新型近红外试剂的合成及其现场二聚体与DNA作用的研究   总被引:6,自引:0,他引:6  
合成了一种新型近红外阴离子染料,并对其水溶液及阳离子表面活性剂CTAB存在下的吸收荧光光谱进行了研究。结果表明,低浓度的CTAB与该近红外阴离子染料形成离子缔合物而使阴离子染料的荧光强度降低,当CTAB的浓度进一步加大时,其胶束前预聚集促使该染料形成非荧光二聚体,导致荧光急剧猝灭。  相似文献

应用紫外-可见吸收光谱研究了光敏试剂亚甲蓝在溶液中的聚合状态及其与表面活性剂之间的关系。当添加阴离子表面活性剂的浓度于其临界胶束浓度时,亚甲蓝二聚体的比例将出现一极大值;用环己烷剂萃取后,测得亚甲蓝二聚体的吸收光谱,其最大吸收波长位于604nm。  相似文献

本文通过荧光光谱法、紫外-可见吸收光谱法和透射电镜并结合电导率测定分别研究了水中卵清蛋白与阴离子表面活性剂十二烷基硫酸钠(SDS)和阳离子表面活性剂十二烷基三甲基溴化铵(DTAB)和十六烷基三甲基溴化铵(CTAB)之间的相互作用。研究结果表明卵清蛋白可以增加SDS和CTAB的临界胶束浓度,但对DTAB的临界胶束浓度没有影响。阴离子表面活性剂可以使卵清蛋白构象完全伸展,而阳离子表面活性剂却不具备此种作用。表面活性剂单体与卵清蛋白的相互作用强于表面活性剂胶束与卵清蛋白的相互作用。  相似文献

采用表面张力、电导率、紫外可见吸收光谱、恒波长同步荧光光谱法分析了典型阴离子表面活性剂十二烷基苯磺酸钠在水溶液中的临界胶束浓度(CMC)。 结果表明,同步荧光光谱法测定CMC时的样品用量少,检测灵敏度高,具有较高的准确性;测定的十二烷基苯磺酸钠的第一临界胶束浓度和第二临界胶束浓度分别为1.48和6.90 mmol/L,与传统的表面张力法和电导率法的测定结果一致。 验证了同步荧光光谱法测定表面活性剂CMC的可靠性。  相似文献

对阳离子荧光染料酚藏花红 (PF)在阴离子表面活性剂溶液状态的吸收光谱和荧光光谱进行了研究。结果表明 ,当阴离子表面活性剂十二烷基硫酸钠 (SDS)的浓度在 0 .8× 10 -3 ~ 2 0× 10 -3 mol/L范围内时 ,酚藏花红形成非荧光现场二聚体 ,且在 5 15nm处出现新的特征吸收峰。现场形成的酚藏花红二聚体作为荧光探针的实验表明 ,核酸可使体系的荧光强度有很大的增强 ,且其增强的程度与核酸加入量呈明显的线性关系 ,据此建立了核酸定量分析的新方法。在最优化条件下 ,用于小牛胸腺DNA、鲱鱼精DNA、酵母RNA的定量测定 ,其线性范围分别为 0 0 6~ 18mg/L、0 0 4~ 2 0mg/L和 0 12~ 2 0mg/L ;检测限分别为 2 1、17和36 μg/L。  相似文献

对阳离子染料灿烂甲酚蓝 (BCB)在阴离子表面活性剂存在时的溶液的吸收光谱进行了研究。同时还研究了BCB在阴离子表面活性剂作用下形成的现场二聚体的荧光性能 ,并用其现场二聚体作为探针 ,首次用于蛋白质的测定 ,结果令人满意。用于牛血清白蛋白的测定 ,线性范围为 0~ 7mg/L ,检测限为 3.6 6× 10 -3mg/L。  相似文献

