A simple and efficient one-pot method for the synthesis of 4-anisyl-2-phenyl-1,2,3,4-tetrahydroquinoline derivatives using a three-component imino Diels-Alder cycloaddition (Povarov reaction) between anilines, benzaldehyde, and trans-anethole in the presence of acidic catalysts is shown. New substituted tetrahydroquinolines are reported and their direct preparation from the anise essential oil is described. Also, a simple procedure of the same tetrahydroquinolines from the anise seeds under supercritical fluid (CO2) conditions has been reported.  相似文献

Herein, we report a copper salt‐controlled divergent reactivity toward α‐diazocarbonyl compounds. By a simple change of the copper counteranion under identical reaction conditions, the reported method allowed an easy access to either (Z)‐α‐fluorovinylphosphonate or alkyl‐SCF2PO(OEt)2 derivatives in good yields. Mechanistic studies were performed and suggested two different pathways to explain the formation of these products.  相似文献

A titanocene‐catalyzed regiodivergent radical arylation is described that allows access to either enantiomerically pure tetrahydroquinolines or indolines from a common starting material. The regioselectivity of epoxide opening that results in the high selectivity of heterocycle formation is controlled by two factors, the absolute configuration of the enantiopure ligands of the (C5H4R)2TiX2 catalyst and the inorganic ligand X (X=Cl, OTs). The overall reaction is atom‐economical and constitutes a radical Friedel–Crafts alkylation.  相似文献

A novel series of tetrahydroquinolines containing acetohydrazide, oxopyrazole, oxothioxodihydropyrazole, and thioxotriazole have been synthesized. Antileishmanial, antitumor, and cytotoxicity activities of synthesized compounds were evaluated in vitro. Antileishmanial activity of the most synthesized compounds showed tremendous activity towards Leishmania major. Most of the test compounds exhibited significant level of tumor inhibition. The tetrahydropyrano[2,3‐b]quinolin‐2‐one 6 and 4‐oxo‐4H‐pyrazol‐3‐yloxytetrahydroquinoline‐3‐carbonitrile derivatives 18 showed 100% tumor inhibition comparable with standard drug vincristine (100% tumor inhibition). Tetrahydroquinolines under investigation showed cytotoxicity with LD50 values in the range 0.56–3.01 μg/mL compared with standard drug MS‐222 with LD50 value of 4.30 μg/mL. The presence of a pyrazole ring markedly improved the activity profiles of tetrahydroquinoline. All newly synthesized compounds were characterized by IR, 1H NMR, and MS.  相似文献

The discovery of sustainable and scalable synthetic protocols leading to gold–aryl compounds bearing N-heterocyclic carbene (NHC) ligands sparked an investigation of their reactivity and potential utility as organometallic synthons. The use of a mild base and green solvents provide access to these compounds, starting from widely available boronic acids and various [Au(NHC)Cl] complexes, with reactions taking place under air, at room temperature and leading to high yields with unprecedented ease. One compound, (N,N′-bis[2,6-(di-isopropyl)phenyl]imidazol-2-ylidene)(4-methoxyphenyl)gold, ([Au(IPr)(4-MeOC6H4)]), was synthesized on a multigram scale and used to gauge the reactivity of this class of compounds towards C−H/N−H bonds and with various acids, revealing simple pathways to gold–based species that possess attractive properties as materials, reagents and/or catalysts.  相似文献

A simple synthesis of medicinally important cis‐2‐methyl‐4‐azapan‐2‐one‐1,2,3,4‐tetrahydroquinolines/cis‐9‐(2‐methyl‐1,2,3,4‐tetrahydroquinolin‐4‐yl)‐9H‐carbazole was reported. Multicomponent one pot synthesis with anilines and N‐vinylcaprolactam/N‐vinyl carbazole via imino Diels‐Alder reaction by using antimony trichloride as catalyst and acetonitrile as solvent was employed. NMR technique (2D) was used to study the regio‐ and stereo‐chemistry of newly synthesized compounds. The cis diastereo‐selectivity of the products was predicted by COSY and NOESY studies.  相似文献

We report a gold-catalyzed intramolecular hydroarylation of unactivated olefins using a combination of AuCl3/AgOTf as the catalytic system affording dihydrobenzopyrans, tetralins and tetrahydroquinolines in good yields. For our preliminary mechanistic studies, we have investigated the kinetic isotope effects with deuterated arene compounds and found that this catalytic hydroarylation is consistent with an electrophilic aromatic substitution process.  相似文献

The interactions between 4‐R‐substituted 2‐furyl‐1,2,3,4‐tetrahydroquinolines (synthesized by the Povarov reaction) and a number of alkenes have been investigated. Maleic, dibromomaleic, dichloromaleic, and citraconic anhydrides, as well as acryloyl, methacryloyl, crotonyl, and cynnamoyl chlorides were used as alkene components. It was shown that the initial N‐acylation of the tetrahydroquinolines was followed by a spontaneous [4+2]‐cycloaddition of an N‐acryloyl substituent to the furan ring. It was established that the intramolecular Diels–Alder reaction of furans is reversible, occurs stereoselectively as exo‐addition, and led to target epoxyisoindolo[2,1‐a]tetrahydroquinolines with moderate yields. Oxidation and aromatization of the synthesized products were carried out.  相似文献

