A new fluorogenic method for the selective and sensitive determination of chromium(VI) in acidic water using rhodamine B hydrazide was developed. This method was based on the oxidation of non-fluorescent rhodamine B hydrazide by potassium dichromate in acidic aqueous conditions to give rhodamine B, which was highly fluorescent, as a product. With the optimum condition described, the fluorescence enhancement at 585 nm was linearly related to the concentration of chromium(VI) in the range of 5.0 × 10−8 to 2.0 × 10−6 mol L−1 (2.60-104 ng mL−1) with a correlation coefficient of R2 = 0.9993 (n = 18) and a detection limit of 5.5 × 10−9 mol L−1 (0.29 ng mL−1). The R.S.D. was 2.2% (n = 5). The proposed method was also applied to the determination of chromium(VI) in drinking water, river water and synthetic samples.  相似文献

A new functional monomer N-(o-carboxyphenyl)maleamic acid (CPMA) was synthesized and chosen for the preparation of surface-grafted ion-imprinted polymers (IIPs) specific for thorium(IV). Polymerizable double bond was introduced to silica gel surface by amidation reaction between -NH2 and maleic anhydride. In the ion-imprinting process, thorium(IV) was complexed with the carboxyl groups, then was imprinted in the polymers grafted to the silica gel surface. The imprinted Th(IV) was removed with 3 mol L−1 HCl. The obtained imprinted particles exhibited excellent selectivity and rapid kinetics process for Th(IV). The relatively selective factor (αr) values of Th(IV)/La(III), Th(IV)/Ce(III), Th(IV)/Nd(III), Th(IV)/U(VI), and Th(IV)/Zr(IV) were 85.7, 88.9, 26.6, 64.4, and 433.8, respectively, which were greater than 1. The precision (R.S.D.), the detection limit (3σ), and the quantification limit (10σ) of the method were 1.9%, 0.51 ng mL−1 and 1.19 ng mL−1, respectively. The prepared IIPs as solid-phase extractants were successfully applied for the preconcentration of trace thorium in natural and certified samples prior to its determination by inductively coupled plasma atomic emission spectrometry (ICP-AES) with satisfactory results.  相似文献

A new method was developed for determination of micro amounts of nucleic acids based on near-infrared (near-IR) fluorescence recovery, employing a two-reagent system which is composed of an anionic tetracarboxy aluminum phthalocyanine (AlC4Pc) and a cationic tetra-N-hexadecylpyridiniumyl porphyrin (TC16PyP). The fluorescence of the AlC4Pc, with the maximum emission wavelength at 701 nm, could be quenched by TC16PyP at its proper concentration, but recovered by adding nucleic acids. Under optimal conditions, the recovered fluorescence is proportional to the concentration of nucleic acids. The calibration graphs are linear over the range of 1-200 ng mL−1 for fish sperm DNA (FS DNA) and 2-400 ng mL−1 for calf thymus DNA (CT DNA). The corresponding detection limits are 0.59 ng mL−1 for FS DNA and 0.82 ng mL−1 for CT DNA, respectively. Four synthetic and three real nucleic acid samples were determined with satisfactory results.  相似文献

In pH 4.2-4.8 HAc-NaAc buffer solution, folic acid (FA) could react with uranium (VI) to form a 2:1 anionic chelate which further reacted with some basic triphenylmethane dyes (BTPMD) such as Ethyl Violet (EV), Methyl Violet (MV) and Crystal Violet (CV) to form 1:2 ion-association complexes. As a result, not only the absorption spectra were changed, but also the intensities of resonance Rayleigh scattering (RRS) were enhanced greatly and the new RRS spectra were observed. The maximum RRS wavelengths were located at 328 nm for EV system, 325 nm for MV system and 328 nm for CV system. The fading degree (ΔA) and RRS intensities (ΔI) of three systems were different. Under given conditions, the ΔA and ΔI were all directly proportional to the concentration of FA. The linear ranges and the detection limits of RRS methods were 0.0039-5.0 μg mL−1 and 1.2 ng mL−1 for EV system, 0.0073-4.0 μg mL−1 and 2.2 ng mL−1 for MV system, 0.014-3.5 μg mL−1 and 4.7 ng mL−1 for CV system. The RRS methods exhibited higher sensitivity, so they are more suitable for the determination of trace FA. The optimum conditions, the influencing factors and the effects of coexisting substances on the reaction were investigated. The method can be applied to the determination of FA in serum and urine samples with satisfactory results. The structure of the ternary ion-association complex and the reaction mechanism were discussed in this work.  相似文献

