 6 in a total yield of 60%. Alkylation of toluene with ethene in the naphthalene–alkali metal systems leads to the formation of higher monoalkylbenzenes. The greatest toluene conversion (48%, 24 h) is observed on using the lithium-containing system (Li:C10H8 = 2:1), in the presence of which a mixture of n-propylbenzene (11), n-pentylbenzene (12), 3-phenylpentane (13) and 3-phenylheptane (14) is produced from ethene and toluene. On the replacement of lithium by sodium or potassium, only 11 and 13 are obtained. A treatment of biphenyl, phenanthrene, trans-stilbene, pyrene and anthracene with alkali metals in THF also gives systems capable of catalyzing the alkylation of toluene with ethene at 22 °C. Of particularly active is the stilbene–lithium system (Li:stilbene = 3:1) which converts toluene into a mixture of 1114, n-heptylbenzene and 5-phenylnonane in a yield of 58%. In all cases, the rate of the alkylation considerably increases in the presence of the solid phase of alkali metal. The mechanism of the reactions found is discussed.  相似文献