The behavior of the even-electron mass 150 ions, (presumably nitrobenzoyl cations) generated by simple α-cleavage of m- and p-nitrobenzoic acid derivatives, undergo two competing secondary fragmentations to yield odd-electron ions by the expulsion of NO and NO2. Expulsion of NO from the mass 150 ions from p-nitro compounds is far greater than expulsion of NO from those generated from m-nitro compounds. In addition, the behavior of the even-electron mass 136 ions, generated from m- and p-nitrobenzyl compounds, was compared to that of the mass 150 ions and they were found to decompose in a similar fashion. From our results we conclude that the decomposing mass 136 ions produced from either the m- or p-nitrobenzyl compounds, or both, do not have the nitropylium structure; our data are more consistent with the decomposing ions having nitrobenzyl structures.  相似文献

Abstract

A simple, efficient, and metal-free methodology for the preparation of 2-mercaptobenzothiazole and derivatives in excellent yields via microwave-assisted pathway is reported. Our condition provides a convenient protocol for the synthesis of a diverse collection of 2-mercaptobenzothiazoles and 6-(4-substituted-1H-1,2,3-triazol-1-yl)-2-mercaptobenzothiazoles with a very simple purification process. This report provides an alternative protocol for fast access to the wide range of compounds for sequence synthesis and biological studies.  相似文献

The identification of new photocatalytic pathways expands our knowledge of chemical reactivity and enables new environmentally friendly synthetic applications. However, the development of miniaturized screening procedures/platforms to expedite the discovery of photochemical reactions remains challenging. Herein, we describe a picomole‐scale, real‐time photoreaction screening platform in which a handheld laser source is coupled with nano‐electrospray ionization mass spectrometry. By using this method, we discovered an accelerated dehydrogenation pathway for the conversion of tetrahydroquinolines into the corresponding quinolines. This transformation is readily promoted by an off‐the‐shelf [Ru(bpy)3]Cl2?6 H2O complex in air at ambient temperature in direct sunlight, or with the aid of an energy‐saving lamp. Moreover, radical cations and trans‐dihydride intermediates captured by the screening platform provided direct evidence for the mechanism of the photoredox reaction.  相似文献

A novel and practical strategy for the construction of imidazo[1,2‐a]pyridin‐2‐amine frameworks has been developed. The present sequential approach involves addition of arylamines to nitriles and I2/KI‐mediated oxidative C?N bond formation without purification of the intermediate amidines. This operationally simple synthetic process provides a facile access to a variety of new 2‐amino substituted imidazo[1,2‐a]pyridines and related heterocyclic compounds in an efficient and scalable fashion.  相似文献

In this work, seven new norcantharimide derivatives were synthesized by an acidolysis method. The compounds were prepared by acidolyzing trans-1,4-diacetate and trans-1,2-chloroacetate structures, which were obtained by stereospecific cleavage of the internal etheric bond of the tricyclic imides. The HCl(gas) was produced from the reaction of H2SO4 with NaCl. The resulting gas was bubbled into the reaction mixture. Trans-1,4-diacetate and trans-1,2-chloroacetate were thus acidolyzed, and the corresponding diol and halohydrin products were obtained respectively in moderate overall yields from low-cost starting materials, using simple and easily scalable chemistry. The products were characterized by means of spectroscopic techniques. The synthesized compounds have high potential as anticancer agents and can be valuable for studies in this area.  相似文献

Photocyclization, irrespective of whether multiple steps (e.g., Norrish–Yang cyclization) or a single concerted step (e.g., 6π photocyclization) are involved, is an intramolecular photochemical process resulting in the formation of one new single bond to afford a ring system. In particular, visible-light-induced photocyclization offers a green and sustainable route to organic cyclic compounds that are difficult to access by thermal reactions. Herein, we describe the ambient light-induced intramolecular photocyclization of a series of donor/acceptor chromophores 1 d – 3 d containing two types of photoresponsive motifs, namely an electron-deficient BF2-chelated ketone fused with an electron-rich thiophene, and probe the solution-phase and solid-state photochromic performance of these compounds. The results reveal that simple variation of R substituents on the diaryl moiety allows one to control the intramolecular photocyclization mechanism with high photochemical selectivity, e.g., under ambient light, methyl-substituted 1 d and 2 d undergo reversible 6π photocyclization, whereas ethyl-substituted 3 d exclusively undergoes irreversible Norrish–Yang photocyclization. Single-crystal X-ray analysis of Norrish–Yang cyclization products reveals the formation of four pairs of conformational enantiomers differing in the dihedral angle between benzothiophene and the BF2 core, namely (±) N -3 d @68°, (±) N -3 d @-77°, (±) N -3 d @-78°, and (±) N -3 d @-102°. The UV/Vis absorption spectra of 1 d – 3 d cover a broad visible-light region (380–572 nm), while DFT and TD-DFT calculations reveal that absorption in this region is dominated by the charge-transfer (CT) transition from the thiophene-centered HOMO to the LUMO of the electron-deficient π-conjugated BF2-chelated unit and the n→π* and π→π* transitions within the latter unit. The spatial separation of the HOMO and LUMO of these dyes promotes triplet-state generation and self-photosensitizes intramolecular photocyclization in the visible-light region. Three-dimensional time-resolved and steady-state emission spectra of 3 d show that the Norrish–Yang photocyclization takes place within milliseconds with excellent conversion efficiency (96 %).  相似文献