A novel class of redox-active molecular tags, poly(o-phenylenediamine)-carried nanogold particles (GPPDs), was first synthesized and functionalized with horseradish peroxidase-anti-prolactin conjugates (HRP-anti-PRL). Thereafter, a specific sandwich-type electrochemical immunoassay was designed for determination of prolactin (PRL) by using GPPD-labeled HRP-anti-PRL conjugates as molecular tags on anti-PRL antibody-modified glassy carbon electrode. Compared with pure gold nanoparticles and poly(o-phenylenediamine) microspheres, the as-prepared GPPDs increased the surface coverage of the nanostructures, and enhanced the immobilization amount of biomolecules. Several labeling protocols compromising GPPD-labeled HRP-anti-PRL, nanogold particles-labeled HRP-anti-PRL and poly(o-phenylenediamine) microspheres-labeled HRP-anti-PRL, were investigated for detection of PRL, and improved analytical features were obtained with the GPPD-based strategy. With the GPPD labeling method, dependence of the electrochemical signals on the incubation time and pH of the assay solution were also studied. The strong attachment of HRP-anti-PRL to the GPPDs resulted in a good repeatability and intermediate reproducibility down to 9.8%. The dynamic concentration range spanned from 0.5 to 180 ng mL−1 PRL with a detection limit of 0.1 ng mL−1 at the 3Sblank level. No significant differences at the 95% confidence level were encountered in the analysis of 10 spiked blank cattle serum samples between the developed immunoassay and enzyme-linked immunosorbent assay method for determination of PRL.  相似文献

No previous publications about percutaneous absorption of polyethylene glycol 25 p-aminobenzoic acid (PEG-25 PABA) have been found in the literature and the expected levels to be found in human urine after sunscreens use are unknown. The method proposed here is suitable to determine PEG-25 PABA in the urine of sunscreens users in order to carry out studies on body accumulation/excretion. It is based on solid-phase extraction (SPE) with size-exclusion liquid chromatography determination. Solid-phase extraction allows the analyte to be retained and subsequently eluted for a clean-up, using a silica-based cartridge. The size-exclusion liquid chromatography of the eluted allows the rest of matrix interferences to be avoided. Fluorescence intensity was measured at λem = 350 nm (λexc = 300 nm). The sensitivity of the proposed method is in the order of 450 ± 5 mL ng−1 and the detection limit (3 Sy/x/b) in the measured solutions is in the order of 13 ng mL−1, that is 2.6 ng mL−1 in urine samples. The method enables PEG-25 PABA to be determined in both, spiked and unspiked human urine samples. Results obtained for spiked human urine samples (11-100 ng mL−1) demonstrated the accuracy of the method. The mean relative standard deviation of the results was in the order of 3-10%. Three volunteers applied a sunscreen lotion containing a 8% PEG-25 PABA sunscreen cream and their urinary excretion was controlled from the moment of application until the excreted amounts were no longer detectable.  相似文献

The supramolecular interaction of cucurbit(n = 7)uril (Q[7]) with berberine chloride (BER) has been studied in aqueous solution at pH 2.0 and room temperature by spectro-fluorimetry. The association constant of the complex was 2.07 × 106 L mol−1 calculated by using a nonlinear least squares method. 1H NMR spectra confirmed that a 1:1 stable complex is formed between Q[7] and BER. This work proposes a possible interaction mode, in which the guest BER is incorporated inside the hydrophobic cavity of the host Q[7] via the isoquinoline ring part of the guest molecule. Based on a significant enhancement of the fluorescence intensity of this supramolecular complex, a spectrofluorimetric method with high sensitivity and selectivity has been developed for the determination of BER in aqueous solution in the presence of Q[7]. The linear range of the method was from 7.43 to 11.2 × 103 ng mL−1with the detection limit 4.2 ng mL−1. There was no interference from the compounds normally used in tablets, serum or urine constituents. The proposed method was applied to the determination of BER in tablets, serum and urine samples with satisfactory results and good consistency with those obtained by the pharmacopoeia method. This shows that it has promising potential for therapeutic drug monitoring and pharmokinetics and for clinical application.  相似文献

