We describe the development of an electrophoresis microchip fabricated by a direct-printing process, based on lamination of printed polyester films with end-channel amperometric detection. The channel structures are defined by polyester (base and cover) and by a toner layer (walls). The polyester-toner devices presented an electroosmotic flow (EOF) magnitude of approximately 10(-5) cm2 V(-1) s(-1), which is generated by a polymeric mixture of the toner and polyester composition. The microelectrodes used for detection were produced combining this laser-printer technology to compact discs. The performance of this device was evaluated by amperometric detection of iodide and ascorbate. The detection limits found were 500 nmol.L(-1) (135 amol) and 1.8 micromol.L(-1) (486 amol) for iodide and ascorbate, respectively.  相似文献

This paper reports for the first time the use of colored toner to produce polyester toner (PT) ME devices. Colored PT devices were designed in drawing software and printed on a polyester film using a color laser printer with 3600 dpi resolution. The colored toner is composed of a copolymer mixture (styrene and acrylate), wax, silicon dioxide, and pigments. The presence of silica in the toner composition has enhanced the EOF magnitude and improved the analytical performance. For a pH range between 2 and 12, the EOF measured on a magenta PT chip, for example, ranged from 3.8 to 5.8 (× 10?4 cm2 V?1 s?1). Typical separations of inorganic cations (K+, Na+, and Li+) were used as model system to investigate the analytical feasibility of the proposed devices. The repeatability for the migration times of all analytes exhibited RSD values lower than 1% (n = 10). The separation efficiencies found on colored PT devices ranged from 10 000 to 49 000 plates/m, which means between 7 and 23% of the maximum theoretical efficiency on this microfluidic platform (1.85 × 105 plates/m). The improvements achieved on the proposed devices are associated with the small additional amount of silica on the toner composition as well as the printing of channels with smoother surfaces and better uniformity when compared to the conventional PT chips printed with monochromatic laser printers.  相似文献

A microfluidic device with integrated waveguides and a long path length detection cell for UV/Vis absorbance detection is presented. The 750 microm U-cell detection geometry was evaluated in terms of its optical performance as well as its influence on efficiency for electrophoretic separations in the microdevice. Stray light was found to have a strong effect on both, the sensitivity of the detection and the available linear range. The long path length U-cell showed a 9 times higher sensitivity when compared to a conventional capillary electrophoresis (CE) system with a 75 microm inner diameter (ID) capillary, and a 22 times higher sensitivity than with a 50 microm ID capillary. The linear range was comparable to that achieved in a 75 microm ID capillary and more than twice as large as in a 50 microm ID capillary. The use of the 750 microm U-cell did not contribute significantly to band broadening; however, a clear quantification was made difficult by the convolution of several other band broadening sources.  相似文献

A new process for the manufacture of microfluidic devices based on deposition of laser-printing toner on glass substrates is described. It is an alternative method to the toner on polyester film (toner-polyester) one, previously introduced. Commercial laser printers cannot print directly on glass, thus the toner must first be printed on a special paper and then transferred by heating under pressure to the glass surface. Although this procedure is more complex than the toner-polyester one, it can be repeated several times, yielding multiple toner layers. Even without special alignment equipment, up to four layers could be satisfactorily piled up. Characterization tests revealed that the toner-glass devices have similar behavior as toner-polyester ones regarding the toner layer porosity. The main advantages of the toner-glass technology are improved mechanical stability, possibility of multiple toner layers, augmented electroosmotic flow (EOF), and improved heat transfer. On the other hand, toner adhesion to glass is weaker than to polyester, which limits the device lifetime and usable liquid media. The measured EOF mobility (3.5 x 10(-4) cm2.V(-1).s(-1) for pH 7) suggests that it is mainly determined by the glass surface, being little influenced by the toner walls. Microchip electrophoresis with contactless conductivity detection and photometric detection were implemented using toner-glass devices.  相似文献

A second generation of a battery-powered portable electrophoresis instrument for the use of ME with electrochemical detection was developed. As the first-generation, the main unit of the instrument (150 mm × 165 mm × 95 mm) consists of four-outputs high-voltage power supply (HVPS) with maximum voltage of 3 KV and acquisition system (bipotentiostat) containing 2-channels for dual electrochemical detection. A new reusable microfluidic platform was designed in order to incorporate the microchips with the portable instrument. In this case, the platform is integrated to the main unit of the instrument so that it is not necessary to have any external cable for the interconnection of both parts, making the use of the complete system easier. The new platform contains all the electrical connections for the HVPS and bipotentiostat, as well as fluidic ports for driving the solutions. The microfluidic electrophoresis instrument is controlled by means of a user-friendly interface from a computer. The possibility of wireless connection (Bluetooth?) allows the use of the instrument without any external cable improving the portability. Therefore, the second generation brings a more compact and integrated electrophoresis instrument for "in situ" applications using microfluidic chips in an easy way. The performance of the electrophoresis system was initially evaluated using single- and dual-channel SU-8/Pyrex microchips with different models of integrated electrodes including microelectrodes and interdigitated arrays. The method was tested in different analytical applications such as separation of neurotransmitters, chlorophenols, purine derivatives, vitamins, polyphenolic acids, and flavones.  相似文献