The tetraphosphine all‐cis‐1,2,3,4‐tetrakis(diphenylphosphinomethyl)cyclopentane (Tedicyp) in combination with [Pd(C3H5)Cl]2 affords an efficient catalyst of the coupling of 2‐chloroacrylonitrile with arylboronic acids. In the presence of 1% catalyst, the 2‐arylacrylonitrile derivatives were obtained in medium to good yields. A variety of substituents such as alkyl, methoxy, fluoro, trifluoromethyl, formyl, or nitro on the arylboronic acid are tolerated. The cross‐coupling reactions of methyl 2‐chloroacrylate with arylboronic acids give simple access to 2‐phenylacrylate derivatives, which are useful precursors for the synthesis of biologically active compounds such as ibuprofen, ketoprofen, and naproxen.  相似文献

Six isomeric C20 alkylthiophenes were synthesized in order to prove the occurrence of such compounds in various deep sea sediments. As expected from the analogy with the corresponding saturated hydrocarbons, 2- and 3-heptadecylthiophene have considerably longer relative retention times than the isomeric isoprenoid compounds, i.e. 2- and 3-(4,8,12-trimethyltridecyl)thiophene and 3- and 4-methyl-2-(3,7,11-trimethyldodecyl)thiophene. Covats retention indices for all compounds were determined by isothermal analysis at 190°C on two different silicone-coated capillary columns. The mass spectra of the monoalkylated thiophenes show a base peak at m/z 98 corresponding to a Mcafferty rearrangement product in the case of the 3-substituted isomers, whereas a simple benzylic cleavage dominates in the mass spectra of 2-alkylthiophenes. 4-Methyl-2-(3,7,11-trimethylodecy)thiophene is an exception from this simple rule because also shows the McLafferty rearrangement as dominant fragmentation. Two of the isoprenoid thiophenes are prominent compounds in various deep sea sediments.  相似文献

In this work, we present a theoretical study (based on DFT‐calculations) of the electronic properties of compounds crystallising in a NH4Pb2Br5 type structure in a wide pressure range. The main focus of this study is to elucidate the nature of bonding of the ns2‐cations at ambient and elevated pressure. For a better understanding of the structure and bonding, the DOS of these compounds are evaluated and discussed on the basis of a simple model assuming mainly ionic interactions. The calculations are complemented by an orbital analysis using the crystal orbital Hamilton population (COHP) and an analysis of the electronic density topology with the electron localisation function (ELF). Structural and theoretical investigations give results that are in excellent agreement: The DFT‐calculations confirm the existence of bonding interactions between the ns2‐cations at elevated pressure. Our study indicates that the “character” of the additional electron pair changes with increasing pressure from nonbonding to bonding in agreement with a simple model system of two interacting ns2‐cations.  相似文献

The reaction of 2-amino-2-oxazolines with ethoxycarbonyl isocyanate was investigated in order to access to fused 1,3,5-triazine-2,4-diones with a potential 5-HT2 antagonist activity. The reaction leads to 2,3,6,7-tetrahydro-4H-oxazolo[3,2-a]-1,3,5-triazine-2,4-diones and to 1-carbethoxy-3-(2-iminooxazolidine)ureas. During the carbamoylation the regioselectivity seems to be related to the strong nucleophilic character of the endo nitrogen atom of 2-amino-2-oxazolines. The structures of two compounds were studied by X-ray crystallography. N-Substituted compounds have been prepared by alkylation of the 2,3,6,7-tetrahydro-7-phenoxymethyl-4H-oxazolo[3,2-a]-1,3,5-triazine-2,4-dione.  相似文献

α-Trifluoromethylated ketones have attracted significant attention as valuable building blocks in organic synthesis. Such compounds are generally accessed through trifluoromethylation of ketones. Here we report an alternative disconnection approach for the construction of α-CF3 carbonyl compounds by using aroyl fluorides as bifunctional reagents for fluoroaroylation of gem-difluoroalkenes through cooperative photoredox and N-heterocyclic carbene (NHC) catalysis. This strategy bypasses the use of expensive or sensitive trifluoromethylation reagents and/or the requirement for ketone pre-functionalization, thus enabling an efficient and general synthetic method to access α-CF3-substituted ketones. A wide variety of gem-difluoroalkenes and aroyl fluorides bearing a diverse set of functional groups are eligible substrates. Notably, the developed methodology also provides rapid access to mono- or difluoroalkyl ketones. Mechanistic studies reveal that merging photoredox catalysis with NHC catalysis is essential for the reaction.  相似文献