Orthogonal array design (OAD) was applied for the first time to optimize cloud point extraction (CPE) conditions for Se(IV) determination by electrothermal atomic absorption spectrometry (ETAAS) in environmental and biological samples. Selenium was reacted with o-phenylenediamine to form piazselenol in an acidic medium (pH 2). Using Triton X-114, as surfactant, piazselenol was quantitatively extracted into small volume (about 30 μL) of the surfactant-rich phase after centrifugation. Five relevant factors, i.e. surfactant concentration, pH, ionic strength, equilibrium time and temperature were selected and the effects of each factor were studied at four levels on the extraction efficiency of Se(IV) and optimized. The statistical analysis revealed that the most important factors contributing to the extraction efficiency are ionic strength, pH and percent of surfactant. Based on the results obtained from the analysis of variance (ANOVA), the optimum conditions for extraction were established as: pH 6; vial temperature = 50 °C; extraction time = 7 min and 0.3% (w/v) of Triton X-114. The method was permitted to obtain a detection limit of 0.09 ng mL−1 and two linear calibration ranges from 0.6 to 1.0 and 1.0 to 80.0 ng mL−1 Se. The precision (%RSD) of the extraction and determination for the six replicates of Se at 20 ng mL−1 was better than 3.6% and the enrichment factor of 63.5 was achieved. The studied analyte was successfully extracted and determined with high efficiency using cloud point extraction method in water and biological matrices.  相似文献

Polyclonal antibody (PAb) with broad-specificity for O,O-diethyl organophosphorus pesticides (OPs) against a generic hapten, 4-(diethoxyphosphorothioyloxy)benzoic acid, was produced. The obtained PAb showed high sensitivity to seven commonly used O,O-diethyl OPs in a competitive indirect enzyme-linked immunosorbent assay (ciELISA) using a heterologous coating antigen, 4-(3-(diethoxyphosphorothioyloxy)phenylamino)-4-oxobutanoic acid. The 50% inhibition value (IC50) was 348 ng mL−1 for parathion, 13 ng mL−1 for coumaphos, 22 ng mL−1 for quinalphos, 35 ng mL−1 for triazophos, 751 ng mL−1 for phorate, 850 ng mL−1 for dichlofenthion, and 1301 ng mL−1 for phoxim. The limit of detection (LOD) met the ideal detection criteria of all the seven OP residues. A quantitative structure-activity relationship (QSAR) model was constructed to study the mechanism of antibody recognition using multiple linear regression analysis. The results indicated that the frontier-orbital energies (energy of the highest occupied molecular orbital, EHOMO, and energy of the lowest unoccupied molecular orbital, ELUMO) and hydrophobicity (log of the octanol/water partition coefficient, log P) were mainly responsible for the antibody recognition. The linear equation was log(IC50) = −63.274EHOMO + 15.985ELUMO + 0.556 log P − 25.015, with a determination coefficient (r2) of 0.908.  相似文献

The supramolecular interaction of a homologous series of cucurbit[n]uril (CB[n], n = 5, 6, 7, 8) hosts and coptisine (COP) was studied by spectrofluorimetry. All of the CB[n]s were found to react with COP to form 1:1 host-guest stable complexes and the fluorescence intensity of the complexes was greatly enhanced. The apparent association constants of the complexes were 1.44 × 104, 1.28 × 104, 1.86 × 104 and 1.26 × 104 L mol−1 for CB[5], CB[6], CB[7] and CB[8], respectively. In addition, CB[5] and CB[7] exhibited a higher fluorescence signal than CB[6] and CB[8]. The fluorescence intensity of the complex with CB[7] was enhanced 70-fold compared to that of the studied drug itself. Based on the significant enhancement of fluorescence intensity of supramolecular complex, a simple, rapid, highly sensitive, and selective spectrofluorimetric method was developed for the determination of COP in aqueous solution in the presence of CB[7]. At the optimum reaction conditions, a linear relationship was obtained in the range from 0.05 to 1700 ng mL−1 with a detection limit of 0.012 ng mL−1. The proposed method was successfully applied for the determination of the drug in urine and serum samples.  相似文献

A highly sensitive and relatively interference-free spectrophotometric method for determination of calcium is described. The method is based on the reaction between calcium ions and carboxyazo-p-CH3 in aqueous citrate medium of pH 7, to form a blue complex with maximum absorption at 716 nm. The calibration is linear up to 0.12 μg ml−1 calcium with a repeatability (R.S.D.) of 1.0% at a concentration of 0.04 μg ml−1 (n=5). The molar absorptivity of the complex and Sandell’s sensitivity are 3.5×105 l mol−1 cm−1 and 0.11 ng cm−2, its 10σ limit of quantification and the 3σ limit of detection were found to be 0.3 ng ml−1 and 0.09 ng ml−1 respectively. The influence of reaction variables and the effect of interfering ions are studied; no interference was observed in clinical samples. The proposed method has been applied directly to the determination of calcium in clinical samples without the need for pre-concentration, masking metal ions and digesting samples.  相似文献