采用芘为荧光探针、二苯甲酮为猝灭剂,以分子荧光探针法首次测定了由刚性基团联接的特殊双亲水基型阴离子表面活性剂——十六烷基二苯醚二磺酸钠(C16-MADS)的胶束聚集数(Nm),并考察了表面活性剂浓度、无机盐浓度和温度对Nm的影响。结果表明,表面活性剂的浓度必须高于临界胶束浓度(cmc)一个数量级时,才能测定出较为合适的Nm;随着C16-MADS浓度的增大,Nm增大,胶束内核的微极性变小;而温度对Nm的影响极小。与传统的单亲水基阴离子表面活性剂相比,C16-MADS的有序聚集体表现出许多不同之处,如Nm仅为传统的单亲水基阴离子表面活性剂胶束聚集数的一半,无机盐浓度对Nm的影响极小,胶束内核的微极性明显增大。  相似文献

在阳离子表面活性剂CTMAB水溶液中,对二甲氨基苯甲酸(DMABOA)的分子内扭转电荷转移(TICT)荧光带随着胶束的形成而显著蓝移,同时伴随着TICT荧光带与正常荧光带强度比Ia/Ib和总荧光强度If的增大;而阴离子表面活性剂SLS胶束的形成并未引起TICT荧光带位置和Ia/Ib的变化,仅使If提高(较在CTMAB胶束中的小)。实验发现在胶束疏水核中的TICT行为与纯有机溶剂中不同,DMABOA与CTMAB胶束的疏水相互作用和静电作用是协同的。  相似文献

This study is aimed at establishing optimal conditions for the use of 2,2'-[1,3-propanediylbis[(dimethyliminio)-3,1-propanediyl-1(4H)-pyridinyl-4-ylidenemethy-lidyne]]bis[3-methyl]-tetraiodide (BOBO-1) as a fluorescent probe in the characterization of lipid/DNA complexes (lipoplexes). The fluorescence spectra, anisotropy, fluorescence lifetimes and fluorescence quantum yields of this dimeric cyanine dye in plasmid DNA (2694 base pairs) with and without cationic liposomes (1,2-dioleoyl-3-trimethylammonium-propane [DOTAP]), are reported. The photophysical behavior of the dye in the absence of lipid was studied for several dye/DNA ratios using both supercoiled and relaxed plasmid. At dye/DNA ratios (d/b) below 0.01 the fluorescence intensity increases linearly, whereas lifetime and anisotropy values of the dye are constant (tau approximately 2.5 ns and = 0.20). By agarose gel electrophoresis it was verified that up to d/b = 0.01 DNA conformation is not considerably modified, whereas for d/b = 0.05-0.06 a single heavy band appears on the gel. For these and higher dye/DNA ratios the fluorescence intensity, anisotropy and average lifetime values decrease with an increase in BOBO-1 concentration. When cationic liposomes are added to the BOBO-1/DNA complex, an additional effect is noticed: The difference in the environment probed by BOBO-1 bound to DNA leads to a decrease in quantum yield and average lifetime values, and a redshift is apparent in the emission spectrum. For fluorescence measurements including energy transfer (FRET), a d/b ratio of 0.01 seems to be adequate because no considerable change on DNA conformation is detected, a considerable fluorescent signal is still measured after lipoplex formation, and energy migration is not efficient.  相似文献

以4-溴-1,8-萘酐为原料,合成脂溶性4-[2-(二甲氨基)乙氧基]-N-十八烷基-1,8-萘酰亚胺。对其进行了1H NMR和IR表征。考察了荧光染料在DMF、乙腈、丙酮、乙酸乙酯和乙醚5种溶剂中的荧光光谱和吸收光谱,发现由于溶剂效应,随溶剂极性由小到大,荧光光谱和吸收光谱的最大峰值波长逐渐红移。考察了不同金属离子和pH对荧光染料荧光光谱的影响,结果表明荧光强度随Fe3+、Zn2+、Co2+浓度增大而逐渐增强,Fe3+的影响最为显著;当pH<7时,荧光强度随着pH的降低逐渐增强;进一步考察了Fe3+、Zn2+、Co2+对吸收光谱的影响,结果发现吸收光谱均蓝移。分析认为荧光染料的光致电子转移被阻碍,实验结果表明,合成的新型荧光染料可用于溶液中金属离子和pH的检测。  相似文献