A simple method to fabricate cylindrical carbon electrodes for use in capillary electrophoresis (CE) microchips is described. The electrodes were fabricated using a metallic wire coated with carbon ink. Several experimental variables were studied in order to establish the best conditions to fabricate the electrode. Finally, the electrodes were integrated in a poly(dimethylsiloxane) microchip and used for the analysis of phenolic compounds. Using the optimum conditions, the analysis of a mixture of dopamine, epinephrine, catechol, and 4-aminophenol was achieved in less than 240 s, showing good linear responses (R2 = 0.999) in the 0.1-190 μM range, and limits of detection (without the use of stacking or a decoupler) of 140 and 105 nM for dopamine and epinephrine, respectively.  相似文献

Miniaturized analytical systems, especially microchip CE (MCE), are becoming a promising tool for analytical purposes including DNA analysis. These microdevices require a sensitive and miniaturizable detection system such as electrochemical detection (ED). Several electroactive DNA intercalators, including the organic dye methylene blue (MB), anthraquinone derivatives, and the metal complexes Fe(phen)3 2+ and Ru(phen)3 2+, have been tested for using in combination with thermoplastic olefin polymer of amorphous structure (Topas) CE-microchips and ED. Two end-channel approaches for integration of gold wire electrodes in CE-ED microchip were used. A 250 microm diameter gold wire was manually aligned at the outlet of the separation channel. A new approach based on a guide channel for integration of 100 and 50 microm diameter gold wire has been also developed in order to reduce the background current and the baseline noise level. Modification of gold wire electrodes has been also tested to improve the detector performance. Application of MCE-ED for ssDNA detection has been studied and demonstrated for the first time using the electroactive dye MB. Electrostatic interaction between cationic MB and anionic ssDNA was used for monitoring the DNA on microchips. Thus, reproducible calibration curves for ssDNA were obtained. This study advances the feasibility of direct DNA analysis using CE-microchip with ED.  相似文献

We present a new approach for contactless conductivity detection for microchip-based capillary electrophoresis (CE). The detector integrates easily with well-known microfabrication techniques for glass-based microfluidic devices. Platinum electrodes are structured in recesses in-plane with the microchannel network after glass etching, which allows precise positioning and batch fabrication of the electrodes. A thin glass wall of 10-15 microm separates the electrodes and the buffer electrolyte in the separation channel to achieve the electrical insulation necessary for contactless operation. The effective separation length is 34 mm, with a channel width of 50 microm and depth of 12 microm. Microchip CE devices with conductivity detection were characterized in terms of sensitivity and linearity of response, and were tested using samples containing up to three small cations. The limit of detection for K+ (18 microM) is good, though an order of magnitude higher than for comparable capillary-based systems and one recently reported example of contactless conductivity on chip. However, an integrated field-amplified stacking step could be employed prior to CE to preconcentrate the sample ions by a factor of four.  相似文献

The application of plastified laser-printed poly(ethylene terephthalate)(PET)-toner microchips to capillary electrophoresis was investigated. Electroosmotic flow was observed in the direction of the cathode for the buffer system studied (phosphate, pH 3–10). Average electroosmotic mobilities of 1.71×10–4 to 4.35×10–4 cm2 V–1 s–1 were observed from pH 3 to 10. This variation suggests that silica fillers in the toner and on the surface of the polymer dominate the zeta potential of the material, which is also confirmed by XPS measurements. Dopamine and catechol were used as model analytes for microchip electrophoresis in combination with electrochemical detection. Results show that these two analytes can be efficiently separated and detected electrochemically with the plastified laser-printed PET-toner microchips.  相似文献

We describe a simple and easy way to construct gold microelectrodes for amperometric detection in capillary electrophoresis (CE). The gold microelectrodes, in single or twin sets, were obtained from recordable compact discs (gold-sputtered type), which present highly reproducible surface characteristics. The performance of these electrodes was evaluated by using a home-made CE equipment. The basic steps for the electrode construction are: drawing on a microcomputer; laser printing of the design on wax paper; heat-transfer of the toner onto the gold surface of a peeled recordable compact disc (CD-R); etching of the gold layer from unprinted regions; removal of the toner with a solvent; sealing of unused electrode areas with varnish. One electrode at a time was connected to a potentiostat (or two, to a bipotentiostat) and operated in a wall jet configuration relative to the CE capillary outlet. The amperometric signals were integrated for quantification purposes. Repetitive injections (n = 10) of a mixture containing iodide, ascorbic acid, dipyrone, and acetaminophen (20, 200, 500, and 100 microM), presented relative standard deviations of 2.9, 4.5, 6.1, and 4.0%, respectively. For these analytes, the detection limits (S/N = 3, 30 s of 100 mm hydrodynamic injection) were 0.1, 0.5, 3.1, and 1.1 microM, respectively.  相似文献