A novel method for determination of indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) in an extract from mung bean sprouts using high performance liquid chromatography (HPLC) with chemiluminescence (CL) detection is described. The method is based on the CL reaction of auxin (indole-3-acetic acid and indole-3-butyric acid) with acidic potassium permanganate (KMnO4) and tris(2,2′-bipyridyl)ruthenium(II), which was immobilized on the cationic ion-exchange resin. The chromatographic separation was performed on a Nucleosil RP-C18 column (i.d.: 250 mm × 4.6 mm, particle size: 5 μm, pore size: 100) with an isocratic mobile phase consisting of methanol-water-acetic acid (45:55:1, v/v/v). At a flow rate of 1.0 mL min−1, the total run time was 20 min. Under the optimal conditions, the linear ranges were 5.0 × 10−8 to 5.0 × 10−6 g mL−1 and 5.0 × 10−7 to 1.0 × 10−5 g mL−1 for IAA and IBA, respectively. The detection limits were 2.0 × 10−8 g mL−1 and 2.0 × 10−7 g mL−1 for IAA and IBA, respectively. The relative standard deviation (RSD) of intra-day were 3.1% and 2.3% (n = 11) for 2 × 10−6 g mL−1 IAA and 2 × 10−6 g mL−1 IBA; The relative standard deviations of inter-day precision were 6.9% and 4.9% for 2 × 10−6 g mL−1 IAA and 2 × 10−6 g mL−1 IBA. The proposed method had been successfully applied to the determination of auxin in mung bean sprouts.  相似文献

The chlorogenic acid (CA) in Honeysuckle is determined and identified by nano-liquid chromatography-electrospray ionization mass spectrometry (nano-LC-ESI/MS) after extraction with microwave-assisted extraction (MAE). As a new sample preparation method for Honeysuckle, the MAE procedure is optimized, validated and compared with conventional methods including reflux extraction (RE) and ultrasonic extraction (USE). It is found that MAE gives the best result due to the highest extraction efficiency within shortest extraction time (only 4 min). Here, CA is determined by nano-LC-ESI/MS based on the calibration curve of its authentic standard. The method linearity, detection limit, precision and recovery are studied. The results show that the combined MAE and nano-LC-ESI/MS method has a linearity (R2 0.991, 0.8-20 ng mL−1), a low limit of detection (0.5 ng mL−1), good precision (R.S.D. = 2.54%) and a recovery (84.8%). The experiment has demonstrated that the nano-LC-ESI/MS following MAE is a fast and reliable method for quantitative analysis of CA in Honeysuckle.  相似文献

This study established a flow injection (FI) methodology for the determination of the total phenolic content in plant-derived beverages based on soluble manganese(IV) chemiluminescence (CL) detection. It was found that mixing polyphenols with acidic soluble manganese(IV) in the presence of formaldehyde evoked chemiluminescence. Based on this finding, a new FI-CL method was developed for the estimation of the total content of phenolic compounds (expressed as milligrams of gallic acid equivalent per litre of drink) in a variety of wine, tea and fruit juice samples. The proposed method is sensitive with a detection limit of 0.02 ng mL−1 (gallic acid), offers a wide linear dynamic range (0.5-400 ng mL−1) and high sample throughput (247 samples h−1). The relative standard deviation for 15 measurements was 3.8% for 2 ng mL−1 and 0.45% for 10 ng mL−1 of gallic acid. Analysis of 36 different samples showed that the results obtained by the proposed FI-CL method correlate highly with those obtained by spectrophotometric methods commonly used for the evaluation of the total phenolic/antioxidant level. However, the FI-CL method was found to be far simpler, more rapid and selective, with almost no interference from non-phenolic components of the samples examined.  相似文献

A method for the simultaneous preconcentration and determination of Hg(II) and MeHg(I) at the ng ml−1 level has been developed. This method is based on solid phase extraction using a newly synthesized chelating resin containing nitrogen and sulphur donor sites of the 1,2-bis(o-aminophenylthio)ethane moiety that is very selective for mercury. The characterization of the resin has been carried out by elemental analyses, infrared spectral data, thermogravimetric analysis and metal ion capacities. The resin is highly selective for Hg(II) and MeHg(I) with an exchange capacity of 0.38 and 0.30 mmol g−1, respectively. Various parameters like pH, column flow rate, desorbing agents are optimized. Cold vapour atomic absorption spectrometry (CVAAS) was used to measure the concentration of both species of mercury. The calibration graph was linear upto 10 ng ml−1 with a 3σ detection limit of 0.09 ng ml−1. The recovery of Hg(II) and MeHg(I) was found to be 98.9±2.0 and 98.0±1.1%, respectively. The method has been used for routine determination of trace levels of mercury species in natural waters to comply with more stringent regulations.  相似文献