Based on a change in structure between spirocyclic (non-fluorescent) and ring-open (fluorescent) forms of rhodamine-based dyes, a new fluorescent and colorimetric Cu(2+) probe was designed and synthesized. Upon treatment with Cu(2+), the weakly fluorescent probe exhibited a strong fluorescence response with high selectivity. The interaction of a new derivative of thiazole orange (TO-3) with calf thymus DNA (ctDNA) has been investigated by fluorescence and absorption spectroscopy. When TO-3 binds to ctDNA, absorption bands exhibit significant hypochromicity at low base pair/dye ratio (BP/D ratio), and high BP/D show hyperchromicity with red shift. The spectral changes are attributed to the different species formed between TO-3 and ctDNA in the titration course of the dye molecule with DNA. Multivariate curve resolutions–alternating least squares (MCR–ALS) is applied to the absorption measurements recorded to recover the concentration profiles and the pure spectra of the DNA/TO-3 complexes involved in the process. The binding constant and size of the binding site have been determined spectrophotometrically using the McGhee von Hippel equation. MCR–ALS has been used to reveal the precise concentration profiles of all detectable species formed between ctDNA and TO-3 and their pure spectral profiles.  相似文献

Two 10-mer DNA probes, or one 20-mer DNA probe, respectively, hybridize with a 21-mer target DNA to form a vacancy or bulge opposite the target nucleotide. The former double-DNA-probe method and the latter bulge form method are applicable to the detection of single-nucleotide polymorphisms (SNPs). A small fluorescent dye enters into the vacancy or bulge and binds with a target nucleotide via a hydrogen bonding interaction, which causes fluorescence quenching. The interaction between fluorescent dye and the target nucleotide is confirmed by measuring the melting temperature and fluorescence spectra. The fluorescent dye, ADMND (2-amino-5,7-dimethyl-1,8-naphthyridine), is found to selectively bind with C over A or G. The methods proposed here are economic, convenient, and effective for the fluorescence detection of SNPs. Finally, the double-DNA-probe method and bulge form method are successfully applied to the detection of C/G and C/A mutations in the estrogen receptor 2 gene and progesterone receptor gene using ADMND.  相似文献

With the development of single-molecule detection and super-resolution fluorescence imaging, rhodamine dyes gain new life. Through the modification of the N-substituents and the replacement of the oxygen atom in xanthene, the wavelength and brightness can be effectively changed. However, the spectra of rhodamine, especially due to the balance between ring-closed non-fluorescent lactone and ring-opened fluorescent zwitterion/cation, are sensitive to interference from various environmental factors. In this way, the spectral data of various rhodamines reported by different research groups under different test conditions lacked comparability, sometimes even lacked accuracy. In order to meet the requirements for the accuracy and uniformity of spectral data in the research of single molecule imaging and dye structure-fluorescence relationship study, we have tested the spectra of fifteen rhodamine dyes that cover the visible and near-infrared regions under exactly the same conditions. By studying the dependence of the spectra on dye concentrations, it was confirmed that 1 μmol/L was ideal for detection less from the interference of dye molecule aggregation. We provide comprehensive and reliable spectral data of these fifteen dyes, which are expected to be used as references for future research. And the direct comparison of different rhodamine spectra would help to understand the structure-fluorescence relationship of rhodamines.  相似文献

本文合成了3个新型中位分别为N,N-二甲基苯胺、对-甲氧基苯基或苯基取代的含稠合外环的硼-二吡咯亚甲基染料。研究了它们的吸收光谱、稳态荧光光谱和电化学性质;采用荧光光谱滴定方法研究了它们在强极性溶剂中对氢离子的响应;将氢离子滴加到N,N-二甲基苯胺取代的硼-二吡咯亚甲基染料(1)的CH3CN-H2O(1∶1,V/V)溶液中,其溶液的荧光显著增强;染料1在可见光激发下,可以作为酸性pH范围内检测氢离子的荧光探针。  相似文献