Separation and determination of dopamine and epinephrine with end-channel electrochemical (EC) detection integrated on a native printed microchip capillary electrophoresis (CE) system was investigated. Factors influencing the separation and detection were investigated and optimized. Results show manipulating EOF, which can be easily achieved by adjusting buffer pH, is a simple and effective way to achieve the baseline separation of dopamine and epinephrine in native polymeric microchips. Without surface modification of microchannel, printed microchips with advantages of low cost and easy preparation can achieve high performance like other microfluidic devices.  相似文献

Inexpensive and disposable polyester microchips were fabricated through photolithographic and wet-chemical etching procedure, followed by replication using an imprinting method at room temperature. Laboratory-scale laser-induced fluorescence equipment was employed as a detection system. The generation of electroosmotic flow (EOF) on the polyester channels was discussed in this paper. Surfactants in the running buffer had a significant effect on the EOF depending on their types. The epsilon potential of the electric double layer formed by adsorbing sodium lauryl sulfate molecules on the wall of polyester channels seemed to be constant within the buffer pH investigated. EOF could also be suppressed to zero by adding polyoxyethylene 23 lauryl ether into the running buffer. The separation of two laser dyes was obtained using polyester chips through both micellar electrokinetic chromatography and capillary zone electrophoresis. The polyester channels modified with 10-undecen-1-ol exhibited a dramatically high-separation efficiency compared with the conventional fused-silica capillary tubes.  相似文献

A contactless conductivity detector integrated into a poly(dimethylsiloxane) microchip for electrophoresis is presented. It adopted the simplest configuration of electrodes commonly used in this detection mode for capillary electrophoresis microchips. Although the chip is based on a simple and effective design, it is able to obtain low detection levels due to the low noise of the detection circuit. A circuit based on a lock-in amplifier was designed on printed circuit boards to read out the signal. The property of the detection cell was studied by applying excitation signals of different frequencies and different amplitudes. It was found that the best detection limit could be achieved with a frequency of 50?kHz and amplitude of 20?V. The performance of the detector was demonstrated by successfully separating and detecting several inorganic ions and also a mixture of heavy metal ions. An average detection limit of 0.4?μM was obtained for inorganic cations. This value is significantly improved compared to similar microchip-based detectors. The presented detector could be promising for mass production due to its properties, such as simple construction, high degree of integration, high performance and low cost.  相似文献

Microchip capillary electrophoresis (CE) with integrated four-electrode capacitively coupled conductivity detection is presented. Conductivity detection is a universal detection technique that is relatively independent on the detection pathlength and, especially important for chip-based analysis, is compatible with miniaturization and on-chip integration. The glass microchip structure consists of a 6 cm etched channel (20 microm x 70 microm cross section) with silicon nitride covered walls. In the channel, a 30 nm thick silicon carbide layer covers the electrodes to enable capacitive coupling with the liquid inside the channel as well as to prevent interference of the applied separation field. The detector response was found to be linear over the concentration range from 20 microM up to 2 mM. Detection limits were at the low microM level. Separation of two short peptides with a pI of respectively 5.38 and 4.87 at the 1 mM level demonstrates the applicability for biochemical analysis. At a relatively low separation field strength (50 V/cm) plate numbers in the order of 3500 were achieved. Results obtained with the microdevice compared well with those obtained in a bench scale CE instrument using UV detection under similar conditions.  相似文献

A precolumn reaction chamber was integrated into a polyester microfluidic device with a miniaturized detection system. The reaction chamber was designed to be a zigzag channel, 70 microm in width, 8 mm in length, followed by a wider straight channel, 150 microm in width, 2 mm in length. The detection system is composed of an embedded light-emitting diode (LED), an integrated optical fiber, and a photomultiplier tube (PMT). A success in amino acid analysis using the integrated microchemical analysis device proved that the precolumn reaction chamber was compatible with the integrated detection system. Three kinds of amino acids, arginine, glycine, and phenylalanine, mixed and reacted with 7-fluoro-4-nitrobenzo-2-oxa-1,3-diazole (NBD-F) in the precolumn reaction chamber to produce fluorescent products, were separated by micellar eletrokinetic chromatography (MEKC) and detected by LED-excited fluorescence. The detection limits for arginine, glycine, and phenylalanine were 1, 1, and 0.5 mM, respectively, which can be improved by further optimizations of the reaction system and detection system.  相似文献