Spectrophotometric determination of titanium(IV) was accomplished with o-carboxyphenylfluorone (OCPF) in the presence of hexadecyltrimethyl ammonium chloride (HTAC) under strongly acidic media. In the determination of titanium(IV), Beer's law was obeyed in the range of 24-340 ng mL−1 with an effective molar absorption coefficient (at 530 nm) and relative standard deviation of 2.24 × 105 dm3 mol−1 cm−1 and 0.64% (n = 8), respectively. The severe interference of iron ions was easily eliminated by the addition of ethylenediaminetetraacetic acid (EDTA); the effects of other foreign substances were low. Equilibrium and kinetic studies under analytical conditions were investigated to quantitatively evaluate the reaction mechanism. The obtained orange complex is considered to be Ti(OCPF)4. Its stability log Kf and rate constant Kobs are 16.88 and 1.65 × 10−2 s−1, respectively. It is suggested that the color of the complex is related to the species of OCPF in the solution.  相似文献

A cloud point extraction process using mixed micelle of the cationic surfactant CTAB and non-ionic surfactant TritonX-114 to extract uranium(VI) from aqueous solutions was investigated. The method is based on the color reaction of uranium with pyrocatechol violet in the presence of potassium iodide in hexamethylenetetramine buffer media and mixed micelle-mediated extraction of complex. The optimal extraction and reaction conditions (e.g. surfactant concentration, reagent concentration, effect of time) were studied and the analytical characteristics of the method (e.g. limit of detection, linear range, preconcentration, and improvement factors) were obtained. Linearity was obeyed in the range of 0.20-10.00 ng mL−1 of uranium(VI) ion and the detection limit of the method is 0.06 ng mL−1. The interference effect of some anions and cations was also tested. The method was applied to the determination of uranium(VI) in tap water, waste-water and well water samples.  相似文献

A highly selective, and low cost extractive spectrofluorometric method has been developed for determination of trace concentrations of chromium (III & VI) in water samples using the fluorescent reagent tetraphenylphosphonium bromide (TPP+·Br). The method was based upon solvent extraction of the produced ion associate [TPP+·CrO3Cl] of TPP+·Br and halochromate in aqueous HCl and measuring the fluorescence quenching of TPP+·Br in chloroform at λex/em = 242/305 nm. The fluorescence intensity of TPP+Br decreased linearly on increasing the chromium (VI) concentration in the range of 1-114 μg L−1. The limits of detection (LOD) and quantification (LOQ) of chromium (VI) were 0.43 and 1.42 μg L−1, respectively. Chromium (III) species after oxidation to chromium (VI) with H2O2 in alkaline solution were also determined. Chemical speciation of chromium (III & VI) species at trace levels was achieved. The method was applied for analysis of chromium in certified reference material (IAEA Soil-7) and in tap- and wastewater samples and compared successfully (>95%) with the inductively coupled plasma-mass spectrometry (ICP-MS) results.  相似文献

A simple and sensitive method for using electrothermal atomic absorption spectrometry (ET AAS) with Rh as permanent modifier determination of Sb(III) and total Sb after separation and preconcentration by N-benzoyl-N-phenylhydroxylamine (BPHA)-chloroform single drop has been developed. Parameters, such as pyrolysis and atomization temperature, solvent type, pH, BPHA concentration, extraction time, drop size, stirring rate and sample volume were investigated. Under the optimized experimental conditions, the detection limits (3σ) were 8.0 ng L−1 for Sb(III) and 9.2 ng L−1 for total Sb, respectively. The relative standard deviations (R.S.Ds.) were 6.6% for Sb(III) and 7.1% for total Sb (c = 0.2 ng mL−1, n = 7), respectively. The enrichment factor was 96. The developed method has been applied successfully to the determination of Sb(III) and total Sb in natural water samples.  相似文献

A highly sensitive and specific enzyme-linked immunosorbent assay (ELISA) method has been developed for the determination of 20(S)-protopanaxatriol (PPT), one of the major aglycones of dammarane-type ginseng saponins. Polyclonal antibodies raised against ginsenoside F1 (GF1)-bovine serum albumin showed high reactivities to PPT and GF1, whereas they exhibited minor or even no cross-reactivities to other ginsenosides and protopanaxadiol (0.19%). The working range of this method extends from 50 pg ml−1 to 20 ng ml−1 of PPT. The assay reported here has been validated against an HPLC technique using PPT-containing samples and was shown to correlate closely (γ=0.993). This ELISA could be a useful tool for the determination of PPT contained in biological fluids and plant materials.  相似文献