A new SU-8 based microchip capillary electrophoresis (MCE) device has been developed for the first time with integrated electrochemical detection. Embedded electrophoretic microchannels have been fabricated with a multilayer technology based on bonding and releasing steps of stacked SU-8 films. This technology has allowed the monolithic integration in the device of the electrochemical detection system based on platinum electrodes. The fabrication of the chips presented in this work is totally compatible with reel-to-reel techniques, which guarantee a low cost and high reliability production. The influence of relevant experimental variables, such as the separation voltage and detection potential, has been studied on the SU-8 microchip with an attractive analytical performance. Thus, the effective electrical isolation of the end-channel amperometric detector has been also demonstrated. The good performance of the SU-8 device has been proven for separation and detection of the neurotransmitters, dopamine (DA) and epinephrine (EP). High efficiency (30,000-80,000 N/m), excellent precision, good detection limit (450 nM) and resolution (0.90-1.30) has been achieved on the SU-8 microchip. These SU-8 devices have shown a better performance than commercial Topas (thermoplastic olefin polymer of amorphous structure) microchips. The low cost and versatile SU-8 microchip with integrated platinum film electrochemical detector holds great promise for high-volume production of disposable microfluidic analytical devices.  相似文献

The fabrication and evaluation of micromolded dual carbon ink electrodes and their integration with a fabricated palladium decoupler for use in microchip electrophoresis is described. As opposed to previous work involving carbon-based dual electrodes with microchip electrophoresis, this approach results in electrodes that are amenable to mass production in a manner where the decoupler/electrode alignment is fixed and reproducible. In this work, electrode sizes and spacings were optimized to result in dual carbon electrodes that are 1 microm in height and separated by 100 microm. Fluorescence microscopy was used to investigate leakage around the electrode/channel interface as well as to investigate what effect the dual electrodes have on band broadening phenomena. The performance of the microelectrodes was demonstrated by the separation and selective dual electrode detection of neurotransmitters in the presence of ascorbic acid. It was also found that addition of SDS to the buffer system improved both the LODs and collection efficiencies. This approach, which is the first involving carbon-based dual electrodes with an on-chip palladium decoupler, will be useful for separating and detecting neurotransmitters that are either collected by in vivo sampling or released from cells on-chip.  相似文献

The detection of underivatized anionic sulfonates, carboxylates, amino acids, sugars, and artificial sweeteners, and of cationic dopamine, ephedrine, and metanephrine in microfabricated electrophoresis devices is demonstrated. This was achieved by high-voltage contactless conductivity measurements with external electrodes. Poly(methyl methacrylate) chips with thin covers to enable sensitive contactless detection were used for most determinations but glass microchips had to be employed for amino acids and sugars. The plastic chips were found not be stable in the alkaline media required to render those two classes of species in the ionic form amenable for separation and detection. The reproducibility of peak area measurements was about 1% or better and the detection limits ranged between 1 and 30 microM for the different compounds examined.  相似文献

The review provides a comprehensive survey of the recent applications of contact and contactless conductivity detection in capillary electrophoretic and chip electrophoretic analyses of a broad scale of compounds, from low-molecular-mass highly mobile small inorganic and organic ions, via medium-molecular-mass peptides and oligo- and polynucleotides up to high-molecular-mass biopolymers, proteins and nucleic acids fragments. The review presents also the recent developments in the construction of different types of conductivity detectors (detectors with galvanic contact of the sensing electrodes with the BGE and sample components, contactless conductivity detectors with capacitively coupled tubular and semitubular electrodes and combined conductivity/optical detectors) applied in the capillary electromigration methods performed in classical fused silica, polytetrafluorethylene, and polyetheretherketone capillaries or on glass and polymethylmethacrylate microchips. In addition, the principle and theoretical bases of conductivity detection in capillary electromigration techniques, zone electrophoresis, ITP, micellar EKC, and electrochromatography are briefly described.  相似文献

A simple, fast, and inexpensive masking technology without any photolithographic step to produce glass microchannels is proposed in this work. This innovative process is based on the use of toner layers as mask for wet chemical etching. The layouts were projected in graphic software and printed on wax paper using a laser printer. The toner layer was thermally transferred from the paper to cleaned glass surfaces (microscope slides) at 130 degrees C for 2 min. After thermal transference, the glass channel was etched using 25% (v/v) hydrofluoric acid (HF) solution. The toner mask was then removed by cotton soaked in acetonitrile. The etching rate was approximately 7.1 +/- 0.6 microm min(-1). This process is economically more attractive than conventional methods because it does not require any sophisticated instrumentation and it can be implemented in any chemical/biochemical laboratory. The glass channel was thermally bonded against a flat glass cover and its analytical feasibility was investigated using capacitively coupled contactless conductivity detection (C(4)D) and laser-induced fluorescence (LIF) detection.  相似文献
